The 23-kDa nuclear-encoded heat-shock protein (HSP) of Chenopodium rubrum L. is regulated by light at the posttranslational level. Higher light intensities are more effective in inducing the accumulation of the mature protein under heat-shock conditions. Based on this and other properties the protein was considered to belong to the group of small chloroplastic HSPs. However, we have now obtained the following evidence that this 23-kDa HSP is localized in the mitochondria: (i) Immunogold-labelled protein was almost exclusively restricted to the mitochondria in electron microscope thin sections. (ii) Using purified, isolated mitochondria from potato tubers the in-vitro-synthesized translation product of 31 kDa was readily transported into mitochondria where it was processed to the 23-kDa product. (iii) The protein could be detected by Western blotting in a preparation of washed mitochondria of Chenopodium, while under the same conditions no signal could be obtained in a preparation of isolated chloroplasts. (iv) Finally, sequence comparison with the published sequences of mitochondrial proteins by Lenne et␣al. (1995, Biochem J 311:805–813) and LaFayette et␣al. (1996, Plant Mol Biol 30:159–169) showed clearly that the 23-kDa protein is considerably more similar to these two proteins than to the group of plastid small HSPs. Beutenbergia cavernae (Groth et al. 1999) is the type species of the genus and is of phylogenetic interest because of its isolated location in the actinobacterial suborder Micrococcineae. B. cavernae HKI 0122(T) is a Gram-positive, non-motile, non-spore-forming bacterium isolated from a cave in Guangxi (China). B. cavernae grows best under aerobic conditions and shows a rod-coccus growth cycle. Its cell wall peptidoglycan contains the diagnostic L-lysine ← L-glutamate interpeptide bridge. Here we describe the features of this organism, together with the complete genome sequence, and annotation. This is the first completed genome sequence from the poorly populated micrococcineal family Beutenbergiaceae, and this 4,669,183 bp long single replicon genome with its 4225 protein-coding and 53 RNA genes is part of the Genomic Encyclopedia of Bacteria and Archaea project.  相似文献

 Responses of stem-volume growth to N application were evaluated in relation to foliar N concentrations. Data from N-fertilization experiments in 28 Pinus sylvestris stands and 21 Picea abies stands were used. Relative stem-growth responses were negatively related to concentrations of N in current-year needles of unfertilized trees. There appeared to be a threshold value of 15–16 mg (g DM)–1 N in current-year needles, above which N-application is unlikely to stimulate growth. However, relations were non-significant between N concentrations in current-year needles and the absolute stem-growth response [dm3 ha–1 (5 years)–1]. Thermovirga lienii Dahle and Birkeland 2006 is a member of the genus Thermovirga in the genomically moderately well characterized phylum 'Synergistetes'. Members of this relatively recently proposed phylum 'Synergistetes' are of interest because of their isolated phylogenetic position and their diverse habitats, e.g. from humans to oil wells. The genome of T. lienii Cas60314(T) is the fifth genome sequence (third completed) from this phylum to be published. Here we describe the features of this organism, together with the complete genome sequence and annotation. The 1,999,646 bp long genome (including one plasmid) with its 1,914 protein-coding and 59 RNA genes is a part of the Genomic Encyclopedia of Bacteria and Archaea project.  相似文献

Sponges (phylum Porifera) are sessile marine filter feeders that have developed efficient defense mechanisms against foreign attackers such as viruses, bacteria, or eukaryotic organisms. Protected by a highly complex immune system, as well as by the capacity to produce efficient antiviral compounds (e.g., nucleoside analogues), antimicrobial compounds (e.g., polyketides), and cytostatic compounds (e.g., avarol), they have not become extinct during the last 600 million years. It can be assumed that during this long period of time, bacteria and microorganisms coevolved with sponges, and thus acquired a complex common metabolism. It is suggested that (at least) some of the bioactive secondary metabolites isolated from sponges are produced by functional enzyme clusters, which originated from the sponges and their associated microorganisms. As a consequence, both the host cells and the microorganisms lost the ability to grow independently from each other. Therefore, it was—until recently—impossible to culture sponge cells in vitro. Also the predominant number of symbiotic bacteria proved to be nonculturable. In order to exploit the bioactive potential of both the sponge and the symbionts, a 3D-aggregate primmorph culture system was established; also it was proved that one bioactive compound, avarol/avarone, is produced by the sponge Dysidea avara. Another promising way to utilize the bioactive potential of the microorganisms is the cloning and heterologous expression of enzymes involved in secondary metabolism, such as the polyketide synthases. Forest N fertilization is a common practice in areas of Sweden that are not affected by high levels of N deposition. The environmental consequences of high N input to closed forests are fairly well known, but the long-term effects following clear-felling are a lot less well known. Thus, residual effects on soil and planted seedlings of previous N additions at an experimental N gradient 11 years after clear-felling were studied at a naturally nutrient-poor forest site in central Sweden. The experimental N gradient had been established by three repeated applications (in 1967, 1974 and 1981) of six dosages of NH4NO3 with increments of 120 kg N ha–1. Thus, in total, the applied N dose ranged between 0 and 1800 kg N ha–1. The study examined extractable base cations and P, soil pH, total-N, total-C, net N-mineralization and potential nitrification in four soil horizons (the humus layer, and 0–5, 5–10 and 10–20 cm in the mineral soil). We also measured the survival and growth of planted Pinus sylvestris L. seedlings. The applied N had no effect on the amounts of extractable-P or base cations in the soil. The soil pH decreased with increasing N dose in the deeper soil horizons, while in the humus the pH showed a weak but statistically significant increase due to the N application. Both total-C and total-N increased as a result of the N application, while the C/N ratio decreased. In the humus layer and the uppermost mineral soil layer NH4 + was the major inorganic N source, in contrast to the lowest mineral soil horizon where NO3 dominated. For most of the studied horizons, there was a positive linear relationship between applied N dose and amount of inorganic N. Both net N-mineralization and potential nitrification showed increases with increasing N dose. As for the plants, no difference in survival or growth was found between the different N treatments. For doses generally applied in forest fertilization no significant differences in any of the studied properties were found.  相似文献
