To understand the role of TGF-β signaling in cardiovascular development, we generated mice with conditional deletion of the TGF-β type II receptor (TβRII) gene (Tgfbr2) in cells expressing the smooth muscle cell-specific protein SM22α. The SM22α promoter was active in tissues involved in cardiovascular development: vascular smooth muscle cells (VSMCs), epicardium and myocardium. All SM22-Cre+/−/Tgfbr2 flox/flox embryos died during the last third of gestation. About half the mutant embryos exhibited heart defects (ventricular myocardium hypoplasia and septal defects). All mutant embryos displayed profound vascular abnormalities in the descending thoracic aorta (irregular outline and thickness, occasional aneurysms and elastic fiber disarray). Restriction of these defects to the descending thoracic aorta occurred despite similar levels of Tgfbr2 invalidation in the other portions of the aorta, the ductus arteriosus and the pulmonary trunk. Immunocytochemistry identified impairment of VSMC differentiation in the coronary vessels and the descending thoracic aorta as crucial for the defects. Ventricular myocardial hypoplasia, when present, was associated to impaired α-SMA differentiation of the epicardium-derived coronary VSMCs. Tgfbr2 deletion in the VSMCs of the descending thoracic aorta diminished the number of α-SMA-positive VSMC progenitors in the media at E11.5 and drastically decreased tropoelastin (from E11.5) and fibulin-5 (from E.12.5) synthesis and/or deposition. Defective elastogenesis observed in all mutant embryos and the resulting dilatation and probable rupture of the descending thoracic aorta might explain the late embryonic lethality. To conclude, during mouse development, TGF-β plays an irreplaceable role on the differentiation of the VSMCs in the coronary vessels and the descending thoracic aorta.  相似文献

Para-aminoclonidine coupled to hemocyanin was used to produce mouse monoclonal antibodies directed against clonidine. The properties of one of these, called mFE7, secreted by a clone of hybrid myeloma, are described. This antibody displayed total crossreactivity with imidazolidines and no crossreactivity at all with catecholamines or other known naturally occurring substances tested. A liquid phase radioimmunoassay permitted the detection of immunoreactivity in human brain extracts. The mFE7 antibody could be useful for immunopurifying the endogenous ligand of Imidazolines Preferring Receptors (IPR) which are catecholamines insensitive.  相似文献

TGR(ASrAOGEN)680, a newly developed transgenic rat line with specific downregulation of astroglial synthesis of angiotensinogen, exhibits decreased brain angiotensinogen content associated with a mild diabetes insipidus and lower blood pressure. Autoradiographic experiments were performed on TGR(ASrAOGEN) (TG) and Sprague-Dawley (SD) control rats to quantify AT(1) and AT(2) receptor-binding sites in different brain nuclei and circumventricular organs. Dose-response curves for drinking response to intracerebroventricular injections of ANG II were compared between SD and TG rats. In most of the regions inside the blood-brain barrier [paraventricular nucleus (PVN), piriform cortex, lateral olfactory tract (LOT), and lateral preoptic area (LPO)], AT(1) receptor binding (sensitive to CV-11974) was significantly higher in TG compared with SD. In contrast, in the circumventricular organs investigated [subfornical organ (SFO) and area postrema], AT(1) receptor binding was significantly lower in TG. AT(2) receptors (binding sensitive to PD-123319) were detected at similar levels in the inferior olive (IO) of both strains. Angiotensin-binding sites sensitive to both CV-11974 and PD-123319 were detected in the LPO of SD rats and specifically upregulated in LOT, IO, and most notably PVN and SFO of TG. The dose-response curve for water intake after intracerebroventricular injections showed a higher sensitivity to ANG II of TG (EC(50) = 3.1 ng) compared with SD (EC(50) = 11.2 ng), strongly suggesting that the upregulation of AT(1) receptors inside the blood-brain barrier of TG rats is functional. Finally, we showed that downregulation of angiotensinogen synthesized by astroglial cells differentially regulates angiotensin receptor subtypes inside the brain and in circumventricular organs.  相似文献

