Significant genetic variation in leaf photosynthetic rate has been reported in grain sorghum [Sorghum biocolor (L.) Moench]. The relationships between leaf photosynthetic rates and total biomass production and grain yield remain to be established and formed the purpose of this experiment. Twenty two grain sorghum parent lines were tested in the field during the 1988 growing season under well-watered and water-limited conditions. Net carbon assimilation rates were measured at mid-day during the 30 day period from panicle initiation to head exertion on upper-most fully expanded leaves using a portable photosynthesis system (LI-6200). Total biomass and grain production were determined at physiological maturity. The lines exhibited significant genetic variation in leaf photosynthetic rate, total biomass production and grain yield. Significant positive correlations existed between leaf photosynthesis and total biomass and grain production under both well-watered and water-limited conditions. The results suggest that leaf photosynthetic rate measured prior to flowering is a good indicator of productivity in grain sorghum.  相似文献

A large-scale method for the isolation of von Willebrand factor (vWF) from human factor VIII concentrates was developed in order to study the structure of this protein and its platelet binding activity. vWF is composed of a number of glycoprotein subunits that are linked together by disulfide bonds to form a series of multimers. These multimers appear to contain an even number of subunits of 270K. Two minor components of Mr 140K and 120K were also identified, but these chains appear to result from minor proteolysis. The smallest multimer of vWF contained nearly equimolar amounts of the 270K, 140K, and 120K subunits, while the largest multimers contained less than 20% of the two minor components. Amino acid sequence analysis, amino acid composition, and cleavage by cyanogen bromide indicate that the 270K subunits are identical and each is a single polypeptide chain with an amino-terminal sequence of Ser-Leu-Ser-Cys-Arg-Pro-Pro-Met-Val-Lys and a carboxyl-terminal sequence of Glu-Cys-Lys-Cys-Ser-Pro-Arg-Lys-Cys-Ser-Lys. Platelet binding in the presence of ristocetin was 8-fold greater with multimers larger than five (i.e., containing more than 10 subunits of 270K) as compared to multimers less than three (containing less than six subunits of 270K). However, partially reduced vWF (Mr 500K), regardless of whether it was prepared from large or small molecular weight multimers, gave platelet binding similar to that of the smallest multimers. Likewise, partial proteolysis by elastase, thermolysin, trypsin, or chymotrypsin produced small "multimer-like" proteins with platelet binding properties similar to either partially reduced vWF or to the smallest multimers. We conclude that human vWF contains identical 270K subunits assembled into a multivalent structure. Disassembly by either partial reduction or partial proteolysis produces essentially monovalent protein with platelet binding properties similar to that of the smallest multimers. Multivalency is likely the primary factor responsible for the increase in biological activity with multimer size.  相似文献

The objective of this study was to assess the species composition, abundance and habitat association of non-volant small mammals from grassland, Acacia woodland, farmland, riverine forest, bushland and wooded grassland habitats in Gibe Sheleko National Park, southwestern Ethiopia. Data were collected using 49 Sherman live traps in 70 × 70 m sized girds and visual observations from December 2018 to August 2020. A total of 937 small mammals belonging to 10 species were captured and directly identified. The identified small mammal species and their relative abundance include Arvicanthis niloticus (7.04%), Crocidura olivieri (1.92%), Grammomys dolichurus (3.84%), Lemniscomys striatus (10.25%), Mastomys awashensis (24.55%), Mastomys natalensis (33.83%), Mus tenellus (0.85%), Myomys fumatus (6.30%), Rattus rattus (4.70%) and Stenocephalemys albipes (6.72%). Hystrix cristata, Xerus rutilus and Tachyoryctes splendens were documented through observation. The highest number of species (6) was registered at farmland and wooded grassland followed by bushland and riverine forest (5) and Acacia woodland and grassland (4). Lemniscomys striatus, M. awashensis and M. natalensis were common in all habitats. More, 216 (23.05%) individuals were recorded in farmland, while the least, 111 (11.85%) were recorded in Acacia woodland. The study area has various topographical land settings additional study is needed using different traps.  相似文献

