Background
Populations of the Oriental White-backed Vulture (Gyps bengalensis) have declined by over 95% within the past decade. This decline is largely due to incidental consumption of the non-steroidal anti-inflammatory veterinary pharmaceutical diclofenac, commonly used to treat domestic livestock. The conservation status of other Gyps vultures in southern Asia is also of immediate concern, given the lack of knowledge regarding status of their populations and the continuing existence of taxonomic uncertainties. In this study, we assess phylogenetic relationships for all recognized species and the majority of subspecies within the genus Gyps. The continuing veterinary use of diclofenac is an unknown but potential risk to related species with similar feeding habits to Gyps bengalensis. Therefore, an accurate assessment of the phylogenetic relationships among Gyps vultures should aid in their conservation by clarifying taxonomic uncertainties, and enabling inference of their respective relatedness to susceptible G. bengalensis. 相似文献2.

This report describes additional studies of the lyo and desmo components of esterase, alkaline phosphatase, acid phosphatase, leucine aminopeptidase, and β-glucuronidase. The techniques used have already been reported (7). Enzyme diffusion occurs to different degrees in different fixatives, and varies somewhat with each enzyme. Loss of enzymatic activity during fixation occurs as a result of both inactivation due to the chemical reaction of the fixative with the enzymic protein, and diffusion of the lyo component into the fixative. The amount of diffusion into formalin can be reduced by the addition of salts, sucrose, or methocel. The pH of the aqueous medium significantly influences the removal of the lyo fraction from the tissue section. A striking similarity can be noted in the proportions of each fraction of enzyme present in the kidney of the rat, dog, and man. The procedure of fixation and paraffin embedding of tissue blocks does not wholly prevent the diffusion of the lyo component from the tissue sections when they are subsequently immersed in the aqueous incubation medium. 相似文献

alpha 1-Proteinase inhibitor (alpha 1-PI), a member of the serine
proteinase inhibitor superfamily, has a primary role in controlling
neutrophil elastase activity within the mammalian circulation. Several
studies have indicated that the reactive center region of alpha 1-PI, the
amino acid sequence of which is critical to recognition of and binding to
target proteinases, is highly divergent within and among species. This
appears to be a consequence of accelerated rates of evolution that may have
been driven by positive Darwinian selection. In order to examine this and
other features of alpha 1-PI evolution in more detail, we have isolated and
sequenced cDNAs representing alpha 1- PI mRNAs of the mouse species Mus
saxicola and Mus minutoides and have compared these with a number of other
mammalian alpha 1-PI mRNAs. Relative to other mammalian mRNAs, the extent
of nonsynonymous substitution is generally high throughout the alpha 1-PI
mRNA molecule, indicating greater overall rates of amino acid substitution.
Within and among mouse species, the 5'-half of the mRNA, but not the
3'-half, has been homogenized by concerted evolution. Finally, the reactive
center is under diversifying or positive Darwinian selection in murid
rodents (rats, mice) and guinea pigs yet is under purifying selection in
primates and artiodactyls. The significance of these findings to alpha 1-PI
function and the possible selective forces driving evolution of serpins in
general are discussed.

Patterns of mitochondrial DNA (mtDNA) variation were examined in 133
mole-rats constituting all four chromosomal species (2n = 52, 2n = 54, 2n =
58, and 2n = 60) of the Spalax ehrenbergi superspecies in Israel, as well
as the peripheral isolates of 2n = 60. In the main range of the complex, a
total of 28 mtDNA haplotypes were found in 64 mole-rats, with most
haplotypes being unique to either a single chromosomal species or
population. mtDNA divergence increased from low to high diploid number in a
north-to-south direction in Israel. Overall levels of mtDNA diversity were
unexpectedly the highest in the 2n = 60, the youngest species of the
complex. The mtDNA haplotypes can be separated into two major groups, 2n =
52-54 and 2n = 58-60, and a phylogenetic analysis for each group revealed
evidence of a few haplotypes not sorted by diploid number. The overall
patterns of mtDNA divergence seen within and among the four chromosomal
species are consistent with the parapatric mode of speciation as suggested
from previous studies of allozyme and DNA hybridization. In a separate data
set the patterns of mtDNA variation were examined across the main
geographic range and across peripheral semi-isolates and isolates of the 2n
= 60 chromosomal species. Fifteen haplotypes were found in 69 mole-rats.
High levels of mtDNA diversity characterized the main range, semi-isolated,
and even some desert isolated populations. The peripheral isolates contain
much mtDNA diversity, including novel haplotypes.

