Summary Cl influx at the luminal border of the epithelium of rabbit gallbladder was measured by 45-sec exposures to36Cl and3H-sucrose (as extracellular marker). Its paracellular component was evaluated by the use of 25mm SCN which immediately and completely inhibits Cl entry into the cell. Cellular influx was equal to 16.7eq cm–2 hr–1 and decreased to 8.5eq cm–2 hr–1 upon removal of HCO 3 from the bathing media and by bubbling 100% O2 for 45 min. When HCO 3 was present, cellular influx was again about halved by the action of 10–4 m acetazolamide, 10–5 to 10–4 m furosemide, 10–5 to 10–4 m 4-acetamido-4-isothiocyanostilbene-2,2-disulfonate (SITS), 10–3 m amiloride. The effects of furosemide and SITS were tested at different concentrations of the inhibitor and with different exposure times: they were maximal at the concentrations reported above and nonadditive. In turn, the effects of amiloride and SITS were not additive. Acetazolamide reached its maximal action after an exposure of about 2 min. When exogenous HCO 3 was absent, the residual cellular influx was insensitive to acetazolamide, furosemide and SITS. When exogenous HCO 3 was present in the salines, Na+ removal from the mucosal side caused a slow decline of cellular Cl influx; conversely, it immediately abolished cellular Cl influx in the absence of HCO 3 . In conclusion, about 50% of cellular influx is sensitive to HCO 3 , inhibitable by SCN, acetazolamide, furosemide, SITS and amiloride and furthermore slowly dependent on Na+. The residual cellular influx is insensitive to bicarbonate, inhibitable by SCN, resistant to acetazolamide, furosemide, SITS and amiloride, and immediately dependent on Na+. Thus, about 50% of apical membrane NaCl influx appears to result from a Na+/H+ and Cl/HCO 3 exchange, whereas the residual influx seems to be due to Na+–Cl contranport on a single carrier. Whether both components are simultaneously present or the latter represents a cellular homeostatic counterreaction to the inhibition of the former is not clear.  相似文献

We used hand-pollination experiments to test the mating systems of and evaluate limits to seed production in two federally listed endangered plants endemic to the Lake Wales Ridge in Florida, USA: Dicerandra frutescens Shinners ssp. frutescens Huck and D. christmanii Huck and Judd (Lamiaceae). Both are nonclonal, short-lived perennials found in gaps created by disturbance (e.g., fire, roads) in Florida scrub. We found that both species require pollen and insect visitation to produce seeds. We detected pollinator limitation of seed production in D. christmanii but not D. frutescens ssp. frutescens, which we suggest is a function of time-since-disturbance or gap size rather than intrinsic differences between the two species. Both species are self-compatible. Inbreeding depression reduced seed set by 60% in D. frutescens ssp. frutescens but did not occur in D. christmanii. We conclude that pollinator limitation (in fire-suppressed populations of both species) and inbreeding depression (in D. frutescens ssp. frutescens) have the potential to limit seed production in these seed-dependent, rare species. Appropriate fire management should mitigate both of these risks, by maintaining large populations and conditions attractive to pollinators. Although these two species are very similar in reproductive biology, comparisons with other Florida scrub endemics and with rare plants in general suggest that potential threats to conservation via reproductive biology are difficult to predict, depending on combinations of ecology, life-history, and phylogenetic history.  相似文献

One of the most reasonable hypotheses regarding the functional significance of the browridge is that the supraorbital torus forms in response to masticatory stress during development. Oyen, Walker, and Rice (1979) have recently proposed a model that tests this hypothesis: if browridges are functionally related to masticatory stresses on the cranial vault, then changes in the biomechanics of the masticatory system ought to be reflected by changes in the browridge. To test their model they attempted to relate biomechanical discontinuities resulting from tooth eruption to episodes of bone deposition on the supraorbital tori of a developmental series of dry Papio crania. This paper reports on a parallel test of the model on a cross-sectional sample of Australian Aboriginal juvenile crania. This sample showed no relation between tooth eruption and the supraorbital surface morphology thought to be indicative of active bone deposition. It is also demonstrated that no significant relationship between tooth eruption and episodes of bone deposition is shown by the Papio sample. It is concluded that the use of small cross-sectional samples of dry crania does not provide a valid test of the model.  相似文献

