Utilization of Nitrogen Sources by Immature Soybean Cotyledons in Culture   总被引:2,自引:0,他引:2

Immature Glycine max (L.) Merrill cotyledons were cultured ina defined medium containing different nitrogen sources. Glutaminewas the most efficient source in terms of protein accumulationin the cotyledons. Asparagine was less efficient (about 70 percent that of glutamine) while allantoin was a poor source ofnitrogen. This was also true for older cotyledons where asparaginaseand allantoinase activities were maximal. The utilization ofboth asparagine and allantoin (but not glutamine) was totallyinhibited by methionine sulfoximine suggesting that their metabolisminvolves ammonia assimilation via glutamine synthetase. Apparently,neither exogenous or endogenously-generated ammonia had mucheffect on glutamine utilization, but ammonia did have a smallinhibitory effect on asparagine, which may in part account forthe lower efficiency observed with this amide. A fluorescent antibody technique was developed for the determination of the capsular-type of strains of Staphylococcus aureus. It compared favourably with the method using serum-soft agar (Yoshida 1972). With the new technique, many populations of encapsulated and unencapsulated strains were investigated. Of 1421 fresh isolates of Staph. aureus , 54 were encapsulated and among these 54·8% and 48·1% were mono- and polyvalent, respectively. Capsular-type antigens A and B were found in 92·5% and 44·4% of strains respectively; capsular-types C and D were found relatively infrequently. In the other group, of unencapsulated strains, capsular-type antigen production was demonstrated in 125 out of 163 strains examined. Mono- and polyvalent capsular-types (A and B antigen producing strains) comprised 77·6% and 22·4%. respectively. In these capsular-types A and B were found in 54·4% and 62·4%, respectively: capsular-type antigen C and D producing strains were again infrequent. These results indicate that a majority of ordinary Staph . aureus strains produce capsular-type antigens although isolation of the encapsulated strains is infrequent.  相似文献

Stand dynamics and self-thinning were analyzed in relation to the dynamics of above-ground biomass in natural Abies sachalinensis stands growing on sand dunes in northern Hokkaido, Japan. This was done in order to examine wave-type regeneration in the stands. Fifty-two plots were established in almost pure Abies stands that ranged from saplings to the mature and collapsing growth stages. Above-ground biomass and tree height reached asymptotic levels prior to the collapsing phase, unlike wave-regeneration Abies stands in central Japan and North America. Stand density was high in the young growth stages, but the self-thinning rate, that is, the density decrease per biomass growth in the study stands was greater than in wave-regeneration stands in central Japan, as indicated by a large self-thinning exponent (–1.26 by reduced major axis regression). The range of tree height distribution was very narrow, and the stands vertical structure was typically single-layered. The slenderness ratio of trees was large, except in young stands. In mature and collapsing stands, advanced seedling density increased markedly. These stand and tree characteristics were considered to be correlated with the wave-type regeneration in the study stands, and it is assumed that prevailing winds affect tree mortality.  相似文献
