Fruits of Pongamia pinnata afforded four new furanoflavonoids, pongapinnol A-D (1-4), and a new coumestan, pongacoumestan (5) along with thirteen known compounds 6-18. Compounds 16 and 17 are isolated for the first time from this plant. The structures of isolated compounds were elucidated on the basis of spectroscopic data interpretation.  相似文献

Nineteen compounds of various classes, such as flavonoid glycosides, pterocarpanoids, lipids, glycolipids, and alkaloids, were isolated and identified from the Desmodium gangeticum whole plant. Aminoglucosyl glycerolipid (8) is reported here for the first time. Its structure has been elucidated by spectroscopic and degradation studies. This novel compound exhibited in vitro antileishmanial and immunomodulatory activities, as it enhanced nitric oxide (NO) production and provided resistance against infection established in peritoneal macrophages by the protozoan parasite Leishmania donovani. Another known compound, glycosphingolipid (cerebroside) (7) was found to possess significant in vitro antileishmanial and immunomodulatory activities against the same parasite. This study focuses on the identification and quantification of airborne pollen grains from allergenic plant species and their relationship with meteorological factors, i.e. maximum and minimum daily temperature, relative humidity, rainfall and wind speed in the city of Islamabad, Pakistan. An aerobiological data set (2010–2012), collected using rotorod samplers in five different sectors of the city, was supplied by the Pakistan Meteorological Department. Pollen of eight allergenic species was identified amongst which Broussonetia papyrifera exceeded the highest pollen level and, therefore, likely played a key role in aggravating the symptoms of pollen allergy in the city. The mean weekly pollen counts were next correlated with the weekly number of allergic patients visiting hospitals during 2010–2011. Clinical data were acquired from the Pakistan Institute of Medical Sciences. The highest number of allergic patients visiting hospital was usually observed during weeks with high pollen level. These results suggest a close relationship between the pollen concentration in the air and the allergy symptoms. Spearman’s rank correlation analysis was performed to establish the relationships between meteorological parameters and daily average pollen counts. A pollen calendar for the Islamabad city was also prepared to provide a guide for the timing and duration of season for all encountered pollen types.  相似文献

Honey is a complex foodstuff found in nature which is used without any processing. Honey has been in use in medicine as well as raw food since ancient times. Essentially, it is a blend of sugars especially fructose and glucose. The objectives of the study were to determine major sugar composition as well as pesticides contamination in honey samples. Further, Hydroxy-methyl-furfuraldehyde (HMF) level was also determined to ascertain the freshness of honey samples. A total of 14 samples were collected from local market and tested for fructose, glucose, sucrose, HMF and organochlorine pesticides using HPLC and GC–MS techniques respectively. The total sugars in the 14 honey samples were found ranging between 50.26 and 74.74 g/100 g of honey. The chromatographic results showed the presence of the sugars like fructose and glucose in all honey samples. The honey sample SH–11 was found to contain the highest amount of fructose (40.63%). On the other hand, the lowest amount of fructose with 29.08% was observed in SH–7. The HPLC analysis also revealed the presence of sucrose in two samples but under the permissible limit. The average ratio of fructose to glucose in these honey samples was 1.3. None of the sample has ratio below 1.0 indicating lesser chances for honey to crystallize on storage. Out of 14 honey samples, 13 samples were found negative for the presence of any of the 63 pesticides tested. Only sample No. 13, was found to contain 15.95 ppb hexachlorobenzene per kilogram of honey. The HMF was not detected in four samples but in remaining samples it was well below the maximum permissible limit. No pesticide and sugar adulteration was observed in any of the honey samples. The honeys collected from Saudi Arabian markets were found to confirm the standards set by the regional and international standardization organization, the GSO and Codex Alimentarius Commission respectively.  相似文献

Introduction

We previously reported that alveolar macrophages from patients with chronic obstructive pulmonary disease (COPD) are defective in their ability to phagocytose apoptotic cells, with a similar defect in response to cigarette smoke. The exact mechanisms for this defect are unknown. Sphingolipids including ceramide, sphingosine and sphingosine-1-phosphate (S1P) are involved in diverse cellular processes and we hypothesised that a comprehensive analysis of this system in alveolar macrophages in COPD may help to delineate the reasons for defective phagocytic function.

Methods

We compared mRNA expression of sphingosine kinases (SPHK1/2), S1P receptors (S1PR1-5) and S1P-degrading enzymes (SGPP1, SGPP2, SGPL1) in bronchoalveolar lavage-derived alveolar macrophages from 10 healthy controls, 7 healthy smokers and 20 COPD patients (10 current- and 10 ex-smokers) using Real-Time PCR. Phagocytosis of apoptotic cells was investigated using flow cytometry. Functional associations were assessed between sphingosine signalling system components and alveolar macrophage phagocytic ability in COPD. To elucidate functional effects of increased S1PR5 on macrophage phagocytic ability, we performed the phagocytosis assay in the presence of varying concentrations of suramin, an antagonist of S1PR3 and S1PR5. The effects of cigarette smoking on the S1P system were investigated using a THP-1 macrophage cell line model.

Results

We found significant increases in SPHK1/2 (3.4- and 2.1-fold increases respectively), S1PR2 and 5 (4.3- and 14.6-fold increases respectively), and SGPL1 (4.5-fold increase) in COPD vs. controls. S1PR5 and SGPL1 expression was unaffected by smoking status, suggesting a COPD “disease effect” rather than smoke effect per se. Significant associations were noted between S1PR5 and both lung function and phagocytosis. Cigarette smoke extract significantly increased mRNA expression of SPHK1, SPHK2, S1PR2 and S1PR5 by THP-1 macrophages, confirming the results in patient-derived macrophages. Antagonising SIPR5 significantly improved phagocytosis.

Conclusion

Our results suggest a potential link between the S1P signalling system and defective macrophage phagocytic function in COPD and advise therapeutic targets.  相似文献

Abstract

To assess the tolerance, the rye-grass L. grown on soil amended with petroleum wastewater (PWW) containing four metals lead, zinc, nickel and mercury. The PWW (25 to 50%) showed remarkable increase in length and biomass. Chlorophyll ‘a and b’ increased with an increase of PWW from 25–50% while such contents decreased on increasing the 75–100% compared to control. The mass balance performed on the system showed the removal of 90–97.6% lead, 85.5–92.9% zinc, 78.9–85.5% nickle and 47.6–27.5% mercury. The model for the maximum metal reduction rate (Rmax) was much better for Pb (89.5) and Zn (72.1) with respect to Ni (57.3) and Hg (32.4). Survival of rye-grass (30-days, statics, and renewal exposures) was increased by 50% as compared to control. The toxicity index Y of PWW showed 0–25% deficiency level, 25–50% tolerance level, 50–90% toxic level and 90–100% lethal level.
