In this paper studies on the role of flavonoids in pea root nodule development are reported. Flavonoid synthesis was followed by localizing chalcone synthase (CHS) mRNA in infected pea roots and in root nodules. In a nodule primordium, CHS mRNA is present in all cells of the primordium. Therefore it is hypothesized that the Rhizobium Nod factor induces cell division in the root cortex by stimulating the production of flavonoids that function as auxin transport inhibitors. In nodules CHS mRNA is predominantly present in a region at the apex of the nodule consisting of meristematic and cortical cells. These cells are not infected by Rhizobium. Therefore it is postulated that CHS plays a role in nodule development rather than in a defence response. In roots CHS mRNA is located at a similar position as in nodules, suggesting that CHS has the same function in both root and nodule development. When nodules are formed by mutants of Rhizobium leguminosarum bv. viciae that are unable to secrete β(1-2) glucan and to synthesize the O-antigen containing LPS I, CHS genes are also expressed in regions of the nodule that are infected by Rhizobium. It is postulated that the impaired development of nodules formed by these mutants is due to an induction of a plant defence response.  相似文献

The development of about 20 relatively small nests of Xylocopa pubescens was studied. After the first offspring had become adult, these nests reached a social stage in which there was only one egg-layer per nest. Freshly emerged (teneral) adults eat a lot of food, collected by a forager, before they fly out of the nest. This food appears to be of major importance to these bees in that it makes them fully agile. It seems therefore, that part of the food needed for the development of a young bee is not given at the larval stage, via the beebread, but at the teneral adult stage. As a result of this, a necessary overlap of generations is accomplished. Besides, less pollen has to be collected for the provision of a brood cell, so more cells can be constructed within a short period of time. There is a high degree of nest competition. Many nests were taken over by conspecific individuals (or by females of X. sulcatipes) that were searching for nesting sites within the study area. However, although more than 50% of the solitary nests were taken over sooner or later, strangers hardly ever intruded into social nests, with more than one adult. This illustrates how important it is for a reproducing female to tolerate the presence of nestmates which guard the nest in her absence. Although in most cases the foundress of a nest proceeded to produce new brood in the presence of her offspring, it happened that a daughter took over reproduction from her mother. In two of these cases, it could be observed that a mother started a new nest elsewhere after having been thrown out by her own daughter. At least in these cases, nest competition between mother and daughter started long before the mother had reached the end of her reproductive capacities. Since nesting possibilities are scarce, it seems a logical strategy to stay in the maternal nest and wait for a chance to become the egg-layer if the mother dies or if she loses her dominance. The occurrence of several social interactions among nestmates is discussed: — trophallaxis was observed often, not only under ‘forced’ conditions, but also as a form of ‘voluntary’ feeding; — nestmates were observed to groom each other.  相似文献

Honeybee queen attendants disperse queen pheromones to supplement pheromone dispersal by direct queen-worker contacts. With time they lose their dispersal function exponentially due mainly to volatilization of queen pheromones carried on their bodies. The elimination of those airborne pheromones together with the air while ventilating the hive is balanced by pheromone release by the queen. This equilibrium results in a certain level of queen pheromones in the broodnest. The change of the pheromone level (for example, due to loss of the colony of its queen) can serve as a signal to alter the behaviour of the workers and the state of the colony.  相似文献

Purpose

This study aimed to investigate the environmental consequences (on climate change and land use) of an increase in preference for grass-based milk in France using a consequential life cycle assessment (CLCA) approach. This increase in preference was assumed to be satisfied domestically, by converting maize silage-based dairy farms (MS farm) to grass-based dairy farms (G farm) while keeping on-farm usable agricultural area and total milk production of farm constant.

