Drought is a major limiting factor for barley production, especially in the primary areas of its cultivation. Wild barley represents a major source of favourable alleles for increasing the genetic variation for multiple traits including resistance to both biotic and abiotic stresses. We used advanced backcross quantitative trait locus (AB-QTL) analysis of a BC3-doubled haploid population developed between the cultivated parent Brenda (Hordeum vulgare ssp. vulgare) and the wild accession HS584 (H. vulgare ssp. spontaneum) to study the contribution of wild barley in improving various agronomic and seed quality traits under post-anthesis drought. The experiment was carried out at two different locations (IPK, Gatersleben and Nordsaat, Böhnshausen) and terminal drought was imposed by withholding water or spraying with potassium iodide at 10 days after flowering under greenhouse or field conditions, respectively. QTL analysis indicated that wild barley contributed favourably to most of the traits studied under both control and drought conditions. A total of seven hot-spot QTL regions with co-localizing QTL for various traits harboured more than 80 % of the stable QTL detected in the present study. For yield and thousand-grain weight and their respective drought tolerance indices, most of the QTL were derived from Brenda. On the other hand, for traits like seed length and seed nitrogen content, all the QTL were contributed by HS584, the parent having higher trait value. A significantly reduced carbon/nitrogen (C/N) ratio in the selected contrasting inferior lines compared to superior ones suggests that C/N ratio could be a potential parameter for screening not just seed quality parameters but also grain weight performance under terminal drought.  相似文献

In order to characterize the functional diversity in CYP63 cluster of tandemly linked P450 genes (pc-1, pc-2, and pc-3) in Phanerochaete chrysosporium, here we report the functional characterization of pc-3 (CYP63A3), a newly cloned member of this group. pc-3 expression was favored in nutrient-limited versus nutrient-rich media in 3–6-day-old cultures and was upregulated by starch as a carbon source or by oxygenation of cultures. pc-3 was induced by various xenobiotics in defined nutrient-limited (3–9-fold) and nutrient-rich (2–5-fold) cultures. Particularly, a range of unsubstituted and substituted aliphatic hydrocarbons (alkanes and fatty acids) induced the expression under the two nutrient conditions albeit in a differential manner. Interestingly, pc-3 was also inducible by certain oxygenated mono aromatics (nitrophenol, benzoate, and resorcinol), lower molecular weight (2 to 4 ring size) polycyclic aromatic hydrocarbons (PAHs) and alkali-treated lignin derivatives in nutrient-rich malt extract cultures. The study further establishes that the three CYP63 genes (CYP63A1, A2, and A3) are independently regulated despite being members of the tandem gene cluster with high gene structural similarity (13–14 introns) and protein sequence homology (59–85%). The pc-3 cDNA (1,812 bp) was expressed in E. coli as a His-tagged protein (∼ 74 kDa). This constitutes the first report on heterologous expression of a P450 monooxygenase enzyme from this model white-rot fungus.  相似文献

Background

Progranulin is an epithelial tissue growth factor (also known as proepithelin, acrogranin and PC-cell-derived growth factor) that has been implicated in development, wound healing and in the progression of many cancers. The single mammalian progranulin gene encodes a glycoprotein precursor consisting of seven and one half tandemly repeated non-identical copies of the cystine-rich granulin motif. A genome-wide duplication event hypothesized to have occurred at the base of the teleost radiation predicts that mammalian progranulin may be represented by two co-orthologues in zebrafish.

