Effect of aerosolization on subsequent bacterial survival.   总被引:7,自引:6,他引:1       下载免费PDF全文
To determine whether aerosolization could impair bacterial survival, Pseudomonas syringae and Erwinia herbicola were aerosolized in a greenhouse, the aerosol was sampled at various distances from the site of release by using all-glass impingers, and bacterial survival was followed in the impingers for 6 h. Bacterial survival subsequent to aerosolization of P. syringae and E. herbicola was not impaired 1 m from the site of release. P. syringae aerosolized at 3 to 15 m from the site of release at a temperature of 12 degrees C and a relative humidity of 80% survived 35- to 65-fold better than P. syringae released at 27 degrees C and a relative humidity of 40%. No difference was observed in the survival of P. syringae and E. herbicola following aerosolization at the same temperature and relative humidity. Bacteria sprayed directly onto bean and oat plants established stable populations at comparable numbers on both plants over an 8-day period following inoculation. Bacteria that inoculated adjacent plants by drifting downwind up to 5 m were detectable at an initial population of 10(2) CFU/g on oats and 10(5) CFU/g on beans 2 h after the spray. However, bacterial populations on both plants were undetectable within 48 h.  相似文献

1. Canopy invertebrate responses to Hurricane Hugo, tree species, and recovery time were examined at the Luquillo Experimental Forest in Puerto Rico during 1991–92 and 1994–95. Six tree species representing early and late successional stages were examined in paired plots representing severe hurricane disturbance (most trees toppled) and light hurricane disturbance (all trees standing and most branches intact). 2. Hurricane disturbance affected invertebrate abundances significantly. Sap-suckers and molluscs were more abundant, and defoliators, detritivores, and emergent aquatic insects were less abundant in recovering tree-fall gaps than in intact forest during this 5-year period. These changes in functional organisation are consistent with comparable studies of arthropod responses to canopy removal during harvest in temperate forests. 3. Tree species also affected invertebrate abundances significantly, but invertebrate communities did not differ significantly between the three early successional and three later successional tree species. 4. Most taxa showed significant annual variation in abundances, but only two Homoptera species showed a significant linear decline in abundance through time, perhaps reflecting long-term trends during recovery. 5. Leaf area missing, an indicator of herbivore effect on canopy processes, showed significant seasonal and annual trends, as well as differences among tree species and hurricane treatments. Generally, leaf area missing peaked during the wet season each year, but reached its highest levels during an extended drought in 1994. Leaf area missing also tended to be higher on the more abundant tree species in each disturbance treatment. 6. Herbivore abundances and leaf area missing were not related to concentrations of nitrogen, phosphorous, potassium, or calcium in the foliage. 7. This study demonstrated that invertebrate community structure and herbivory are dynamic processes that reflect the influences of host species and variable environmental conditions.  相似文献

Despite frequent reporting of blood pressure (BP) during profound passive heat stress, both with and without a hypotensive challenge, the method by which BP is measured often varies between laboratories. It is unknown whether auscultatory and finger BP measures accurately reflect intra-arterial BP during dynamic changes in cardiac output and peripheral resistance associated with the aforementioned conditions. The purpose of this investigation was to test the hypothesis that auscultatory BP measured at the brachial artery, and finger BP measured by the Penaz method, are valid measures of intra-arterial BP during a passive heat stress and a heat-stressed orthostatic challenge, via lower body negative pressure (LBNP). Absolute (specific aim 1) and the change in (specific aim 2) systolic (SBP), diastolic (DBP), and mean BPs (MBP) were compared at normothermia, after a core temperature increase of 1.47 ± 0.09°C, and during subsequent LBNP. Heat stress did not change auscultatory SBP (6 ± 11 mmHg; P = 0.16), but Penaz SBP (-22 ± 16 mmHg; P < 0.001) and intra-arterial SBP (-11 ± 13 mmHg P = 0.017) decreased. In contrast, DBP and MBP did not differ between methods throughout heat stress. Compared with BP before LBNP, the magnitude of the reduction in BP with all three methods was similar throughout LBNP (P > 0.05). In conclusion, auscultatory SBP and Penaz SBP failed to track the decrease in intra-arterial SBP that occurred during the profound heat stress, while decreases in arterial BP during an orthostatic challenge are comparable between methodologies.  相似文献

