Aims
Shallow soils on acidic bedrock in dry areas of Central Europe support dry grasslands and heathlands that were formerly used as extensive pastures. These habitats are of high conservation value, but their abandonment in the 20th century triggered slow natural succession that poses a threat to specialized plant species. We asked how this vegetation and its plant diversity have changed over the past three decades and whether protected areas have positively affected habitat quality.Location
Southwestern and central Moravia, Czech Republic.Methods
In 2018–2019, we resurveyed 94 vegetation plots first sampled in 1986–1991 at 47 acidic dry grassland and heathland sites. We compared the number of all vascular plant species, Red List species and alien species per plot using parametric and non-parametric tests, life-form spectra using the chi-square test, species composition using detrended correspondence analysis, and indicator values using a permutation test. We also compared these changes between sites within and outside protected areas.Results
Vegetation changes over the past three decades have been relatively small. However, we detected a decrease in total species richness, the number of Red List species and the number of characteristic species of dry grasslands. Neophytes were infrequent, while archaeophytes increased slightly. The competitive tall grass Arrhenatherum elatius, annual species and young woody plants increased in abundance or newly established at many sites. Indicator values did not change except for a slight increase in nutrient values. These negative trends occurred both within and outside protected areas but were more pronounced outside.Conclusions
Formerly grazed acidic dry grasslands and heathlands in Moravia are slowly losing habitat specialists, including threatened plant species, and are increasingly dominated by Arrhenatherum elatius. Conservation management, especially cutting in protected areas, slows down the negative trends of decline in plant diversity and habitat quality but is insufficient to halt these processes completely. 相似文献3.

The complex and dynamic pattern of Hoxb3 expression in the developing hindbrain and the associated neural crest of mouse embryos is controlled by three separate cis-regulatory elements: element I (region A), element IIIa, and the r5 enhancer (element IVa). We have examined the cis-regulatory element IIIa by transgenic and mutational analysis to determine the upstream trans-acting factors and mechanisms that are involved in controlling the expression of the mouse Hoxb3 gene in the anterior spinal cord and hindbrain up to the r5/r6 boundary, as well as the associated neural crest which migrate to the third and posterior branchial arches and to the gut. By deletion analysis, we have identified the sequence requirements within a 482-bp element III482. Two Hox binding sites are identified in element III482 and we have shown that in vitro both Hoxb3 and Hoxb4 proteins can interact with these Hox binding sites, suggesting that auto/cross-regulation is required for establishing the expression of Hoxb3 in the neural tube domain. Interestingly, we have identified a novel GCCAGGC sequence motif within element III482, which is also required to direct gene expression to a subset of the expression domains except for rhombomere 6 and the associated neural crest migrating to the third and posterior branchial arches. Element III482 can direct a higher level of reporter gene expression in r6, which led us to investigate whether kreisler is involved in regulating Hoxb3 expression in r6 through this element. However, our transgenic and mutational analysis has demonstrated that, although kreisler binding sites are present, they are not required for the establishment or maintenance of reporter gene expression in r6. Our results have provided evidence that the expression of Hoxb3 in the neural tube up to the r5/r6 boundary is auto/cross-regulated by Hox genes and expression of Hoxb3 in r6 does not require kreisler. 相似文献

The interactions of five different lectins: peanut (PNA), lentil (LEN), wheat germ (WGA), soybean (SBA), Asparagus pea (FBP) with leukaemic cells obtained from 31 children: 25 with acute lymphoblastic leukaemia (ALL) and 6 with acute myeloblastic leukaemia (AML) were examined in this study. The relationship of lectin-binding ability to cells cytomorphological, cytochemical and immunological features and its potential clinical application were investigated. It has been shown that PNA and LEN receptors were found in the majority of blast cells. The SBA reacting cells were found only in few patients and FBP binding was not found in studied ALL and AML cells. There was a clear difference in the WGA binding capacity in ALL cells with L1 and L2 characteristics respectively. No differences were found in PNA. WGA and LEN reactivity between PAS negative and PAS positive leukaemic cells. Only PNA of all studied lectins seemed to differentiate T- from B-ALL blast cells. Only WGA binding of ALL cells showed the positive correlation to the risk index value. 相似文献

Expression of nonspecific cross-reacting antigen species in myeloid leukemic patients and healthy subjects 总被引:1,自引:0,他引:1
The reactivity of two monoclonal antibodies recognizing NCA-95 and NCA-55 (MAb 47 and MAb 192, respectively) with a polyclonal anti-NCA serum in myeloid leukemic cells isolated by density gradient centrifugation was compared using an immunofluorescence test (IF). It was observed that the blood myeloid cells in 78.8% of the patients with different types of myelocytic leukemias and all granulocytes of 15 normal donors showed similar expression of the NCA species studied. The leukocytes of the remaining patients did not synthesize the NCA-95 species regardless of the maturation stage of the cells studied. In two patients, synthesis of this NCA form was limited to the fractions containing myelocytes and metamyelocytes. We have found that all anti-NCA antibodies studied recognized different antigenic epitopes in a myeloid cell series. A relationship between the patient's survival and the proportion of NCA-containing cells was also observed. 相似文献