The spatial ecology of golden jackal Canis aureus was studied on farmland adjacent to the Bale Mountains National Park in southern Ethiopia during 1998–2000. Three adult and four subadult jackals were captured in leg‐hold traps and radiotagged. The range size of the adult jackals varied from 7.9 to 48.2 km2 and the subadults from 24.2 to 64.8 km2 . These ranges are the largest recorded for this species. Range overlap of the tagged jackals averaged 54%, which, in conjunction with observations of associations between individuals, suggested that all the tagged jackals belonged to one social group. Tagged jackals were observed alone on 87% of occasions despite the extensive overlap in individual ranges. Pairs consisting of a male and female were the most commonly observed group and larger groups were seen on only five occasions. Jackals in this population appeared less gregarious than observed elsewhere. The jackals used all the habitats available to them, particularly at night when they foraged in Artemesia and Hypericum bush and farmland. During the day they were more frequently found in Hagenia and Juniper woodland and their diurnal resting sites were characterized by thick cover. This is the first detailed study of golden jackals in a human‐modified landscape in Africa and further demonstrates the flexibility in behaviour and ecology exhibited by this species throughout its range.  相似文献

Osmotic adjustment, defined as a lowering of osmotic potential (ψπ) due to net solute accumulation in response to water stress, has been considered to be a beneficial drought tolerance mechanism in some crop species. The objective of this experiment was to determine the relative contribution of passive versus active mechanisms involved in diurnal ψπ changes in sorghum (Sorghum bicolor L. Moench) leaf tissue in response to water stress. A single sorghum hybrid (cv AT×623 × RT×430) was grown in the field under variable water supplies. Water potential, ψπ, and relative water content were measured diurnally on expanding and the uppermost fully expanded leaves before flowering and on fully expanded leaves during the grain-filling period. Diurnal changes in total osmotic potential (Δψπ) in response to water stress was 1.1 megapascals before flowering and 1.4 megapascals during grain filling in comparison with 0.53 megapascal under well-watered conditions. Under water-stressed conditions, passive concentration of solutes associated with dehydration accounted for 50% (0.55 megapascal) of the diurnal Δψπ before flowering and 47% (0.66 megapascal) of the change during grain filling. Net solute accumulation accounted for 42% (0.46 megapascal) of the diurnal Δψπ before flowering and 45% (0.63 megapascal) of the change during grain filling in water-stressed leaves. The relative contribution of changes in nonosmotic volume (decreased turgid weight/dry weight) to diurnal Δψπ was less than 8% at either growth stages. Water stress did not affect leaf tissue elasticity or partitioning of water between the symplasm and apoplasm.  相似文献

Studies were conducted on the ecology of potentially pathogenic Vibrio parahaemolyticus in three coastal areas of Kii Channel, Tokushima, Japan. Seawater and seaweed samples were collected seasonally between June 2003 and May 2004. Total and toxigenic strains of V. parahaemolyticus were isolated using most probable number culture and colony blot hybridization. Toxigenic strains were serotyped and further characterized by random amplified polymorphic DNA (RAPD) and ribotyping. Six thousand strains of V. parahaemolyticus were isolated and 18 were found positive for tdh. V. parahaemolyticus were detected in all samples during summer and autumn, and from some samples during winter and spring. Among the toxigenic strains seven serotypes, five ribotypes and RAPD patterns were observed. Seven strains belonged to O3:K6 clone with identical ribotypes and RAPD patterns to that of a pandemic reference strain. The presence of toxigenic V. parahaemolyticus with pandemic potential might indicate a human health risk due to consumption of marine food sources.  相似文献

Habitat fragmentation is an increasing threat to wildlife species across the globe and it has been predicted that future biodiversity will decrease rapidly without the intervention of scientifically-based management. In this study we have applied a landscape genetics approach to suggest a network design that will maintain connectivity among populations of the endangered mountain Nyala (Tragelaphus buxtoni) in the fragmented highlands of Ethiopia. DNA was obtained non-invasively from 328 individuals and genetic population structure and gene flow were estimated using 12 microsatellite markers. In addition, a 475-bp segment of the mitochondrial control region was sequenced for 132 individuals. Potential dispersal corridors were determined from least-cost path analysis based on a habitat suitability map. The genetic data indicated limited gene flow between the sampled mountain Nyala populations of the Bale Massif and the Arsi Massif. The genetic differentiation observed among five sampling areas of the Bale Massif followed a pattern of isolation by distance. We detected no impact of habitat resistance on the gene flow. In the future, however, the current expanding human population in the highlands of Ethiopia may reduce the current mountain Nyala habitat and further limit migration. Hence, maintaining habitat connectivity and facilitating survival of stepping-stone populations will be important for the future conservation of the species. The approach used here may also be useful for the study and conservation of other wildlife species inhabiting areas of increasing human encroachment.  相似文献