The extent to which heat stress compromises blood pressure control is variable among individuals, with some individuals becoming very intolerant to a hypotensive challenge, such as lower body negative pressure (LBNP) while heat stressed, while others are relatively tolerant. Heat stress itself reduces indexes of ventricular filling pressure, including central venous pressure, which may be reflective of reductions in tolerance in this thermal condition. This study tested the hypothesis that the magnitude of the reduction in central venous pressure in response to heat stress alone is related to the subsequent decrement in LBNP tolerance. In 19 subjects, central hypovolemia was imposed via LBNP to presyncope in both normothermic and heat-stress conditions. Tolerance to LBNP was quantified using a cumulative stress index (CSI), and the difference between normothermic CSI and heat-stress CSI was calculated for each individual. The eight individuals with the greatest CSI difference between normothermic and heat-stress tolerances (LargeDif), and the eight individuals with the smallest CSI difference (SmallDif), were grouped together. By design, the difference in CSI between thermal conditions was greater in the LargeDif group (969 vs. 382 mmHg × min; P < 0.001). Despite this profound difference in the effect of heat stress in decreasing LBNP tolerance between groups, coupled with no difference in the rise in core body temperatures to the heat stress (LargeDif, 1.4 ± 0.1°C vs. SmallDif, 1.4 ± 0.1°C; interaction P = 0.89), the reduction in central venous pressure during heat stress alone was similar between groups (LargeDif: 5.7 ± 1.9 mmHg vs. SmallDif: 5.2 ± 2.0 mmHg; interaction P = 0.85). Contrary to the proposed hypothesis, differences in blood pressure control during LBNP are not related to differences in the magnitude of the heat-stress-induced reductions in central venous pressure.  相似文献

Catalase incorporation into enumeration media caused a significant increase (greater than 63%) in the colony-forming abilities of airborne bacteria. Incubation for 30 to 60 min of airborne bacteria in collection fluid containing catalase caused a greater than 95% increase in colony-forming ability. However, catalase did not have any effects on enumeration at high relative humidities (80 to 90%).  相似文献

Because the death mechanisms of freeze-dried and air-dried bacteria are thought to be similar, freeze-drying was used to investigate the survival differences between potentially airborne genetically engineered microorganisms and their wild types. To this end, engineered strains of Escherichia coli and Pseudomonas syringae were freeze-dried and exposed to air, visible light, or both. The death rates of all engineered strains were significantly higher than those of their parental strains. Light and air exposure were found to increase the death rates of all strains. Application of death rate models to freeze-dried engineered bacteria to be released into the environment is discussed.  相似文献

Summary A bioaerosol research chamber was constructed and used to evaluate wind-gust release of previously depositedPseudomonas syringae, spores ofBacillus subtilis var.niger, and fluorescent microspheres (FM) on oat plants, and the airborne survival of the releasedP. syringae. Observations of wind gusts on the releasedBacillus spores and FM showed they had similar particle size distributions and therefore FM could act as bacterial sized surrogates in particle dispersion. Microscopic examination of the released FM containing particles revealed that 84% were associated with either fungal spores and hyphae were epiphytic on the plant, or with plant and soil debris. The release rate ofBacillus spores decreased as the number of gusts experienced by the plants increased, with a greater proportion of larger particles removed. The larger the particle associated with the releasedP. syringae, the longer the bacteria survived.  相似文献

Decontamination treatments of burning and biocide application, alone and in combination with tillage, were evaluated for their ability to reduce populations of bacteria applied to the leaves of plants in field plots. In addition, the effects of these control methods on indigenous leaf and soil bacteria and fungi were assessed. Field plots of bush beans (Phaseolus vulgaris), sprayed with the bacterium Pseudomonas syringae, Pseudomonas fluorescens, or Erwinia herbicola, received the following treatments: (i) control, (ii) tillage, (iii) burning, (iv) burning plus tillage (burn-tillage), (v) Kocide (cupric hydroxide), (vi) Kocide plus tillage, (vii) Agri-Strep (streptomycin sulfate), and (viii) Agri-Strep plus tillage. Leaves and soil from the plots were sampled at 1 day before and at 1, 3, 7, 10, 14, 21, and 30 days after application of the decontamination treatments. The burn and burn-tillage treatments produced the most significant reductions in bacterial populations. The Agri-Strep treatment was more effective than the Kocide treatment in eliminating applied bacteria, but neither biocide produced consistent or persistent control. In contrast, the tillage treatment, alone or in combination with the Agri-Strep or Kocide treatments, had a short-term stimulatory effect and increased populations of applied bacteria and also levels of indigenous fungi and bacteria. Agri-Strep and Kocide treatments caused significant reductions in indigenous bacterial populations up to 14 days after application and in indigenous fungal populations on day 7 after application. Our results suggest that conventional plant disease control methods may not provide satisfactory control of genetically engineered microorganisms and indicate a need for further development of effective and selective methods to control release microorganisms at field sites.  相似文献
