Distribution of iron-containing oxidases in aging nodal rootsof rice and wheat was studied. Activities of cytochrome c oxidase(1.9.3.1

, donor:H2O2 oxidoreductase) in wheat roots were comparatively higherthan were those in rice roots at corresponding stages. Cytochromec oxidase in roots remained active throughout the lives of therice and wheat crops. In rice roots, catalase seemed to playa distinct role around the panicle formation stage. Decay ofcatalase activity took place earlier than did that of peroxidaseand cytochrome c oxidase activities. In wheat roots similarenzyme activity changes were not observed. Data may suggestthat the high activity of iron containing oxidases at the panicleformation stage (I) may be chiefly due to catalase activityin rice roots.

Abstract Mitochondria play a central role to provide ATP for fertilization and preimplantation embryo development in the ooplasm. The mitochondrial dysfunction of oocyte has been proposed as one of the causes of high levels of developmental retardation and arrest that occur in preimplantation embryos generated using Assisted Reproductive Technology. Cytoplasmic transfer (CT) from a donor to a recipient oocyte has been applied to infertility due to dysfunctional ooplasm, with resulting pregnancies and births. However, neither the efficacy nor safety of this procedure has been appropriately investigated. In order to improve embryogenesis, we observed the mitochondrial distribution in ooplasma under the several conditions using mitochondrial GFP-transgenic mice (mtGFP-tg mice) in which the mitochondria are visualized by GFP. In this report, we will present our research about the mitochondrial distribution in ooplasm during early embryogenesis and the fate of injected donor mitochondria after CT using mtGFP-tg mice. The mitochondria in ooplasm from the germinal vesicle stage to the morula stage were accumulated in the perinuclear region. The mitochondria of the mtGFP-tg mouse oocyte transferred into the wild type mouse embryo could be observed until the blastocysts stage, suggesting that the mtGFP-tg mice oocyte is very useful for visual observation of the mitochondrial distribution in the oocyte, and that the aberrant early developmental competences due to the oocyte mitochondrial dysfunction may be overcome by transferring the \"normal\" mitochondria. 相似文献

The effect of abscisic acid (ABA) on the tolerance to oxidative stress in a freshwater green alga, Chlamydomonas reinhardtii, was investigated. Exogenously added ABA enhanced the growth of this alga, which was observed under continuous illumination but not in the dark. The cells treated with ABA for 24 h showed tolerance to oxidative stress caused by exposure to paraquat or hydrogen peroxide. In the ABA‐treated cells, the activities of two antioxidant enzymes, catalase (CAT) and ascorbate peroxidase (APX), were significantly higher than those in the untreated control. The result suggests that ABA plays a role in the enhancement of tolerance to oxidative stress by increasing the activity of antioxidant enzymes. 相似文献

This experiment studies the comparative activities of terminaloxidases (Fe-containing oxidases, Cu-containing oxidases andCN-resistant oxidases) in roots of rice and wheat seedlingsand the effect of root pruning on these terminal oxidases inthe roots of those crops. Data indicate that (i) excepting achange in the activity of Cu-containing oxidases no appreciabledifference in respiration or in Fe-containing and CN-resistantoxidases was noticed in the two types of seedling roots and(ii) root pruning did not produce any noticeable change in terminaloxidase activities in roots of wheat seedlings. A drastic changein terminal oxidase activities in roots of rice seedlings wasobserved, suggesting that a close relationship exists betweenCN-resistant oxidase and transplanting time. The effects of EDTA·Mg or GEDTA·Mg on the uptakeof nutrient ions, the release of Ca++ and nucleotides into themedium and the nucleic acid contents in rice and red bean rootswere investigated.
- Both EDTA and EDTA·Mg induced similarly the release ofCa from roots.
- EDTA·Mg as well as EDTA brought abouta significant repressionin K uptake, but had an insignificantor no effect on P, NH4and NO3 uptakes. EDTA·Ca did notrepress K, P, NH4 andNO3 uptakes.
- EDTA or GEDTA acceleratedthe degradation of nucleic acid andthe release of nucleotides,while EDTA·Mg or GEDTA·Mghad no such effect.

An acid α-glucosidase (EC 3.2.1.20) was purified to homogeneity from the culture medium of Tetrahymena thermophila CU 399. Its general molecular, catalytic and immunological properties were compared to those of the T. pyriformis W enzyme. The enzyme from T. thermophila was a 105-kD monomer and the N-terminus (25 amino acid residues) displayed some homology with that of T. pyriformis enzyme. The purified enzyme was most active at 56° C and showed resistance to thermal inactivation. The acid α-glucosidase appears to have α-1,6-glucosidase as well as α-1,4-glucosidase activity. The Km values determined with p-nitrophenyl-α-glucopyranoside, maltose, isomaltose and glycogen were 0.7 mM, 2.5 mM, 28.5 mM and 18.5 mg/ml, respectively. The enzyme was antigenically distinct from T. pyriformis acid α-glucosidase. 相似文献

对日本冲绳岛亚热带天然次生常绿阔叶林的水文生态特性进行了为期3年的定位观测.结果表明,观测期间年平均降雨量高达3325mm,台风降雨(包括直接和间接影响)占年降雨量的42.3%;穿透雨量和树干流量分别为年降雨量的53.9%和30.9%,较高的树干流量可能与高风速、强度降雨以及优势树种的树冠构造紧密相关.林冠截留损失占年降雨量的15.2%,低于世界范围内其它多数常绿阔叶林(15%~30%).年平均地表径流和壤中流量分别是1092mm和613mm,分别占年降雨量的32.8%和18.4%;较高的径流量可能是由不良的土壤物理性质引起的.试验林土壤尤其是下层土壤非毛管孔隙度低(8%~12%)、粘粒含量极高(51.9%~60.5%),水分渗透缓慢(12ml·min-1).0~70 cm土层的有效蓄水量和最大蓄水量分别为85 mm和324mm,林地凋落物层的最大持水量为2.8 mm,接近或低于中国亚热带和热带山地雨林. 相似文献

不同坡位对琉球松人工林凋落物及其养分归还量的影响 总被引:8,自引:0,他引:8
对日本冲绳岛北部不同坡位的琉球松人工林凋落物量及其养分归还量进行了为期3年的观测研究,结果表明,琉球松成林年凋落物量为6.54~8.05Mg·hm^-2·年^-1,其凋落高峰出现在6、7月份.台风干扰对年凋落物量及其季节动态影响显著.凋落物不同组分的养分含量差异明显.年均养分归还总量为113.4~154.6kg·hm^-2·年^-1,其中氮素归还量最大,占42.2%;各养分归还量的大小依次为N>Ca>K>Mg>Na>P.两调查林分由于立地条件的显著差异,年均凋落物量及其相应的养分归还量亦存在明显差异.在立地条件较好的下坡,林分P1的年均凋落物量达8.05Mg·hm^-2·年^-1,高于立地条件相对较差的上坡的林分P2的23.1%;其相应的年养分归还量为:N66、04,P1.63,K17.42,Ca48.31,Mg14.65和N。6、57kg·hm^-2·年^-1,分别高于林分P2的39.7%、48.8%、39.4%、32.9%、24.8%和49.3%.两林分凋落物的养分利用效率分别是N为122和138,P为4934和5945,K为462和523,林分P1明显低于林分P2,这与林分P2的立地条件较差相关.可见,所调查的琉球松林是一种高效的养分利用系统。 相似文献
