Detection of Nitrosomonas spp. by polymerase chain reaction 总被引:2,自引:0,他引:2
Abstract A unique genomic DNA fragment was isolated from Nitrosomonas europaea ATCC 19718. Based on the sequence of this fragment, oligonucleotide primers for polymerase chain reaction amplification were prepared which amplify sequences of 775 and 658 bp. The predicted DNA fragments were both amplified from the genome of N. europaea and a Nitrosomonas spp. isolated from a local oxidation pond. The primers failed to amplify DNA from the genomes of the ammonia oxidiser Nitrosolobous multiformis , the nitrite oxidiser Nitrococcus mobilis as well as from the genomes of other unrelated heterotrophic bacteria. These DNA sequences could be amplified from 0.01 ng of N. europaea genomic DNA or from 100 intact cells, and it was possible to detect Nitrosomonas DNA in a DNA mixture extracted from water samples drawn from a local oxidation pond. 相似文献

Summary Bacteria utilizing high concentrations of acetonitrile as the sole carbon source were isolated and identified asChromobacterium sp. andPseudomonas aeruginosa. Maximum growth was attained after 96 h of incubation andP. aeruginosa grew slightly faster thanChromobacterium sp. The strains were able to grow and oxidize acetonitrile at concentrations as high as 600 mM. However, higher concentrations inhibited growth and oxygen uptake. Degradation studies with (14C)acetonitrile indicated 57% of acetonitrile was degraded byPseudomonas aeruginosa as compared to 43% byChromobacterium. In situ gel-forming systems have drawn much attention of current researchers to overcome the poor bioavailability from the conventional
eye drops. The present work described formulation and pharmacoscintigraphic evaluation of timolol-maleate-loaded chitosan/hydroxy
propyl methyl cellulose (HPMC)-based polymer matrix for enhanced ocular retention. Chitosan and HPMC ratio was optimized and
formulation was characterized for various in vitro parameters. The ocular retention was studied on New Zealand rabbits by gamma scintigraphy, which is a very simple and noninvasive
technique. For scintigraphy study, the drug timolol maleate was radiolabeled 99mTc by direct labeling method using SnCl2·2H2O as reducing agent. The labeling procedure was optimized to get maximum labeling efficiency (>98%). In vitro stability of the radiolabeled drug (99mTc-timolol maleate complex) was checked and it was found to be stable for up to 24 h. Plain drug eliminates rapidly as significant
activity was recorded in kidney and bladder after 2 h of ocular administration. It was evident from the scintigraphic images
and the time–activity curve plotted from the data that the plain drug solution cleared very rapidly from the corneal region
and reached into systemic circulation via nasolachrymal drainage system, as significant activity was recorded in kidney and
bladder after 2 h of ocular administration. Developed formulation cleared at a slow rate and remained at corneal surface for
longer time duration. No radioactivity was observed in systemic circulation after 2 h. Ocular irritation of the developed
formulation was also checked by hen’s egg chorioallantoic membrane test and formulation was found to be practically nonirritant.
The study signified the potential of gamma scintigraphy in evaluation of novel drug delivery systems in a noninvasive manner. 相似文献

An approach of combining flow cytometry (FCM) analysis with morphological and chemical profiling was used to assess the genetic
stability and bioactive compound diversity in a Scutellaria baicalensis Georgi (Huang-qin) germplasm collection that was clonally maintained in in vitro for a period of over 6 years. Based on the
FCM analysis of nuclei samples from young shoots, the nuclear DNA content of S. baicalensis was calculated as 0.84 pg/2C. FCM analysis showed no significant variation in the nuclear DNA contents and ploidy levels
in the long-term in vitro maintained germplasm lines. Germplasm lines, acclimatized to ex vitro conditions, exhibited distinctive
plant growth and bioactive compound production capacities. The high level of genetic stability observed in in vitro maintained
S. baicalensis lines opens up a variety of opportunities such as allowing long-term aseptic preservation and easy distribution of well-characterized
germplasm lines of this medicinal plant species. This study represents a novel approach for continuous maintenance, monitoring,