The aim of our present study is to investigate the interaction between angiotensin II (ANG II) and sympathetic nervous system (SNS) on matrix metalloproteinase MMP-2 and MMP-9 expression and activity in juvenile rat aorta under normal conditions. Sympathectomy with guanethidine and blockade of the ANG II receptors (AT1R) by losartan were performed alone or in combination on new-born rats. mRNA, protein expression and activity of MMP-2 and MMP-9 were examined by Q-RT-PCR, immunoblotting and zymography, respectively. MMP-2 mRNA and protein amount were decreased after sympathectomy or AT1R blockade and an additive effect was observed after combined treatment. However, MMP-9 expression was reduced to the same level in the three treated groups. There were some detectable gelatinolytic activity of the MMPs in both control and treated rats. We concluded that ANG II stimulates directly and indirectly (via sympathostimulator pathway) the MMP-2 expression but seems unable to affect MMP-9 expression through direct pathway. Combined inhibition of SNS and ANG II were more efficient than a single inhibition in reducing MMP amounts in rat vessels.  相似文献

Climate change models predict a strong reduction of average precipitation, especially of the summer rainfall, and an increase in intensity and frequency of drought events in the Mediterranean region. The research aim was to understand how four dominant grass species (Arrhenatherum elatius, Cynosurus cristatus, Elymus repens, and Lolium perenne) in sub-Mediterranean meadows (central Apennines, Italy) modulate their resource acquisition and conservation strategies to short-term variation of the pattern of summer water supply. During summer 2016, using a randomized block design, we tested the effect of three patterns of summer water supply, differing in water amount and watering frequency, on leaf area, leaf dry mass, specific leaf area (SLA), leaf senescence, and plant height. Our results showed that dominant grass species can modulate their strategies to variation of the pattern of summer water supply, but the response of leaf traits and plant height is mediated by the set of functional characteristics of the species. E. repens and A. elatius, with summer green leaves, lower SLA, later flowering period, and deeper roots, were less influenced by changes in water amount. C. cristatus and L. perenne, which display acquisitive strategies (persistent leaves, higher SLA values), earlier flowering, and shallower roots were more influenced by changes in the pattern of summer water supply. Our results suggest that a short-term decrease in water availability might affect primarily species with trait syndromes less adapted to face summer drought.

Plant functional traits are now frequently used instead of species identity to identify how plant species co-exist in assemblages. One notion is that species inhabiting the same environment have more characteristics in common than species from different habitats, leading to different prevailing dominant traits along environmental gradients, and also to a lesser diversity of traits in habitats that impose a stronger filter on these traits. Though such patterns have been demonstrated for different environmental drivers and different traits, studies using easily available traits connected to above ground processes (i.e. traits of the leaf-height-seed, or LHS, strategy scheme) are largely overrepresented in these analyses. Here we combined data on clonal and bud bank traits, representing the ability to reproduce and spread vegetatively, with LHS trait data and examined how these traits varied in relation to the vegetational composition of 29 Central-European habitat types. Our analysis focused on determining whether clonal/bud bank or LHS traits play an important role for environmental filtering along gradients approximated by Ellenberg indicator values (EIV) across these habitats. Our results show that clonal and bud bank traits are at least as – if not more – important for the differentiation of the 29 habitat types. Overall, diversity and dominance of clonal and bud bank traits was more strongly correlated with gradients of light availability, temperature, moisture, soil reaction, and nutrient availability across these habitats than it was the case for traits of the leaf-height-seed scheme. Our results call for a stronger integration of belowground traits into the functional traits approach in plant ecology and for an extension of efforts to collect such data.  相似文献

Patients having the nephrogenic syndrome of inappropriate antidiuresis present either the R137C or R137L V2 mutated receptor. While the clinical features have been characterized, the molecular mechanisms of functioning of these two mutants remain elusive. In the present study, we compare the pharmacological properties of R137C and R137L mutants with the wild-type and the V2 D136A receptor, the latter being reported as a highly constitutively active receptor. We have performed binding studies, second messenger measurements and BRET experiments in order to evaluate the affinities of the ligands, their agonist and antagonist properties and the ability of the receptors to recruit β-arrestins, respectively. The R137C and R137L receptors exhibit small constitutive activities regarding the Gs protein activation. In addition, these two mutants induce a constitutive β-arrestin recruitment. Of interest, they also exhibit weak sensitivities to agonist and to inverse agonist in term of Gs protein coupling and β-arrestin recruitment. The small constitutive activities of the mutants and the weak regulation of their functioning by agonist suggest a poor ability of the antidiuretic function to be adapted to the external stimuli, giving to the environmental factors an importance which can explain some of the phenotypic variability in patients having NSIAD.  相似文献
