The cortex cells of pea roots (Pisum sativum L.) grown for 144 h in the presence of cadmium, chromium and lead at the concentration 10(-4) M were the object of the present studies. Applied metals reduced dry mass content and concentration of nuclei in meristematic and differentiation zones. Chromium only enhanced nuclear mass concentration in the differentiation zone. The metals also made dry mass content and concentration of cytoplasm reduce, but they diminished mostly the concentration of cytoplasm in the meristematic zone and its dry mass in differentiation zone. Stimulation of cytoplasmic dry mass concentration was visible in the 1st mm (Cr) and 7th mm (Cd). Moreover, chromium caused a marked increase of cytoplasmic dry mass content in the 3rd mm of root. The studied metals reduced nuclear size, calculated as surface area, in the meristematic and differentiation zones. The increment of nuclear dimensions was observed only in the 1-3rd mm (Pb), 3rd (Cr) and 7th mm (Cd). In the presence of the applied metals the surface area of cytoplasm increased only in the 3rd mm and in 5th mm (chromium only). The present observations have shown that the toxicity of studied metals is as follows; Cd greater than Pb greater than Cr (nucleus - dry mass content and concentration, cytoplasmic area), Pb greater than Cd greater than Cr (cytoplasmic dry mass content and concentration and Cd greater than Cr greater than Pb (nuclear dimensions).  相似文献

Résumé Les granulations qui constituent les structures les plus primitives de la plaque cellulaire contiennent des enzymes hydrolytiques caractéristiques pour les sphérosomes de la cellule végétale; la plupart de ces enzymes sont présentes dans les lysosomes de la cellule animale. Dans les granulations de la plaque cellulaire nous avons constaté l'activité des enzymes suivantes: estérases non spécifiques, lipases, phosphatases acide et alcaline, aryl-sulfatase, désoxyribonucléase acide et probablement-glucuronidase. Le caractère chimique et morphologique de ces granulations, ainsi que leur équipement enzymatique correspondent aux sphérosomes.Dans la littérature les images de la cytocinèse au microscope électronique étant divergentes, le problème de l'origine et de la nature des granulations en question ne peut pas être pour le moment totalement résolu.
Zusammenfassung Die Körnchen, die die ursprünglichen Strukturen der Zellplatte bilden, enthalten die hydrolytischen Enzyme, die für die Sphärosomen der pflanzlichen Zelle und meistens für die Lysosomen der tierischen Zelle charakteristisch sind. Es sind die folgenden Enzyme: Esterasen (nicht spezifisch), Lipasen, saure Phosphatase, Aryl-sulfatase, saure Desoxyribonuklease und-Glukuronidase. Außerdem enthalten diese Körnchen die alkalische Phosphatase. Der morphologische und chemische Charakter dieser Körnchen und ihr enzymatischer Inhalt entsprechen den Sphärosomen.Wegen großer Unterschiede der in der Literatur vorhandenen Beschreibungen der Zellplatte in elektronenmikroskopischen Bildern kann das Problem der Entstehung und der Natur dieser Körnchen zur Zeit nicht völlig entschieden werden.

Summary It was shown that Golgi structures abundantly appearing in tapetal cells ofDelphinium Ajacis L. developing anthers, prior to meiocytes meiosis, show a fine fibrous material within their vesicles. At the time of the formation of tapetal cell wall this fibrous component, released by an exocytotic process, is incorporated into the cell wall. The membrane of dictyosomes derived vesicles participates in the development of plasma membrane. Fibrous material appears to be morphologically similar to the fibrils of tapetal cell wall; this cell wall gives a positive reaction for cellulose and pectins, as visible in the light microscope. Moreover, the fibrous and pectinase resistant compound of dictyosomes derived vesicles and the fibrils of cell wall disappear partly after cellulase digestion which proves their cellulosic character. On the other hand pectinase treatment as well as ruthenium red staining suggest associated with cellulose pectins within Golgi vesicles.  相似文献

