Three East African lichens, Cladonia praetermissa var. modesta comb, nov., Phaeophyscia fumosa, Pyxine nubila are recorded from cool temperate Australia and their distribution and ecology are discussed. The disjunction in distribution is considered unlikely to be due to long-distance dispersal and the species may represent ancient relicts. 相似文献

A wide variety of the barrier crossing strategies exist among migrating songbirds, ranging from strict nocturnal flights to non‐stop flights over a few days. We evaluate barrier crossing strategies in a nocturnally migrating songbird crossing the Mediterranean Sea and the Sahara Desert, the great reed warbler, exploring variation between the sexes and within individuals. We used data from 31 year‐round light‐level geolocators tracks from 26 individuals (13 males and 13 females), with four individuals tracked for 2–3.5 consecutive years. Almost all individuals (25 of 26) prolonged their flights into the day at least on one occasion. The mean duration of these prolonged flights was 19.9 h and did not differ between sexes or seasons. Fifteen birds performed non‐stop flights during more than one full day and night (≥ 24 h; mean = 31.9 h; max = 55 h) in autumn and/or spring, but these flights were generally too short to cross an entire barrier (such as the Sahara Desert) in one non‐stop flight. Patterns of prolonged flights showed considerable within‐individual variation in females between seasons (autumn versus spring) and in both males and females between years, suggesting high individual flexibility in migration strategy. Significantly more males than females performed prolonged flights during autumn migration, but not spring, possibly reflecting sex‐specific carry‐over effects. We conclude that great reed warblers have the ability to conduct prolonged continuous flights for up to several nights and days, which potentially would allow them to cross the Sahara Desert in one non‐stop flight. However, they typically use a mixed strategy of several nocturnal flights with intermittent stopovers in combination with 1–3 prolonged flights. Prolonged flights covered less than half (44%) of the total flight time across the barriers, and the diurnal parts of the flights covered only 18% of this time. 相似文献

We demonstrate a method for simultaneous structure and function determination of integral membrane proteins. Electrical impedance spectroscopy shows that Staphylococcus aureus α-hemolysin channels in membranes tethered to gold have the same properties as those formed in free-standing bilayer lipid membranes. Neutron reflectometry provides high-resolution structural information on the interaction between the channel and the disordered membrane, validating predictions based on the channel's x-ray crystal structure. The robust nature of the membrane enabled the precise localization of the protein within 1.1 Å. The channel's extramembranous cap domain affects the lipid headgroup region and the alkyl chains in the outer membrane leaflet and significantly dehydrates the headgroups. The results suggest that this technique could be used to elucidate molecular details of the association of other proteins with membranes and may provide structural information on domain organization and stimuli-responsive reorganization for transmembrane proteins in membrane mimics. 相似文献

Seed yield is a trait of major interest for the key grassland species Lolium perenne L. An F2 mapping population of perennial ryegrass (VrnA), recently characterised for vernalisation response, was assessed in a glasshouse for traits related to seed yield based on a lattice design with four replications over 2 years. The traits heading date, plant height, length of panicles, number of panicles per plant, seed yield per panicle, flag leaf length, flag leaf width and seed yield per plant revealed repeatabilities ranging from 41 to 76% and a considerable amount of genetic variation in the VrnA population. Path analysis partitioned the direct and indirect effects of seed yield components on seed yield per plant. Seed yield per panicle showed the highest effect on total seed yield. The adjusted mean values of each trait and a genetic linkage map consisting of 97 anonymous and 85 gene associated DNA markers were used for quantitative trait loci (QTL) analysis. Of particular interest were two QTL on linkage group (LG) 1 and LG 2, explaining 41 and 18%, respectively, of the observed phenotypic variation for the trait seed yield per panicle. Both QTL co-located with two major QTL for total seed yield per plant possibly representing the S and Z loci of the gametophytic self incompatibility (SI) system of perennial ryegrass. The diversity of SI alleles in mapping parents and the degree of heterozygosity at SI loci in the full sib progeny determines the interference of self incompatibility with seed production. 相似文献

