The internal pH (pHi) of Toxoplasma gondii was estimated by measuring the accumulation of the weak base 9-aminoacridine in buffers with various ionic compositions. The pHi of the metabolizing parasite increased when the extracellular K+ was elevated in alkaline medium or when the external pH (pHc) was substantially increased in medium employing high external K+ (90 mM). The parasite in mouse peritoneal fluid, or in potassium sulfate buffer (pH 8.2), where the pHi was demonstrated to be increased to 7.9, became motile when acidic buffer was substituted for the original suspension medium. This acid-induced independent movement subsided within 5 min but was repeatedly induced if the pHc was serially lowered to 6.0. Basic buffers, on the other hand, abolished motility when applied to the moving parasites. Nigericin, which is known to collapse pH gradients across the membrane, also abolished motility.  相似文献

Quantitative trait loci (QTL) analysis of aluminium (Al) tolerance was performed using Ler/Cvi recombinant inbred (RI) lines of Arabidopsis thaliana. Relative root length (RRL) (root length with 4 µm Al/root length with no Al at pH 5.0) on day 5 was used as the Al tolerance index for QTL analysis. Al tolerance judged by RRL was well correlated to tolerance judged by other indexes, including accumulation of callose, reactive oxygen species in the root apex and growth performance on acid soil containing a large amount of exchangeable Al. Using data sets with an hb2 of 0.91, two QTLs were detected at the top of chromosome 1 and bottom of chromosome 3. These QTLs explained 40 and 16% of the phenotypic variation of Al tolerance, respectively, and the positive effect of the Cvi allele. The QTL on chromosome 1 overlapped with a major QTL in another recombinant inbred population, and is possibly related to malate excretion. A complete pair-wise search revealed 11 sets of epistatic interacting loci pairs, which accounted for the transgressive segregation among the RI population. Several epistatic interactions shared the same chromosomal region, indicating the possible involvement of regulatory proteins in Al tolerance in Arabidopsis.  相似文献

蛋白质在生物体内电荷转移过程中所起的作用迄今仍然是一个有争议的问题.其争论焦点是蛋白质在生物电荷转移过程中是否提供特殊的电子传递通道或者是仅仅作为普通的有机介质.应用飞秒时间分辨瞬态吸收光谱研究由光合细菌天线分子和平均粒径为8 nm的TiO2组装而成的超分子系统中长程电荷转移.晶体结构研究表明,光合细菌天线分子具有由多个α-脱辅基和β-脱辅基蛋白跨膜螺旋构成的双层空心柱面体结构,其中α-脱辅基蛋白跨膜螺旋构成的小环状体套于β-脱辅基蛋白跨膜螺旋构成的大环状体中.小环状体的空腔直径约为3.6 nm.光合色素细菌叶绿素和β-胡萝卜素分子处于两环之间.细菌叶绿素距离外周胞质膜最近,预计为1 nm.本研究试图将TiO2纳米颗粒部分装入光合细菌膜蛋白的腔体中,探讨细菌叶绿素与TiO2纳米颗粒间进行的光致长程电荷转移,进而揭示蛋白质在电荷转移过程中所起的作用.实验观察到细菌叶绿素B850在LH2/TiO2中的基态漂白恢复的时间常数明显地比在LH2中短,应用长程电荷转移模型,将蛋白质视为普通介电媒体,由电荷转移速率推算得到细菌叶绿素与TiO2纳米颗粒最近表面的距离为0.6 nm,表明TiO2纳米颗粒已经成功地部分装入光合细菌天线分子的空腔中.  相似文献

  1. 1) When yeast cells grown anaerobically were adapted to aerobicculture in a normal medium, catalase formation was markedlyenhanced after the earlier stage of exponential growth of thecells. The same thing occurred with cells transferred from ananaerobic culture into a nitrogen deficient medium.
  2. 2) Thecatalase activity of aerobically grown cells declinedprogressivelyuntil glucose, which had been added to the mediumwas profoundlyexhausted. This decline was followed by a progressiverecoveryof activity to a normal level with the growth of thecells.Similar behavior of catalase was also seen at low concentrationsof glucose, except that an abrupt rise in activity was observedat the beginning of incubation. Even when cells which had declinedto a minimum of catalase activity were aerated in phosphatebuffer, they continued to synthesize catalase.
  3. 3) The patternof alteration of catalase activity during cellgrowth was accompaniedby a comparable pattern of alterationin respiratory capacity.On the basis of this finding, togetherwith the fact that antimycinA causes intensive depression incatalase formation, it maybe inferred that the formation ofthe respiratory chain conductsthe formation of catalase.
  4. 4) In the presence of ethyl alcoholas the carbon source inplace of glucose, a rise in both catalaseactivity and respiratorycapacity occurred from initiation ofincubation. This fact canbe interpreted to mean that the repressiveeffect of glucoseon catalase formation depends on the aerobiccharacter of thecells.