Methods

The possible consequences of an increase in preference for grass-based milk were identified based on cause and effect relationships. The conversion from MS to G farm reduced the use of soybean meal, changed the on-farm cropping pattern and produced more animals but less wheat and no rapeseed. Effects on on-farm soil C were predicted with the RothC model and on global land use change (LUC) with models of global agricultural markets (Global Trade Analysis Project (GTAP) and Landbouw Economisch Instituut Trade Analysis Project (LEITAP)). System expansion using animals from a suckler beef production system was applied to estimate the impacts of milk and animal co-products from the dairy system. Land occupation and climate change impacts were estimated. The consequences of farm conversion were attributed only to the milk, as preference for grass-based milk drove the conversion process.

Results and discussion

The conversion from the MS to G farm increases land occupation and climate change impacts for the G farm, respectively, by 9 and 7 % according to GTAP and 14 and 51 % according to LEITAP. Land occupation and climate change impacts of milk produced by the G farm after conversion increased, respectively, by 82 and 13 % with GTAP and 123 and 97 % with LEITAP relative to those for the MS farm (before conversion). The production of additional wheat and rapeseed outside the G farm increased impacts of the G farm (by 29–69 % depending on impacts and model used). Results indicate that the farm conversion would probably have consequences on global LUC and that it is important to account for this in a LCA approach.

Conclusions

Land use and land use change (LULUC) contributed to the impacts of grass-based milk, and results were highly sensitive to the LULUC model used. The many possible chain-of-event pathways that follow a change in preference for a given product yield high uncertainty in CLCA results. This study only assessed one possible way to meet the increase in preference for grass-based milk; it is necessary to perform a sensitivity analysis to investigate other possible scenarios resulting from this increase in preference.  相似文献

The aim of this article is to develop a methodological approach allowing to assess the influence of parameters of one or more elementary processes in the foreground system, on the outcomes of a life cycle assessment (LCA) study. From this perspective, the method must be able to: (1) include foreground process modeling in order to avoid the assumption of proportionality between inventory data and reference flows; (2) quantify influences of foreground processes’ parameters (and, possibly, interactions between parameters); and (3) identify trends (either increasing or decreasing) for each parameter on each indicator in order to determine the most favorable direction for parametric variation. These objectives can be reached by combining foreground system modeling, a set of two different sensitivity analysis methods (each one providing different and complementary information), and LCA. The proposed method is applied to a case study of hemp‐based insulation materials for buildings. The present study will focus on the agricultural stage as a foreground system and as a first step encompassing the entire life cycle. A set of technological recommendations were identified for hemp farmers in order to reduce the crop's environmental impacts (from –11% to –89% according to the considered impact category). One of the main limitations of the approach is the need for a detailed model of the foreground process. Further, the method is, at present, rather time‐consuming. However, it offers long‐term advantages given that the higher level of model detail adds robustness to the LCA results.  相似文献

Arap1 is a protein that interacts with angiotensin II type 1 (AT(1)) receptors and facilitates increased AT(1) receptor surface expression in vitro. In the present study, we assessed the tissue localization and regulation of Arap1 in vivo. Arap1 was found in various mouse organs, with the highest expression in the heart, kidney, aorta, and adrenal gland. Renal Arap1 protein was restricted to the vasculature and to glomerular mesangial cells and was absent from tubular epithelia. A similar localization was found in human kidneys. To test the hypothesis that angiotensin II may control renal Arap1 expression, mice were subjected to various conditions to alter the activity of the renin-angiotensin system. A high-salt diet (4% NaCl, 7 days) upregulated Arap1 expression in mice by 47% compared with controls (0.6% NaCl, P = 0.03). Renal artery stenosis (7 days) or water restriction (48 h) suppressed Arap1 levels compared with controls (-64 and -62% in the clipped and contralateral kidney, respectively; and -50% after water restriction, P < 0.01). Angiotensin II infusion (2 μg·kg(-1)·min(-1), 7 days) reduced Arap1 mRNA levels compared with vehicle by 29% (P < 0.01), whereas AT(1) antagonism (losartan, 30 mg·kg(-1)·day(-1), 7 days) enhanced Arap1 mRNA expression by 52% (P < 0.01); changes in mRNA were paralleled by Arap1 protein abundance. Experiments with hydralazine and epithelial nitric oxide synthase-/- mice further suggested that Arap1 expression changed in parallel with angiotensin II, rather than with blood pressure per se. Similar to in vivo, Arap1 mRNA and protein were suppressed by angiotensin II in a time- and dose-dependent manner in cultured mesangial cells. In summary, Arap1 is highly expressed in the renal vasculature, and its expression is suppressed by angiotensin II. Thus Arap1 may serve as a local modulator of vascular AT(1) receptor function in vivo.  相似文献

