DNA binding activity of rabbit antiserum against calf spleen DNA's modified by thiophosphamide (DNA-T) was studied by means of solid enzyme immunoassays (ELISA). The studies demonstrated the preferential binding of the immobilized DNA-T compared to immobilized single-stranded DNA (ss-DNA) and only small preference compared to native DNA. Two antisera against DNA-T were purified by affinity chromatography on a ss-DNA-CNBr agarose from antibodies to calf spleen ss-DNA. They interacted only with the immobilized DNA-T, but not with ss-DNA or native DNA. These results demonstrated that DNA modification by thiophosphamide, decreases the immunogenicity of usual nitrogen-containing DNA bases, but detected new immunogenic specificity for adducts. Detection of new immunogenic specificity in DNA's alkylated by thiophosphamide, resulted in the development of a sensitive enzyme immunoassay for the detection of these adducts in nucleic acids, in monitoring their formation, persistence and repair damages in DNA. 相似文献

Two new types of calcium channels were discovered during research in ionic currents in the somatic membrane ofHelix pomatia neurons, using an intracellular perfusion technique. Apart from the principal calcium current described in the literature with a holding potential of about –110 mV, an additional calcium current was observed activated at depolarizations of –40 to –80 mV and was not reduced when the cell was perfused with solutions containing fluoride anions. The kinetics of this current were well described in the context of the Hodgkin and Huxley model with a time constant of activation of 6–8 msec and of inactivation of 300–600 msec. It increased in amplitude as the Ca++ rose in the cellular environment but was reduced by extracellular addition of the Ca++ antagonists Co++, Ni++, and Cd++, and the organic blockers nifedipine and verapamil. The association constants of these substances with corresponding channels determined from the maximum of the current-voltage relationship were 2 (Ca++), 3 (Co++), 0.06 (nifedipine), and 0.2 mM (verapamil). The properties detected in this component of calcium conductance are compared with those of calcium channels in other excitatory formations and its possible functional role is discussed.A. A. On 202 male rats of Wistar line, a study was carried out of the effect of chronic and acute deprivations of the brain CA-systems activity resulting from administration of 6-OHDA on investigating behaviour and learning. Chronic deprivation of CA-systems activity by neonatal administration of 6-OHDA (100 mg/kg subcutaneously) and their acute deprivation by intracerebral administration of 6-OHDA to adult rats (150 mkg in each lateral ventriculus) was accompanied by similar deep changes of behaviour. Both forms of deprivation reduced the investigating activity of the animals in the open field. In both cases, the above 6-OHDA dozes sharply impeded the learning of animals with emotionally negative reinforcement, with no significant influence on learning with emotionally positive reinforcement. Both forms of deprivation of CA-systems activity weakened the reaction of frustration elicited by a sharp reduction of food reinforcement. 相似文献

A neurotoxin 6-hydroxydopamine (6-OHDA) was introduced to 1, 2 and 3 day old male Wistar rats. Falck-Hillarp histochemical fluorescent method was used to analyze hypothalamic and hypophysial structures containing catecholamines. Statistically reliable decrease in catecholamine fluorescence intensity in hypothalamus and hypophysis, accumulation of catecholamines in nerve cells of supraoptic nucleus and in tanycytes of median eminence differed in 25- and 90-days rats. Disturbance of catecholaminergic systems of hypothalamus and hypophysis induced by neonatal introduction of 6-OHDA leads to reliable relaxation of orienting reaction and deep violation of learning that becomes stronger with age. 相似文献

Resistance of 1280 strains of Salmonella of various serological types isolated in the Zakarpatskaya region within 1967-1976 was studied with respect to benzylpenicillin, streptomycin tetracycline, levomycetin, monomycin, neomycin, erythromycin and oleandomycin. An Increase in the resistance of Salmonella to streptomycin, tetracycline, levomycetin, monomycin and neomycin was shown. During the last years Salmonellae carrying 4-8 resistance determinants were spreading in the region. Among S. typhimurium strains with 7-8 resistance determinants predominated (58.8 per cent). The notion of a dynamic compensatory system is discussed, characterized by the alternation of species occupying the leading position in bumblebee assemblages, while the total density of these pollinators in island ecosystems remains at similar levels. The functioning of the compensatory system is regulated by both abiotic factors (the weather and climate) and biotic factors (competition for trophic resources). Polypeptides with cytostatic activity are known to be present in animal tissues during winter dormancy. A 1-10 kDa polypeptide fraction with cytostatic activity was obtained from brain tissue of hibernating ground squirrels and cold-adapted Yakut horses. The pattern of cytostatic activity of this fraction towards tumor cells is of great interest. We present results testifying to cytostatic activity of this fraction towards the Ehrlich ascitic carcinoma cells. The cytostatic effect is realized in tumor cells at the genetic level. 相似文献
