Siddiqi, M. Y. and Glass, A. D. M. 1987. Regulation of K+ influxin barley: Evidence for a direct control of influx by K+ concentrationof root cells.—J. exp. Bot. 38: 935–947. The kinetics of K+ (86Rb+) influx into intact roots of barley(Hordeum vulgare L. cv. Fergus) seedlings having different combinationsof root and shoot [K+], different growth rates and differentroot:shoot weight ratios were studied. K+ influx was stronglycorrelated with root [K+]; shoot [K+], growth rates, and root:shoot ratios appeared to have little effect on K+ influx. Adetailed study showed that both Vmax and Km for K+ influx wereaffected by root [K+] but not by shoot [K+]. We have suggestedthat factors such as growth rates and root: shoot ratio mayaffect K+ influx indirectly primarily via their influence onroot factors such as root [K+]. We have reiterated that othertypes of kinetic control, e.g. increased or decreased synthesisof ‘carrier systems’, may operate in addition todirect (allosteric?) control of K+ influx by root [K+]. Net accumulation of Cl by intact barley plants was virtuallyeliminated in roots and reduced by 40% in shoots when externalmedia (0.5 mol m–3 CaSO4 plus 0–5 mol m–3KCI) were supplemented with 0.25 mol m Ca(NO3)2. Plasmalemma36Cl influx (oc) was shown to be insensitive to externalNO3- in plants which had previously been grown in solutionslacking –3, but oc became sensitive to NO3-after a lagperiod of 3–6 h. Kinetic analyses revealed that the inhibitionof 36C1 influx by external NO3- was complex. At 0.25mol m–3 NO3- the Vmax for Cl influx was reducedby greater than 50%, with insignificant effects upon Km. At0.5 mol m–3 NO3- there was no further effect upon Vmaxbut Km for influx increased from 38±5 mmol m–3to 116±26 mmol m–3. By contrast, Cl effluxwas found to be insensitive to external NO3-. A model for theregulation of Cl influx is proposed which involves bothnegative feedback effects from vacuolar NO3- +Cl) concentrationand (external) NO3- inhibition of Cl influx at the plasmalemma.These combined effects serve to discriminate against Claccumulation, favouring NO3- accumulation, when the latter ionis available. Such observations are inconsistent with recentproposals for the existence of bona fide homeostats for chlorideaccumulation in higher plants. In plants that have been deprived of nitrate for a significant length of time, a constitutive high‐affinity nitrate transport system (cHATS) is responsible for initial nitrate uptake. This absorbed nitrate leads to the induction of the major nitrate transporters and enzymes involved in nitrate assimilation. By use of 13NO3 influx measurements and Blue Native polyacrylamide gel electrophoresis we examined the role of AtNRT2.5 in cHATS in wild type (WT) and various T‐DNA mutants of Arabidopsis thaliana. We demonstrate that AtNRT2.5 is predominantly expressed in roots of nitrate‐deprived WT plants as a 150 kDa molecular complex with AtNAR2.1. This complex represents the major contributor to cHATS influx, which is reduced by 63% compared with WT in roots of Atnrt2.5 mutants. The remaining cHATS nitrate influx in these mutants is due to a residual contribution by the inducible high‐affinity transporter encoded by AtNRT2.1/AtNAR2.1. Estimates of the kinetic properties of the NRT2.5 transporter reveal that its low Km for nitrate makes this transporter ideally suited to detect and respond to trace quantities of nitrate in the root environment.  相似文献

Movement of individuals promotes colonization of new areas, gene flow among local populations, and has implications for the spread of infectious agents and the control of pest species. Wild Norway rats ( Rattus norvegicus ) are common in highly urbanized areas but surprisingly little is known of their population structure. We sampled individuals from 11 locations within Baltimore, Maryland, to characterize the genetic structure and extent of gene flow between areas within the city. Clustering methods and a neighbour-joining tree based on pairwise genetic distances supported an east–west division in the inner city, and a third cluster comprised of historically more recent sites. Most individuals (~95%) were assigned to their area of capture, indicating strong site fidelity. Moreover, the axial dispersal distance of rats (62 m) fell within typical alley length. Several rats were assigned to areas 2–11.5 km away, indicating some, albeit infrequent, long-distance movement within the city. Although individual movement appears to be limited (30–150 m), locations up to 1.7 km are comprised of relatives. Moderate F ST, differentiation between identified clusters, and high allelic diversity indicate that regular gene flow, either via recruitment or migration, has prevented isolation. Therefore, ecology of commensal rodents in urban areas and life-history characteristics of Norway rats likely counteract many expected effects of isolation or founder events. An understanding of levels of connectivity of rat populations inhabiting urban areas provides information about the spatial scale at which populations of rats may spread disease, invade new areas, or be eradicated from an existing area without reinvasion.  相似文献

Marine primary producers adapted over eons to the changing chemistry of the oceans. Because a number of metalloenzymes are necessary for N assimilation, changes in the availability of transition metals posed a particular challenge to the supply of this critical nutrient that regulates marine biomass and productivity. Integrating recently developed geochemical, biochemical, and genetic evidence, we infer that the use of metals in N assimilation – particularly Fe and Mo – can be understood in terms of the history of metal availability through time. Anoxic, Fe-rich Archean oceans were conducive to the evolution of Fe-using enzymes that assimilate abiogenic     and     The N demands of an expanding biosphere were satisfied by the evolution of biological N2 fixation, possibly utilizing only Fe. Trace O2 in late Archean environments, and the eventual 'Great Oxidation Event' c . 2.3 Ga, mobilized metals such as Mo, enabling the evolution of Mo (or V)-based N2 fixation and the Mo-dependent enzymes for     assimilation and denitrification by prokaryotes. However, the subsequent onset of deep-sea euxinia, an increasingly-accepted idea, may have kept ocean Mo inventories low and depressed Fe, limiting the rate of N2 fixation and the supply of fixed N. Eukaryotic ecosystems may have been particularly disadvantaged by N scarcity and the high Mo requirement of eukaryotic     assimilation. Thorough ocean oxygenation in the Neoproterozoic led to Mo-rich oceans, possibly contributing to the proliferation of eukaryotes and thus the Cambrian explosion of metazoan life. These ideas can be tested by more intensive study of the metal requirements in N assimilation and the biological strategies for metal uptake, regulation, and storage.  相似文献

Eight cvs of barley (Hordeum vulgare L.) were separately plantedwith Wild Oats (Avena fatua L., genetically pure line CS40)in a sand culture with two external K+ concentrations. Substantialdifferences were observed among barley cvs in their abilityto compete with wild oat. The variety Fergus was highly competitiveat both high and low [K+]e, whereas Steptoe was competitiveonly at high [K+]e, and Compana was only weakly competitivewith wild oat. The differences between barley cvs were relatedto their previously reported efficiencies of K+ uptake and utilization.