Results

The cDNAs encoding two zebrafish granulin precursors, progranulins-A and -B, were characterized and found to contain 10 and 9 copies of the granulin motif respectively. The cDNAs and genes encoding the two forms of granulin, progranulins-1 and -2, were also cloned and sequenced. Both latter peptides were found to be encoded by precursors with a simplified architecture consisting of one and one half copies of the granulin motif. A cDNA encoding a chimeric progranulin which likely arises through the mechanism of trans-splicing between grn1 and grn2 was also characterized. A non-coding RNA gene with antisense complementarity to both grn1 and grn2 was identified which may have functional implications with respect to gene dosage, as well as in restricting the formation of the chimeric form of progranulin. Chromosomal localization of the four progranulin (grn) genes reveals syntenic conservation for grna only, suggesting that it is the true orthologue of mammalian grn. RT-PCR and whole-mount in situ hybridization analysis of zebrafish grns during development reveals that combined expression of grna and grnb, but not grn1 and grn2, recapitulate many of the expression patterns observed for the murine counterpart. This includes maternal deposition, widespread central nervous system distribution and specific localization within the epithelial compartments of various organs.

Conclusion

In support of the duplication-degeneration-complementation model of duplicate gene retention, partitioning of expression between grna and grnb was observed in the intermediate cell mass and yolk syncytial layer, respectively. Taken together these expression patterns suggest that the function of an ancestral grn gene has been devolved upon four Paralogues in zebrafish.  相似文献

This article highlights recent advances in the design of noninvasive neural interfaces based on the scalp electroencephalogram (EEG). The simplest of physical tasks, such as turning the page to read this article, requires an intense burst of brain activity. It happens in milliseconds and requires little conscious thought. But for amputees and stroke victims with diminished motor-sensory skills, this process can be difficult or impossible. Our team at the University of Maryland, in conjunction with the Johns Hopkins Applied Physics Laboratory (APL) and the University of Maryland School of Medicine, hopes to offer these people newfound mobility and dexterity. In separate research thrusts, were using data gleaned from scalp EEG to develop reliable brainmachine interface (BMI) systems that could soon control modern devices such as prosthetic limbs or powered robotic exoskeletons.  相似文献