Recent advances in tracking technology are based on the use of miniature sensors for recording new aspects of individual migratory behaviour. In this study, we have used activity data loggers with barometric and temperature sensors to record the flight altitudes as well as ground elevations during stationary periods of migratory songbirds. We tracked one individual of red‐backed shrike and one great reed warbler along their autumn migration from Europe to Africa. Both individuals performed their migration stepwise in travel segments and climbed most metres during the passage across the Mediterranean Sea and the Sahara Desert and least metres during the first flight segment in Europe. The great reed warbler reached its highest flight altitude of 3950 m a.s.l. during the travel segment from Europe to west Africa, while the red‐backed shrike reached 3650 m a.s.l as maximum flight altitude during its travel segment from Sahel to southern Africa. Both individuals used both lowlands and highlands for resting periods along their migrations. Furthermore, temperature decreased with increasing altitude during migratory flights for both individuals, highlighting the potential to determine flight duration from temperature measurements. Finally, we discuss how barometric data could be used to investigate birds’ responses to changes in air pressure as a cue for departures on migratory flights. This new technique, i.e. using a miniature data logger with barometric pressure sensor to estimate flight altitudes and ground elevations, will open up new avenues for research and importantly advance our understanding on how small birds behave during migratory flights. 相似文献

Hydrodynamic properties as well as structural dynamics of proteins can be investigated by the well-established experimental method of fluorescence anisotropy decay. Successful use of this method depends on determination of the correct kinetic model, the extent of cross-correlation between parameters in the fitting function, and differences between the timescales of the depolarizing motions and the fluorophore's fluorescence lifetime. We have tested the utility of an independently measured steady-state anisotropy value as a constraint during data analysis to reduce parameter cross correlation and to increase the timescales over which anisotropy decay parameters can be recovered accurately for two calcium-binding proteins. Mutant rat F102W parvalbumin was used as a model system because its single tryptophan residue exhibits monoexponential fluorescence intensity and anisotropy decay kinetics. Cod parvalbumin, a protein with a single tryptophan residue that exhibits multiexponential fluorescence decay kinetics, was also examined as a more complex model. Anisotropy decays were measured for both proteins as a function of solution viscosity to vary hydrodynamic parameters. The use of the steady-state anisotropy as a constraint significantly improved the precision and accuracy of recovered parameters for both proteins, particularly for viscosities at which the protein's rotational correlation time was much longer than the fluorescence lifetime. Thus, basic hydrodynamic properties of larger biomolecules can now be determined with more precision and accuracy by fluorescence anisotropy decay. 相似文献

In the presence of Mn2+, an activity in a preparation of purified Bacillus subtilis RecN degrades single-stranded (ss) DNA with a 3′ → 5′ polarity. This activity is not associated with RecN itself, because RecN purified from cells lacking polynucleotide phosphorylase (PNPase) does not show the exonuclease activity. We show here that, in the presence of Mn2+ and low-level inorganic phosphate (Pi), PNPase degrades ssDNA. The limited end-processing of DNA is regulated by ATP and is inactive in the presence of Mg2+ or high-level Pi. In contrast, the RNase activity of PNPase requires Mg2+ and Pi, suggesting that PNPase degradation of RNA and ssDNA occur by mutually exclusive mechanisms. A null pnpA mutation (ΔpnpA) is not epistatic with ΔrecA, but is epistatic with ΔrecN and Δku, which by themselves are non-epistatic. The addA5, ΔrecO, ΔrecQ (ΔrecJ), ΔrecU and ΔrecG mutations (representative of different epistatic groups), in the context of ΔpnpA, demonstrate gain- or loss-of-function by inactivation of repair-by-recombination, depending on acute or chronic exposure to the damaging agent and the nature of the DNA lesion. Our data suggest that PNPase is involved in various nucleic acid metabolic pathways, and its limited ssDNA exonuclease activity plays an important role in RecA-dependent and RecA-independent repair pathways. 相似文献