We report the use of polyamidoamine (PAMAM-NH2) dendrimers along with other non-viral vehicles for the in vitro transfection of human bone marrow mesenchymal stem cells (hMSCs) and for engineering MSCs to secrete brain-derived neurotrophic factor (BDNF). Different generations of cationic polyamidoamine dendrimers (generations 3–6) were tested on HEK 293T cells. hMSCs were then transfected with PAMAM-NH2 G4 dendrimers and Lipofectamine 2000, which elicited the expression of GFP reporter in around 6 and 20% of the cells, respectively. Both vehicles were then shown to elicit the expression of BDNF in MSCs from a bicistronic cassette. Non-virally induced neurotrophin expression may be a safe and easy method for adapting autologous stem cells for therapeutic treatment of diseases and neural system injuries.  相似文献

This study describes the isolation of three HeLa cell clones after exposure of HeLa cells to ethidium bromide (EB) in culture medium for either 14 days, or 14 days plusreexposure for 30 days. All three EB-induced clones differed from the parental HeLa cell in various physical properties of their mitochondria. The ratio of mitochondrial DNA component I to component II was altered in clone HeLa-2A. In addition, the cytochrome content of the respiratory chain a + a3, b and C1 decreased, while the cytochroms c content remained unchanged. The amount of cytochromes b and c1; which were not reduced by KCN treatment, but were reduced by dithionite, increased in clone HeLa-2A. The ultrastructure of HeLa-2A cells revealed several alterations characteristic of EB treatment. Some mitochondria had enlarged profiles, a reduced number of cristae and a more lucent electron density of the matrix. Other mitochondria were tightly packed with cristae, which occasionally showed a whorled configuration. These changes were observed 4 months (20–25 passages) after the omission of EB from the medium.  相似文献

Callus of the halophyte Mesembryanthemum crystallinum was used to study the effect of NaCl on the response to Botrytis cinerea infection. The fungus easily colonized the callus surface and the intercellular spaces. However, in the NaCl-adapted tissues the incidence of penetration was 67 % lower than in the inoculated control tissue. The modification of the infection pattern found in the salt-adapted callus could be related to metabolic adaptations to salinity. This was manifested by the enhanced antioxidant potential of ascorbate, the up-regulated activities of ascorbate peroxidase, as well as guaiacol and syringaldazine peroxidases together with the increased detoxification capacity of glutathione transferase in the NaCl-adapted callus. The post-inoculation changes in NaCl-adapted and non-adapted calli were roughly similar and supported the prooxidative nature of B. cinerea infection.  相似文献

Effects of a surface-active agent (Spumol BJ) and toxic molasses compounds on development and ultrastructure of Aspergillus niger strain R-16 mycelium producing citric acid in surface fermentation on molasses medium with 80% yield are presented. Microscopic observations showed that Spumol BJ in concentration of 5 microliters/100 cm3 as well as toxic molasses compounds stimulated the process of swelling and germinating of conidia. Giant conidia unable to germinate, appeared along with typical ones. Delicate mycelium outgrowing from germinating conidia however, sank after 20-24 h culture. Observations of hyphae in electron microscope showed changes in the ultrastructure and dimensions of mitochondria followed by successive degeneration of protoplast. The data obtained indicate that one of the reasons disqualifying raw molasses for biotechnological processes might be the excessive amount of surface-active antifoaming additives.  相似文献

Changes in tocopherol, chlorophyll and TBARS levels and the activities of antioxidant enzymes i.e., GSH-Px, GST, and SOD in chloroplasts of tomato plants subjected to moderate (50 mM) and severe (150 mM) NaCl stress were determined. Increase in tocopherol content around the second day under both stresses did not correlate with the chlorophyll degradation while such correlation was observed from the fifth day of severe stress. The activities of GSH-Px and GST as well as TBARS content showed NaCl-induced enhancement which was dose- and time-dependent. However, chloroplastic SOD was rather not involved in the response of tomato plants to NaCl stress. The obtained results suggest that under the moderate stress similarly as in the early phase of severe stress tocopherol functions as a typical antioxidant, while in the late phase of the latter it may be involved in senescence signaling pathway and enables the recovery and recycling of the compounds significant for a plant organism.  相似文献
