There is ample evidence for ecological responses to recent climate change. Most studies to date have concentrated on the effects of climate change on individuals and species, with particular emphasis on the effects on phenology and physiology of organisms as well as changes in the distribution and range shifts of species. However, responses by individual species to climate change are not isolated; they are connected through interactions with others at the same or adjacent trophic levels. Also from this more complex perspective, recent case studies have emphasized evidence on the effects of climate change on biotic interactions and ecosystem services. This review highlights the ‘knowns’ but also ‘unknowns’ resulting from recent climate impact studies and reveals limitations of (linear) extrapolations from recent climate-induced responses of species to expected trends and magnitudes of future climate change. Hence, there is need not only to continue to focus on the impacts of climate change on the actors in ecological networks but also and more intensively to focus on the linkages between them, and to acknowledge that biotic interactions and feedback processes lead to highly complex, nonlinear and sometimes abrupt responses.  相似文献

Background

Elevated cardiac troponin I (cTnI) is frequently observed in patients with severe sepsis and septic shock. However, the mechanisms underlying cTnI release in these patients are still unknown. To date no data regarding coagulation disturbances as a possible mechanism for cTnI release during sepsis are available.

Methodology/Principal Findings

Consecutive patients with systemic inflammatory response syndrome (SIRS), sepsis or septic shock without evidence of an acute coronary syndrome were analyzed. Coagulation parameters (clotting time (CT), clot formation time (CFT), maximum clot firmness (MCF), α-angle) were assessed in native whole blood samples, and using specific activators to evaluate the extrinsic and intrinsic as well as the fibrin component of the coagulation pathway with the use of rotational thrombelastometry (ROTEM).Thirty-eight patients were included and 22 (58%) were cTnI-positive. Baseline characteristics between TnI-positive and -negative patients were similar. The CT, CFT, MCF and the α-angle were similar between the groups with trends towards shorter CT in the extrinsic and fibrin activation.

Conclusions/Significance

We found no differences in coagulation parameters analyzed with rotational thrombelastometry between cTnI-positive and -negative patients with SIRS, severe sepsis, and septic shock. These findings suggest that pathophysiological mechanisms other than thrombus-associated myocardial damage might play a major role, including reversible myocardial membrane leakage and/or cytokine mediated apoptosis in these patients.  相似文献

Physiol.81(5): 1917-1923, 1996.A potential pathogenetic cofactor for thedevelopment of acute mountain sickness and high-altitude pulmonaryedema is an increase in capillary permeability, which could occur as aresult of an inflammatory reaction and/or free radical-mediatedinjury to the lung. We measured the systemic albumin escape byintravenously injecting 5 µCi of 125I-labeled albumin and theplasma concentrations of cytokines, F2-isoprostanes (products of lipidperoxidation), and acute-phase proteins in 24 subjects exposed to 4,559 m. Ten subjects developed acute mountain sickness, and four subjectsdeveloped high-altitude pulmonary edema. The transcapillary escaperate of albumin was 6.9 ± 2.0%/h (SD) at low (550 m) and 6.3 ± 1.9%/h at high (4,559 m) altitude (P = 0.23; n = 24). The subjects withhigh-altitude pulmonary edema had a modest but insignificant increasein the transcapillary escape rate of albumin (4.6 ± 1.9%/h at lowvs. 5.7 ± 1.9%/h at high altitude;P = 0.42;n = 4). Plasma concentrations offibrinogen, 1-acidglycoprotein, C-reactive protein, and interleukin-6 were unchanged inthe early phases and significantly increased by the end of theobservation period in the subjects with high-altitude pulmonary edema,whereas tumor necrosis factor- andF2-isoprostanes did not change atall. This suggests that the inflammatory reaction was rather aconsequence than a causative factor of high-altitude pulmonary edema.In summary, these data argue against a dominant role for increasedsystemic capillary permeability in the development of acute mountainsickness and high-altitude pulmonary edema.

Background and Study Aims

Despite a pronounced reduction of lethality rates due to upper gastrointestinal bleeding, esophageal variceal bleeding remains a challenge for the endoscopist and still accounts for a mortality rate of up to 40% within the first 6 weeks. A relevant proportion of patients with esophageal variceal bleeding remains refractory to standard therapy, thus making a call for additional tools to achieve hemostasis. Self-expandable metal stents (SEMS) incorporate such a tool.

Methods

We evaluated a total number of 582 patients admitted to our endoscopy unit with the diagnosis “gastrointestinal bleeding” according to our documentation software between 2011 and 2014. 82 patients suffered from esophageal variceal bleeding, out of which 11 cases were refractory to standard therapy leading to SEMS application. Patients with esophageal malignancy, fistula, or stricture and a non-esophageal variceal bleeding source were excluded from the analysis. A retrospective analysis reporting a series of clinically relevant parameters in combination with bleeding control rates and adverse events was performed.

Results

The initial bleeding control rate after SEMS application was 100%. Despite this success, we observed a 27% mortality rate within the first 42 days. All of these patients died due to non-directly hemorrhage-associated reasons. The majority of patients exhibited an extensive demand of medical care with prolonged hospital stay. Common complications were hepatic decompensation, pulmonary infection and decline of renal function. Interestingly, we found in 7 out of 11 patients (63.6%) stent dislocation at time of control endoscopy 24 h after hemostasis or at time of stent removal. The presence of hiatal hernia did not affect obviously stent dislocation rates. Refractory patients had significantly longer hospitalization times compared to non-refractory patients.

