- Recent evidence suggests that periphytic algae stimulate plant litter heterotrophs (fungi and bacteria) in the presence of light, but few studies have tested whether this stimulation varies across gradients of light, which may covary with temperature.
- We exposed field-conditioned Typha domingensis litter to fully-crossed, short-term gradients of temperature (15, 20, 25, and 30°C) and light (0, 25, 53, 123, and 388 µmol quanta m−2 s−1) and measured responses of litter-associated algal, fungal, and bacterial production rates and β-glucosidase, β-xylosidase, and phenol oxidase enzyme activities in the laboratory.
- Increased light stimulated algal production rates, from immeasurable production under darkness to >200 µg algal C g−1 detrital C hr−1 at the highest light level, with the greatest light sensitivity and maximal photosynthetic rates at 25°C. In turn, increased light stimulated fungal production rates, especially at the two highest temperatures and most strongly at 25°C where light stimulated fungal production by a mean of 65 µg C g−1 detrital C hr−1, indicating 2.1-fold stimulation by light. Bacterial production rates also responded to light, indicated by stimulation of a mean of 16 µg C g−1 detrital C hr−1 (1.6-fold) at 15°C, but stimulation was weaker at higher temperatures. Enzyme activities increased strongly with elevated temperature but were not affected by light.
- Our experimental evidence suggests algae differentially stimulate litter-associated bacteria and fungi in a light-dependent manner that further depends on temperature. These findings advance understanding of the onset of algal stimulation of heterotrophy, including algal-induced priming effects during litter decomposition, in response to common covarying environmental gradients subject to global change.

The effective molecular weight cut-off values of dialysis membranes for carrageenan and alginate oligosaccharides were evaluated by gel permeation chromatography and nuclear magnetic resonance spectroscopy. For the different membranes tested, i.e. Medicell, Spectra Por 1000D and 3500D, the porous sizes are analogous to tri- and tetrasaccharides. More than 100 BOX elements are randomly distributed in intergenic regions of the pneumococcal genome. Here we demonstrate that these elements can affect expression of neighboring genes and present evidence that they are mobile. We have hypothesized a state of zinc deficiency in sickle cell disease (SCD). This could at least partially explain the growth problems, hypogonadism, and slow healing leg ulcers associated with SCD. Preliminary findings revealed abnormally low red blood cell zinc levels in 10 of 16 patients studied. Before suggesting zinc supplementation in SCD we thought it important to look at the effect of zinc on red cell metabolism and function. It was found that zinc chloride added to normal and SCD blood to a final concentration of 1.5 × 10?3 M caused a left-shift of the blood oxygen affinity curve (increased oxygen affinity) varying from 1.5 to 3.5 mm Hg change in half saturation (p50). This curve shifting property has important implications for SCD since recent work with cyanate suggests that such shifts are very beneficial in treatment of SCD. Thus zinc supplementation in SCD, in addition to its potential role in correcting wound healing and growth problems, may have a beneficial effect on the basic pathological process. Data are given which suggest that zinc and 2, 3-diphosphoglycerate may not be competing for the same site on the hemoglobin molecule. 相似文献

- The transport of guanine in autospores of light-dark synchronizedChlorella fusca was studied using radioactive guanine in the concentration range of 4 nM to 50 μM.
- The transport system was constitutive, it had high specificity for the permeant, and theQ 10 value was in the range of 1.5 to 2.2. At concentrations lower than 0.2 μM the half saturating constant, S0.5 was 1 μM both for cells kept in dark and cells kept in light. At higher concentrations the S0.5 of darkened cells was about 0.23 μM, while that of illuminated cells was unchanged. Only above 0.2 μM guanine did illumination of the cells or addition of glucose increase the transport rate.
- Guanine which had accumulated did not leak out at temperatures below 45°C or by treatment with 10 μM dinitrophenol, which completely inhibited transport. Furthermore, the accumulated guanine did not exchange with exogenous guanine.
- The guanine accumulated, more than 105-fold over the external concentration, showing that the transport, was active.
- The initial transport rate per cell revealed annual fluctuations.

Microfibril-associated glycoprotein (MAGP) 1 and 2 are evolutionarily related but structurally divergent proteins that are components of microfibrils of the extracellular matrix. Using mice with a targeted inactivation of Mfap5, the gene for MAGP2 protein, we demonstrate that MAGPs have shared as well as unique functions in vivo. Mfap5−/− mice appear grossly normal, are fertile, and have no reduction in life span. Cardiopulmonary development is typical. The animals are normotensive and have vascular compliance comparable with age-matched wild-type mice, which is indicative of normal, functional elastic fibers. Loss of MAGP2 alone does not significantly alter bone mass or architecture, and loss of MAGP2 in tandem with loss of MAGP1 does not exacerbate MAGP1-dependent osteopenia. MAGP2-deficient mice are neutropenic, which contrasts with monocytopenia described in MAGP1-deficient animals. This suggests that MAGP1 and MAGP2 have discrete functions in hematopoiesis. In the cardiovascular system, MAGP1;MAGP2 double knockout mice (Mfap2−/−;Mfap5−/−) show age-dependent aortic dilation. These findings indicate that MAGPs have shared primary functions in maintaining large vessel integrity. In solid phase binding assays, MAGP2 binds active TGFβ1, TGFβ2, and BMP2. Together, these data demonstrate that loss of MAGP2 expression in vivo has pleiotropic effects potentially related to the ability of MAGP2 to regulate growth factors or participate in cell signaling. 相似文献

In field experiments, clones of Norway spruce [Picea abies (L.) Karst.] showed different degrees of resistance against pathogenic fungi inoculated into the bark that correlate with
differences in polyphenolic parenchyma (PP) cells of the bark. Cells of spruce callus cultures, particularly towards the callus
surface, resemble PP cells and this study looks at changes in callus cells during infection and the relative resistance of
cultures from clones of low (weak) or high (strong) resistance to fungal infection. Callus cultures, initiated from trees
with different resistance, were co-inoculated with Ceratocystis polonica (Siem.) C. Moreau and Heterobasidion annosum (Fr.) Bref. Callus cells from strong clones resemble PP cells of bark tissue from strong clones, having more polyphenolic
bodies, while callus cells from weak clones are more similar to PP cells from those clones, which have less extensive phenolic
bodies. Callus cultures from trees with weak resistance were more quickly overgrown by both species of pathogenic fungi than
cultures from trees with strong resistance. Callus cells of infected cultures showed changes similar to activated PP cells
of bark, including enhanced accumulation of polyphenolics. Phenolic bodies were more numerous and more extensive (larger and
denser) in callus cells of strong versus weak clones under all conditions. Thus, callus cells may perform similar functions
in defense as PP cells in the bark. Callus from trees of varying resistance seem to reflect the relative resistance of the
trees from which they are derived, and this study indicates that some mechanisms of resistance can be studied using callus
