An in vitro evolution model was used to study changes in the genetic diversity of 24 strains of Pichia guilliermondii isolated from the midgut of bark beetles of the genus Dendroctonus . The genetic diversity of P. guilliermondii strains over 400 generations was analysed using multilocus enzyme electrophoresis (MLEE) and random amplified polymorphic DNA polymerase chain reaction (RAPD-PCR) markers. Resemblance relationships among strains were observed by cluster analysis. From the MLEE and RAPD markers, it was shown that the effective number of alleles, polymorphism and expected heterozygosity varied over the generations. The average heterozygosity among generations was statistically significant. Both the genetic diversity and the average heterozygosity were statistically significant among generations. The reduction in the population size from 109 to 105 yeast mL−1 associated with each transfer in P. guilliermondii strains and the clonal population structure observed along 400 generations suggest that genetic diversity changes and the observed replacement of genotypes are a consequence of a genetic drift process and not of the reproductive mode.  © 2008 The Linnean Society of London, Biological Journal of the Linnean Society , 2008, 93 , 475–486.  相似文献

The boundary between the Nearctic and Neotropical regions has been delineated using different approaches, methods and taxa. Using a panbiogeographical approach, identification of nodes can help understand the dynamics and evolution of the boundary. We analysed the distribution patterns of 46 Mexican land mammal species belonging to the Nearctic biotic component and delineated generalized tracks and nodes, in order to determine the southernmost boundary of the Nearctic region in Mexico. We found six generalized tracks and nine nodes; the latter located largely in the Sierra Madre Oriental, Transmexican Volcanic Belt, Sierra Madre del Sur and Chiapas biogeographical provinces. The highlands of Chiapas were found to represent the southernmost area inhabited by Nearctic taxa. The other biogeographical provinces, together with the Sierra Madre Occidental and Balsas Basin provinces, represent the Mexican transition zone in the strict sense. Arthropods have the capacity of recognizing self from non-selfin various defense phenomena including hemolymph clotting, phagocytosis,encapsulation, melanization and clearance of the foreign matterwhich must be initiated by a first step of molecular recognition."Natural" and experimentally induced humoral factors have beendescribed which act as hemagglutinins, bacterial agglutinins,precipitins, bactericidal factors, bacteriolysins, hemolysins,opsonins, clotting factors, and lysozymes. Their exact rolein recognition functions has not been fully explored and theirfunction remains unclear. Among these factors, the carbohydrate-bindingmolecules (lectins) are the best characterized in their specificity,physicochemical properties and molecular structure. Arthropodlectins are multimeric, high molecular weight protein (glycoprotein)molecules with a certain degree of heterogeneity in their specificityand structure. In particular, serum lectins from chelicerates(horseshoe crabs, scorpions and spiders) share a common property:the specificity for sialic acids. Arachnids and merostomes divergedin the earliest Cambrian. Since they occupy markedly differenthabitats, the sialic acid specific lectins most probably area relict rather than an adaptative character. In addition tothis common feature of specificity, lectins from cheliceratesand other arthropods represent heterogeneous populations whichcan bind a wide variety of carbohydrates, many of them presenton bacteria as D-galactose, 2-keto-3-deoxyoctonate, glucuronicacid, N-acetylmuramic acid, and colominic acid. Multiplicityin specificity suggests that serum lectins might contributeas a carbohydrate-based recognition system for the non-self.The requirement for avoiding self recognition would be thatcarbohydrate structures potentially recognized by the systemwould be absent, masked or out of reach of this humoral factoror cell associated recognition factors.  相似文献