Background  

The Actinomycete Actinosynnema pretiosum ssp. auranticum has commercial importance due to its production of ansamitocin P-3 (AP-3), a potent antitumor agent. One way to increase AP-3 production would be to constitutively express selected genes so as to relieve bottlenecks in the biosynthetic pathway; however, an integrative expression vector for A. pretiosum is lacking. Gut microbiota carry out key functions in health and participate in the pathogenesis of a growing number of diseases. The aim of this study was to develop a custom microarray that is able to identify hundreds of intestinal bacterial species. We used the Entrez nucleotide database to compile a data set of bacterial 16S rRNA gene sequences isolated from human intestinal and fecal samples. Identified sequences were clustered into separate phylospecies groups. Representative sequences from each phylospecies were used to develop a microbiota microarray based on the Affymetrix GeneChip platform. The designed microbiota array contains probes to 775 different bacterial phylospecies. In our validation experiments, the array correctly identified genomic DNA from all 15 bacterial species used. Microbiota array has a detection sensitivity of at least 1 pg of genomic DNA and can detect bacteria present at a 0.00025% level of overall sample. Using the developed microarray, fecal samples from two healthy children and two healthy adults were analyzed for bacterial presence. Between 227 and 232 species were detected in fecal samples from children, whereas 191 to 208 species were found in adult stools. The majority of identified phylospecies belonged to the classes Clostridia and Bacteroidetes. The microarray revealed putative differences between the gut microbiota of healthy children and adults: fecal samples from adults had more Clostridia and less Bacteroidetes and Proteobacteria than those from children. A number of other putative differences were found at the genus level.In the healthy adult, there are 1011 to 1014 bacteria colonizing the intestine. This outnumbers the total tissue cells in the body by at least an order of magnitude. The composition and activity of this complex microbial system (called microbiota or microflora) have a major influence on health and disease (9). Commensal microbiota contribute to the trophic functions of the gut (producing fermentation products and vitamins that can be used by intestinal epithelial cells), stimulate the immune function of the gastrointestinal tract, transform or excrete toxic substances, protect the host against invasion by pathogenic species, and modulate gut motility (28, 36). At the same time, recent research incriminates a dysfunctional cross-talk between the host and the microbiota in the pathogenesis of a growing number of disorders, such as irritable bowel syndrome, inflammatory bowel disease, allergic diseases, and gastrointestinal cancer (28).While the intestine in a newborn contains no microbes, immediately after birth the intestine of the infant is colonized by enterobacteria and enterococci. Gradual changes in microbiota composition occur during childhood, with a general reduction in the number of aerobes and facultative anaerobes and an increase in the populations of obligate anaerobic species (27). It is considered that by 2 years of age the microbiota resembles that of an adult, which is dominated in health and disease by species from only four phyla, Firmicutes (predominantly Clostridia; 50 to 70% total bacterial numbers), Bacteroidetes (10 to 30%), Proteobacteria (up to 10%), and Actinobacteria (up to 5%), with 90% believed to be obligate anaerobes (4, 10, 11, 22).Traditionally, microorganisms were detected in intestinal samples and feces by microscopic, biochemical, or physiological methods, or by culturing on selective nutrient media. However, since most intestinal microbiota species are obligate anaerobes, their isolation and culturing are difficult (21, 38, 42). In recent years, new methods based on the use of microarray technology have been utilized for the characterization of complex microbial communities (18, 32, 41, 43, 47). Microarrays represent an excellent choice for the high-throughput analysis of bacterial populations, because many different probes can be placed on one slide or synthesized on one chip, and samples thus can be tested for the presence of many different species simultaneously. Environmental and clinical samples can be interrogated directly, circumventing any need for culturing, and thus nonculturable species can be reliably detected.Several types of microarrays have been used to date to characterize the composition of microbial communities (47). Community genome arrays are constructed using whole genomic DNA (gDNA) isolated from pure culture strains (46). Functional gene arrays contain genes encoding key enzymes that are involved in various biochemical processes, and they are useful for monitoring physiological changes in microbial communities (14, 45). Phylogenetic oligonucleotide arrays contain probes derived from rRNA sequence information and are ideally suited for the analysis of microbial community composition structure and variance. Different types of phylogenetic arrays have been designed for these purposes (26, 30, 31).A number of projects performed in the last several years focused on sampling the diversity of human microbiota by the cloning and subsequent sequencing of the 16S rRNA genes isolated from gastrointestinal and fecal samples (5, 10, 13, 23, 38). In this project, we have designed, developed, and validated a custom microbiota microarray containing 16S rRNA genes probes to 775 different microbial phylospecies of human intestinal bacteria. We also have tested the applicability of this array to profiling the microbiota populations in fecal samples isolated from two adult and two child volunteers.  相似文献

The regulation of cellular processes by the modulation of protein phosphorylation/dephosphorylation is fundamental to a large number of processes in living organisms. These processes are carried out by specific protein kinases and phosphatases. In this study, a previously uncharacterized gene (Rv0018c) of Mycobacterium tuberculosis, designated as mycobacterial Ser/Thr phosphatase (mstp), was cloned, expressed in Escherichia coli, and purified as a histidine-tagged protein. Purified protein (Mstp) dephosphorylated the phosphorylated Ser/Thr residues of myelin basic protein (MBP), histone, and casein but failed to dephosphorylate phospho-tyrosine residue of these substrates, suggesting that this phosphatase is specific for Ser/Thr residues. It has been suggested that mstp is a part of a gene cluster that also includes two Ser/Thr kinases pknA and pknB. We show that Mstp is a trans-membrane protein that dephosphorylates phosphorylated PknA and PknB. Southern blot analysis revealed that mstp is absent in the fast growing saprophytes Mycobacterium smegmatis and Mycobacterium fortuitum. PknA has been shown, whereas PknB has been proposed to play a role in cell division. The presence of mstp in slow growing mycobacterial species, its trans-membrane localization, and ability to dephosphorylate phosphorylated PknA and PknB implicates that Mstp may play a role in regulating cell division in M. tuberculosis.  相似文献