The effects of nitrogen, potassium, water stress, and phytohormones were studied using a Salvia stenophylla (Burch. ex Benth.) microplant system. In vitro regeneration was monitored and then followed with volatile secondary metabolite profiling
through headspace solid-phase microextraction gas chromatography. Plantlet growth was most prolific on half-strength Murashige
and Skoog medium without phytohormones. An increase in macronutrients supplied to the microplants enhanced accumulation of
the commercially important (−)-α-bisabolol, while no significant changes to the relative abundance of β-bisabolene, α-muurolene,
α-patchoulene, and D-limonene (among others) became apparent. Water-stressed plants, treated with sorbitol and polyethylene
glycol, had a lowered rooting capacity in vitro. Overall, as a plant production system, micropropagation did not have deleterious
effects on the biochemistry of S. stenophylla as no significant differences in metabolic profiles existed between conventional garden plants and in vitro propagules, regardless
of phytohormone treatment. We also show that nutrient manipulation can be used efficiently as a strategy for positively altering
secondary metabolism. This will ultimately benefit the domestication of this commercially important medicinal herb. 相似文献

Multiple Sclerosis (MS) is a progressive autoimmune inflammatory and demyelinating disease of the central nervous system (CNS). T cells play a key role in the progression of neuroinflammation in MS and also in the experimental autoimmune encephalomyelitis (EAE) animal models for the disease. A technology for quantitative and 3 dimensional (3D) spatial assessment of inflammation in this and other CNS inflammatory conditions is much needed. Here we present a procedure for 3D spatial assessment and global quantification of the development of neuroinflammation based on Optical Projection Tomography (OPT). Applying this approach to the analysis of rodent models of MS, we provide global quantitative data of the major inflammatory component as a function of the clinical course. Our data demonstrates a strong correlation between the development and progression of neuroinflammation and clinical disease in several mouse and a rat model of MS refining the information regarding the spatial dynamics of the inflammatory component in EAE. This method provides a powerful tool to investigate the effect of environmental and genetic forces and for assessing the therapeutic effects of drug therapy in animal models of MS and other neuroinflammatory/neurodegenerative disorders. 相似文献

The brain's biological clock, which, in mammals, is located in the suprachiasmatic nucleus (SCN), generates circadian rhythms in behaviour and physiology. These biological rhythms are adjusted daily (entrained) to the environmental light/dark cycle via a monosynaptic retinofugal pathway, the retinohypothalamic tract (RHT). In this review, the anatomical and physiological evidence for glutamate and pituitary adenylate cyclase-activating polypeptide (PACAP) as principal transmitters of the RHT will be considered. A combination of immunohistochemistry at both the light- and electron-microscopic levels and tract-tracing studies have revealed that these two transmitters are co-stored in a subpopulation of retinal ganglion cells projecting to the retino-recipient zone of the ventral SCN. The PACAP/glutamate-containing cells, which constitute the RHT, also contain a recently identified photoreceptor protein, melanopsin, which may function as a "circadian photopigment". In vivo and in vitro studies have shown that glutamate and glutamate agonists such as N-methyl- D-aspartate mimic light-induced phase shifts and that application of glutamate antagonists blocks light-induced phase shifts at subjective night indicating that glutamate mediates light signalling to the clock. PACAP in nanomolar concentrations has similar phase-shifting capacity as light and glutamate, whereas PACAP in micromolar concentrations modulates glutamate-induced phase shifts. Possible targets for PACAP and glutamate are the recently identified clock genes Per1 and Per2, which are induced in the SCN by light, glutamate and PACAP at night. 相似文献
