A new series of fluorinated and non-fluorinated 2-phenylbenzimidazoles bearing oxygenated substituents on the phenyl ring has been synthesized. Synthesis of the new series was based on our previous discovery of 2-(3,4-dimethoxyphenyl)-5-fluorobenzothiazole (PMX 610) as a potent and selective antitumour agent in vitro (sub-nanomolar GI50 in sensitive human cancer cell lines), but with poor aqueous solubility and lack of a definitive cellular target limiting further development. In this study we test the hypothesis that 2-phenylbenzimidazoles with similar substitution patterns to PMX 610 would retain potent antitumour activity but with potentially superior pharmaceutical properties. In general the new compounds were less active than the former benzothiazole series in vitro when tested against the breast cancer cell lines MCF-7 and MDA 468; however the two most active compounds in the present series (3j and 3k) exhibit low micromolar GI50 values in both cell lines and provide the opportunity for further chemical derivatization with a view to target identification.  相似文献

Experimental results show that benzil (1,2‐diphenyl‐1,2‐ethanedione), an achiral compound that crystallizes as a racemic conglomerate, yields by solidification polycrystalline scalemic mixtures of high enantiomeric excesses. These results are related to those previously reported in this type of compounds on deracemizations of racemic mixtures of crystal enantiomorphs obtained by wet grinding. However, the present results strongly suggest that these experiments cannot be explained without taking into account chiral recognition interactions at the level of precritical clusters. Microtubules are hollow cylindrical filaments of the eukaryotic cytoskeleton characterized by extremely low shear modulus. A remarkable controversy has occurred in the literature, regarding the length dependence of flexural rigidity of microtubules predicted by the classical elastic beam model. In this study, a higher order shear deformable beam model for microtubules is employed to study unexplained length-dependent flexural rigidity and Young’s modulus of microtubules reported in the literature. The formulation allows for warping of the cross-section of the microtubule and eliminates the need for using arbitrary shear correction coefficients as in other theories. It is showed that vibration frequencies predicted by the present parabolic shear deformation theory (PSDT) are much lower than that given by the approximate isotropic beam model for shorter microtubules, although the two models give almost identical results for sufficiently long microtubules. It is confirmed that transverse shearing and the warping of the cross-section of microtubules are mainly responsible for the length-dependent flexural rigidity of an isolated microtubule reported in the literature, which cannot be explained by the widely used Euler-Bernoulli beam model. Indeed, the length-dependent flexural rigidity predicted by the present model is found to be in qualitative agreement with the existing experimental data ( [Kurachi et al., 1995] and [Pampaloni et al., 2006]). These results recommend that the parabolic shear deformation-beam theory offers a unified simple 1D model, which can capture the length dependence of flexural rigidity and be applied to various static and dynamic problems of microtubule mechanics.  相似文献

In this study, we report the design, synthesis and antioxidant activity of a series of substituted 2-(4-aminophenyl)-1H-indoles and 2-(methoxyphenyl)-1H-indoles. The new compounds are structurally related to the known indole-based antioxidant lead compound melatonin (MLT), and the antitumour 2-(4-aminophenyl)benzothiazole and 2-(3,4-dimethoxyphenyl)benzothiazole series. Efficient access to the target 2-phenylindoles was achieved via Fischer indole synthesis between substituted phenylhydrazines and acetophenones. 2-(4-Aminophenyl)indoles (such as the 6-fluoro analogue 3b) in particular showed potent antioxidant activity in the DPPH and superoxide radical scavenging assays (80% and 81% inhibition at 1 mM concentration of 3b, respectively), at a level comparable with the reference standard MLT (98% and 75% at 1 mM).  相似文献

Purpose

This life cycle assessment (LCA) study compares energy consumption, greenhouse gas emissions, and environmental damages for two methods of constructing retaining structures, a traditional method involving a retaining wall backfilled with sand, and an alternative method involving a retaining wall backfilled with shredded tires.

Methods

Taking into account the extraction and production of the used construction materials, loading, transport and installation, the cumulative energy demand (CED), global warming potential (GWP), acidification potential, Human Health Criteria Air-mobile, aquatic eutrophication potential, ozone depletion potential, and smog potential is determined for each construction method. The seven environmental impact categories are calculated using the software tool, ATHENA® Environmental Impact Estimator (ATHENA® EIE) for Buildings v5.2.0118.

Results and discussion

The seven impact categories were reduced significantly by using shredded tires as retaining wall backfill; this is due to the decrease in the amounts of concrete, reinforcing steel, and fuel quantity consumed by building machines and vehicles transporting construction materials.

Conclusions

The study concludes that in all examined impact categories alternative method provides a larger environmental benefit than the traditional method. Also, the results clearly demonstrate that the use of shredded tires is very effective as a sustainable alternative to retaining structures.