We have investigated the effect of some metabolic drugs, 3-(3,4-dichlorophenyl)-1,1-dimethylurea (DCMU), 2,4-dinitrophenol (DNP), sodium azide (NaN3), on the photobehavior of single cells of Euglena gracilis, in order to clarify the relevance of different metabolic pathways in the process of photoperception and sensory transduction in this alga. The results obtained show that the photophobic response of Euglena is not affected by the action of these drugs. Summary In the epithelium of rabbit gallbladder, in the nominal absence of bicarbonate, intracellular Cl activity is about 25mm, about 4 times higher than intracellular Cl activity at the electrochemical equilibrium. It is essentially not affected by 10–4 m acetazolamide and 10–4 m 4-acetamido-4-isothiocyanostilbene-2,2-disulfonate (SITS) even during prolonged exposures; it falls to the equilibrium value by removal of Na+ from the lumen without significant changes of the apical membrane potential difference. Both intracellular Cl and Na+ activities are decreased by luminal treatment with 25mm SCN; the initial rates of change are not significantly different. In addition, the initial rates of change of intracellular Cl activity are not significantly different upon Na+ or Cl entry block by the appropriate reduction of the concentration of either ion in the luminal solution. Luminal K+ removal or 10–5 m bumetanide do not affect intracellular Cl and Na+ activities or Cl influx through the apical membrane. It is concluded that in the absence of bicarbonate NaCl entry is entirely due to a Na+–Cl symport on a single carrier which, at least under the conditions tested, does not cotransport K+.  相似文献

Gram-negative bacteria most often use N-acyl homoserine lactones (AHLs) as intercellular quorum-sensing signal molecules. In this study, it was demonstrated that rice plants contain AHL mimic molecules that are very sensitive to the highly specific AiiA lactonase enzyme and can activate three different AHL bacterial biosensors, indicating that the compounds have a homoserine lactone structure and could be AHLs. The possible source and biological significance of this finding are discussed.  相似文献

We have used nitroblue tetrazolium (NBT) as a color reagent to localize antibody-bound alkaline phosphatase in frozen tissue sections. In the method described, NBT is reduced to a stable black diformazan reaction product that contrasts well with nuclear counterstains such as hematoxylin and stands out strongly in black and white photographs. We have found NBT to be a suitable color reagent for the alkaline phosphatase: anti-alkaline immunohistochemical technique. The reaction product also contrasts well with fast red and can therefore be used as second reagent for two color immunoenzyme studies. In this report, we describe a novel two color immunoenzyme method to assess the ex vivo binding of antibodies against Class II histocompatibility antigens in whole organs connected to a perfusion circuit.  相似文献

Summary The protein and gene structure of the Hp Johnson variant (Hp3) were analyzed in two related heterozygous individuals. The molecular weight (23kd) and amino acid composition of Hp3 alpha chain were in agreement with the triplicated structure first suggested by Smithies in 1964. Direct gene analysis by Southern blotting showed a three-fold tandem repeat of the same 1.7 kb DNA segment implicated in the Hp2 gene duplication. On the basis of these data a nine exon model for the Hp3 gene is proposed.  相似文献

A simple and inexpensive aqueous two-phase system for the affinity partitioning of proteins is introduced. An aqueous solution consisting of maltodextrin (M100; molecular mass, 1800) and polyvinylpyrrolidone (PVP360; molecular mass, 360,000) formed two phases at 4 degrees C when the concentration of the polymers was 22.5% (w/w) and 4.0% (w/w), respectively. When the amino derivatives of chlorotriazine textile dyes or other azo textile dyes were added to the two-phase system they partitioned asymmetrically, favoring the upper, less dense, PVP360-rich phase. The association of the textile dyes with PVP360 did not prevent them from acting as affinity ligands for proteins. Three of the dyes screened increased the partition coefficient of purified lysozyme nearly 50-fold over a control containing no dye. Parameters such as pH, ionic strength, and dye concentration modulated the affinity-partitioning effect of the system. The partition coefficient of lysozyme in an egg white protein mixture increased severalfold as the total protein content of the system approached 4% (w/w), indicating that protein concentration is also important in determining the partitioning characteristics of this two-phase system. Proteins were efficiently freed of PVP360 and textile dye by recovery in a high-salt solution when another two-phase system was formed upon the addition of a solution of concentrated potassium phosphate to the isolated upper phase of a PVP360/M100/textile dye two-phase system. The affinity-partitioning system presented here allows one to screen large numbers of potentially useful protein ligands to optimize protein separation, followed by direct scaleup to a system size determined by the user.  相似文献