Purpose

Despite the farm being considered by many as the most suitable level of decision-making and strategic management in agriculture, there is an increasing interest in evaluating agricultural management strategies at the regional level. Recent initiatives attempted to aggregate and generalise farm-level lifecycle inventory (LCI) data and lifecycle impact assessment (LCIA) results to describe the environmental performance of agricultural regions. This article describes our development and application of a regional statistics-based approach for constructing virtual representative farms (VRFs), representing dominant farm types for a given region, as a tool for comparing alternative regional agricultural strategies in contexts of insufficient farm (e.g. LCI) data.

Methods

Based on statistical sources, we constructed VRFs of the dominant farm types in the largely agricultural region of Brittany, France. Environmental impacts of different agricultural management strategies were estimated at the regional level by modelling the strategies as changes in VRF-based LCIs, calculating LCIAs and extrapolating their mean per-ha impacts to the total land use in the region. Based on this assessment, performed using a regional lifecycle assessment framework, we analysed relative environmental impacts of each management strategy on the region. A strategy-comparison table was built to allow decision makers to understand the potential regional environmental consequences of implementing each strategy.

Results and discussion

Once VRFs impact assessment results were extrapolated to the regional level, all strategies show environmental impacts per ha similar to those of the baseline, with differences ranging from ?15 to +6%. The scenario featuring centralised fodder drying by 50% of cattle farms (50FOD) is the only one featuring surpluses for all products, due to associated cattle diet adjustments including reduced maize silage intake and partial substitution of concentrate feeds. The scenario featuring grass specialisation by all cattle farms (100GRA) shows a large deficit of grassland products, suggesting that a region-wide extensification strategy would not be self-sufficient.

Conclusions

The method developed enables comparing environmental consequences of region-wide implementation of agricultural strategies, yet, for our case study, it is particularly difficult to identify a “best” one. Nonetheless, the method serves as an initial step for preselecting strategies to investigate at a more detailed level. Prioritisation of a given strategy would likely be based on the environmental pressures considered most pressing by regional decision makers.

Little is known about the extra- and intracellular stimuli inducing renal stem/progenitor cells to develop into three-dimensionally structured tubules. To study this specific development in a controlled environment, we used an advanced culture technique. Embryonic tissue derived from neonatal rabbit kidney was placed in a perfusion culture container at the interface of an artificial interstitium made of a polyester fleece. Culture was carried out in chemically defined Iscove’s Modified Dulbecco’s Medium (IMDM) for 13 days. Development of tubules was histochemically detected on cryosections labeled with Soybean Agglutinin (SBA). The experiments showed that aldosterone exerts a specific tubulogenic effect. Application of aldosterone (1 × 10−7 M) raised numerous SBA-labeled tubules, while in the absence of the steroid hormone the development of tubules was lacking. Specificity of hormone action was analyzed by the use of aldosterone antagonists. Administration of spironolactone (1 × 10−4 M) and canrenoate (1 × 10−5 M) completely inhibited the development of tubules. Finally, disrupting the intracellular molecular complex of the mineralocorticoid receptor (MR) and heat shock proteins by geldanamycin (2 μg/ml) prevented the development of tubules. Our results suggest that the tubulogenic effect induced by aldosterone is attributed to both hormone binding and an undisturbed intracellular response of the MR.  相似文献