Conclusions

Self-expandable metal stents for esophageal variceal bleeding seem to be safe and efficient after failed standard therapy. Stent migration appeared to be a common incident that did not lead to reactivation of bleeding in any of our patients. SEMS should be considered a reasonable treatment option for refractory esophageal variceal bleeding after treatment failure of ligature and sclerotherapy and non-availability of or contraindication for other measures (e.g. TIPS).  相似文献

Introduction

Telomere shortening is a cell-intrinsic mechanism that limits cell proliferation by induction of DNA damage responses resulting either in apoptosis or cellular senescence. Shortening of telomeres has been shown to occur during human aging and in chronic diseases that accelerate cell turnover, such as chronic hepatitis. Telomere shortening can limit organ homeostasis and regeneration in response to injury. Whether the same holds true for pancreas regeneration in response to injury is not known.

Methods

In the present study, pancreatic regeneration after acute cerulein-induced pancreatitis was studied in late generation telomerase knockout mice with short telomeres compared to telomerase wild-type mice with long telomeres.

Results

Late generation telomerase knockout mice exhibited impaired exocrine pancreatic regeneration after acute pancreatitis as seen by persistence of metaplastic acinar cells and markedly reduced proliferation. The expression levels of p53 and p21 were not significantly increased in regenerating pancreas of late generation telomerase knockout mice compared to wild-type mice.

Conclusion

Our results indicate that pancreatic regeneration is limited in the context of telomere dysfunction without evidence for p53 checkpoint activation.  相似文献

Neurogenesis in the adult human brain occurs within two principle neurogenic regions, the hippocampus and the subventricular zone (SVZ) of the lateral ventricles. Recent reports demonstrated the isolation of human neuroprogenitor cells (NPCs) from these regions, but due to limited tissue availability the knowledge of their phenotype and differentiation behavior is restricted. Here we characterize the phenotype and differentiation capacity of human adult hippocampal NPCs (hNPCs), derived from patients who underwent epilepsy surgery, on various feeder cells including fetal mixed cortical cultures, mouse embryonic fibroblasts (MEFs) and PA6 stromal cells. Isolated hNPCs were cultured in clonal density by transferring the cells to serum-free media supplemented with FGF-2 and EGF in 3% atmospheric oxygen. These hNPCs showed neurosphere formation, expressed high levels of early neuroectodermal markers, such as the proneural genes NeuroD1 and Olig2, the NSC markers Nestin and Musashi1, the proliferation marker Ki67 and significant activity of telomerase. The phenotype was CD15low/-, CD34-, CD45- and CD133-. After removal of mitogens and plating them on poly D-lysine, they spontaneously differentiated into a neuronal (MAP2ab+), astroglial (GFAP+), and oligodendroglial (GalC+) phenotype. Differentiated hNPCs showed functional properties of neurons, such as sodium channels, action potentials and production of the neurotransmitters glutamate and GABA. Co-culture of hNPCs with fetal cortical cultures, MEFs and PA6 cells increased neurogenesis of hNPCs in vitro, while only MEFs and PA6 cells also led to a morphological and functional neurogenic maturation. Together we provide a first detailed characterization of the phenotype and differentiation potential of human adult hNPCs in vitro. Our findings reinforce the emerging view that the differentiation capacity of adult hNPCs is critically influenced by non-neuronal mesodermal feeder cells.  相似文献

Recently, there has been increasing evidence of species' range shifts due to changes in climate. Whereas most of these shifts relate ground truth biogeographic data to a general warming trend in regional or global climate data, we here present a reanalysis of both biogeographic and bioclimatic data of equal spatio-temporal resolution, covering a time span of more than 50 years. Our results reveal a coherent and synchronous shift in both species' distribution and climate. They show not only a shift in the northern margin of a species, which is in concert with gradually increasing winter temperatures in the area, they also confirm the simulated species' distribution changes expected from a bioclimatic model under the recent, relatively moderate climate change.  相似文献

Aim  Many species are currently expanding their ranges in response to climate change, but the mechanisms underlying these range expansions are in many cases poorly understood. In this paper we explore potential climatic factors governing the recent establishment of new palm populations far to the north of any other viable palm population in the world.
Location  Southern Switzerland, Europe, Asia and the world.
Methods  We identified ecological threshold values for the target species, Trachycarpus fortunei , based on gridded climate data, altitude and distributional records from the native range and applied them to the introduced range using local field monitoring and measured meteorological data as well as a bioclimatic model.
Results  We identified a strong relationship between minimum winter temperatures, influenced by growing season length and the distribution of the palm in its native range. Recent climate change strongly coincides with the palm's recent spread into southern Switzerland, which is in concert with the expansion of the global range of palms across various continents.
Main conclusions  Our results strongly suggest that the expansion of palms into (semi-)natural forests is driven by changes in winter temperature and growing season length and not by delayed population expansion. This implies that this rapid expansion is likely to continue in the future under a warming climate. Palms in general, and T. fortunei in particular, are significant bioindicators across continents for present-day climate change and reflect a global signal towards warmer conditions.  相似文献