Background
Accurate, inexpensive point-of-care CD4+ T cell testing technologies are needed that can deliver CD4+ T cell results at lower level health centers or community outreach voluntary counseling and testing. We sought to evaluate a point-of-care CD4+ T cell counter, the Pima CD4 Test System, a portable, battery-operated bench-top instrument that is designed to use finger stick blood samples suitable for field use in conjunction with rapid HIV testing.Methods
Duplicate measurements were performed on both capillary and venous samples using Pima CD4 analyzers, compared to the BD FACSCalibur (reference method). The mean bias was estimated by paired Student''s t-test. Bland Altman plots were used to assess agreement.Results
206 participants were enrolled with a median CD4 count of 396 (range; 18–1500). The finger stick PIMA had a mean bias of −66.3 cells/µL (95%CI −83.4−49.2, P<0.001) compared to the FACSCalibur; the bias was smaller at lower CD4 counts (0–250 cells/µL) with a mean bias of −10.8 (95%CI −27.3−+5.6, P = 0.198), and much greater at higher CD4 cell counts (>500 cells/µL) with a mean bias of −120.6 (95%CI −162.8, −78.4, P<0.001). The sensitivity (95%CI) of the Pima CD4 analyzer was 96.3% (79.1–99.8%) for a <250 cells/ul cut-off with a negative predictive value of 99.2% (95.1–99.9%).Conclusions
The Pima CD4 finger stick test is an easy-to-use, portable, relatively fast device to test CD4+ T cell counts in the field. Issues of negatively-biased CD4 cell counts especially at higher absolute numbers will limit its utility for longitudinal immunologic response to ART. The high sensitivity and negative predictive value of the test makes it an attractive option for field use to identify patients eligible for ART, thus potentially reducing delays in linkage to care and ART initiation. 相似文献6.

Biosurfactant production by Pseudomonas aeruginosa EBN-8 mutant was studied in shake flasks on separate wastes from canola, soybean and corn oil refineries. Of the substrates
tested, canola oil refinery waste (COD=20 g l−1) supplemented with sodium nitrate (at COD/N=20) showed the best microbial growth (4.50 g l−1) and rhamnolipid production (8.50 g l−1), at 10 d of incubation with the specific growth rate of 0.316 h−1 and specific product yield of 0.597 g g−1 h. The interconnection network is one of the key architectural components in any parallel computer. The distribution of the traffic
injected into the network is among the factors that greatly influences network performance. The uniform traffic pattern has
been adopted in many existing network performance evaluation studies due to the tractability of the resulting analytical modelling
approach. However, many real applications exhibit non-uniform traffic patterns such as hot-spot traffic. K-ary n-cubes have been the mostly widely used in the implementation of practical parallel systems. Extensive research studies have
been conducted on the performance modelling and evaluation of these networks. Nonetheless, most of these studies have been
confined to uniform traffic distributions and have been based on software simulation. The present paper proposes a new stochastic
model to predict message latency in k-ary
n-cubes with deterministic routing in the presence of hot-spot traffic. The model has been validated through simulation experiments
and has shown a close agreement with simulation results.

Cadmium (Cd) accumulation in edible crops is undesirable due to its hazardous influences on human health. The objectives of this study were: i) to evaluate the spatial variability of grain Cd and its relationships with soil properties in 4000 km2 wheat farms; ii) to evaluate the effect of wheat cultivar on the soil properties vs. grain Cd relationships. A number of 255 soil (0–20 cm) and grain samples were taken and Cd concentrations in grain samples and some soil properties were measured. Grain Cd concentrations in 95 percent of the samples exceeded the threshold of 0.2 mg kg?1. Durum wheat had more potential to accumulate Cd in grain (0.76 mg kg?1) than bread (0.69 mg kg?1). There was significant (p < 0.01) correlation between grain Cd and organic carbon (r = 0.66), CEC (r = 0.77) and DTPA-extractable Cd (p < 0.05) (r = 0.57) of the soils. Greater Pearson coefficient values for durum wheat showed that, in the studied calcareous soils, organic carbon, CEC, Cd-DTPA had more effects on durum wheat than bread wheat cultivar. The obtained Kriging map of grain Cd identified three hotspots at the east (durum wheat cultivation), the west (intensive irrigated wheat farms), and south (wheat farms around petrochemical industries) of the region. Agricultural mismanagement due to overusing P-fertilizers increased Cd concentration in the topsoils and grains of wheat farms in the study area. 相似文献