The phylogenetic relationships of subtribe Chloraeinae, a group of terrestrial orchids endemic to southern South America, have not been satisfactorily investigated. A previous molecular phylogenetic analysis based on plastid DNA supported the monophyly of Chloraeinae and Gavilea, but showed that Chloraea is non‐monophyletic and that the sole species of Bipinnula analysed is sister to Geoblasta. However, that analysis included only 18 of the 73 species belonging to this subtribe. Here, the phylogenetic relationships of Chloraeinae were assessed by analysing aproximately 7500 bp of nucleotide sequences from nuclear ribosomal internal transcribed spacer (ITS) and plastid DNA (rbcL, matK, trnL‐trnF, rpoB‐trnC) for 42 species representing all four currently accepted genera of Chloraeinae and appropriate outgroups. Nuclear and plastid data were analysed separately and in combination using two different methods, namely parsimony and Bayesian inference. Our analyses support the monophyly of Chloraeinae and their inclusion in an expanded concept of Cranichideae, but none of the genera of Chloraeinae that includes more than one species is monophyletic. Gavilea and Bipinnula are paraphyletic, with Chloraea chica nested in Gavilea and Geoblasta penicillata in Bipinnula. As currently delimited, Chloraea is polyphyletic. The taxonomic changes proposed recently are for the most part not justifiable on phylogenetic grounds, except for recognition of the monotypic genus Correorchis. The lack of resolution for the relationships among species of ‘core’Chloraea suggests a relatively recent diversification of this group. Scolytine bark beetles are the most destructive pests of conifers; they sometimes aggregate in such large numbers that they actually kill their hosts. They maintain close relationships with yeasts and fungi, in particular those that are assumed to aid in digestive, detoxification processes and pheromone production. In this study, 403 yeast strains were isolated from the guts, ovaries, eggs and frass of nine bark beetle species in the genus Dendroctonus Erichson. The beetles were collected from 10 conifer species at 34 locations in Mexico, Guatemala and the USA. Yeast identification was based on partial DNA sequences from 18S rDNA, 26S rDNA and internal transcribed spacer (ITS1), as well as morphological and physiological characteristics. A combined phylogenetic analysis delimited 11 clades with sequences similar to Candida arabinofermentans , C. ernobii , C. membranifaciens (including C. lessepsii , Pichia mexicana and P. scolyti ), C. oregonensis , C. piceae , Kuraishia capsulata (including K. capsulata and K. cf. molischiana ), Pichia americana , P. canadensis , P. glucozyma , P. guilliermondii and an undescribed species of Candida . Nucleotide divergences between the major clades were at least 5% while, with the exception of 30 isolates, yeasts within clades differed from named reference species at fewer than 1% of the nucleotide sites. There do not appear to be obligate relationships between particular yeasts and specific anatomical partitions, nor between particular yeasts and bark beetle species. Quaternary climatic fluctuations have left contrasting historical footprints on the neutral genetic diversity patterns of existing populations of different tree species. We should expect the demography, and consequently the neutral genetic structure, of taxa less tolerant to particular climatic extremes to be more sensitive to long‐term climate fluctuations. We explore this hypothesis here by sampling all six pine species found in the Iberian Peninsula (2464 individuals, 105 populations), using a common set of chloroplast microsatellite markers, and by looking at the association between neutral genetic diversity and species‐specific climatic requirements. We found large variation in neutral genetic diversity and structure among Iberian pines, with cold‐enduring mountain species (Pinus uncinata, P. sylvestris and P. nigra) showing substantially greater diversity than thermophilous taxa (P. pinea and P. halepensis). Within species, we observed a significant positive correlation between population genetic diversity and summer precipitation for some of the mountain pines. The observed pattern is consistent with the hypotheses that: (i) more thermophilous species have been subjected to stronger demographic fluctuations in the past, as a consequence of their maladaptation to recurrent glacial cold stages; and (ii) altitudinal migrations have allowed the maintenance of large effective population sizes and genetic variation in cold‐tolerant species, especially in more humid regions. In the light of these results and hypotheses, we discuss some potential genetic consequences of impending climate change.  相似文献

The local density of individuals is seldom uniform in space and time within natural populations. Yet, formal approaches to the process of isolation by distance in continuous populations have encountered analytical difficulties in describing genetic structuring with demographic heterogeneities, usually disregarding local correlations in the movement and reproduction of genes. We formulate exact recursions for probabilities of identity in continuous populations, from which we deduce definitions of effective dispersal () and effective density (De) that generalize results relating spatial genetic structure, dispersal and density in lattice models. The latter claim is checked in simulations where estimates of effective parameters obtained from demographic information are compared with estimates derived from spatial genetic patterns in a plant population evolving in a heterogeneous and dynamic habitat. The simulations further suggest that increasing spatio‐temporal correlations in local density reduce and generally decrease the product , with dispersal kurtosis influencing their sensitivity to density fluctuations. As in the lattice model, the expected relationship between the product and the genetic structure statistic ar holds under fluctuating density, irrespective of dispersal kurtosis. The product D σ2 between observed census density and the observed dispersal rate over one generation will generally be an upwardly biased (up to 400% in simulations) estimator of in populations distributed in spatially aggregated habitats.  相似文献

The weevil subtribe Listroderina belongs in the tribe Rhytirrhinini (subfamily Cyclominae), and has 25 genera and 300 species in the Americas. The distributional history of this subtribe was reconstructed applying dispersal-vicariance analysis (DIVA) using its genera as terminals. The results suggest that Listroderina originated within an area presently represented by the Central Chile, Paramo, Puna, Patagonia and Subantarctic subregions of the Andean region. Posteriorly, the subtribe was affected by extinctions and was confined to Central Chile, Paramo and Subantarctic subregions. Later, extinctions and dispersals took place and the subtribe was restricted to the Paramo and Puna subregions. From there, a dispersal event to the Subantarctic subregion occurred, enlarging again the geographical range of the subtribe. Subsequently, a vicariant event separated the Puna and Paramo subregions from the Subantarctic one. While the Macrostyphlus generic group was confined to the Paramo and Puna subregions and from there dispersed to other areas, the Antarctobius , Falklandius , Listronotus , and Listroderes generic groups diversified in the Subantarctic subregion. The results obtained by DIVA may be linked to major geological events of South America. Thus, the geobiotic scenarios recorded in this subcontinent since the late Cretaceous could be used to interpret the biogeographical events which drove Listroderina evolution.  © 2003 The Linnean Society of London, Biological Journal of the Linnean Society , 2003, 80 , 339–352.  相似文献
