Effect of removal of snow cover in winter was investigated in an 80-year-old sugar maple (Acer saccharum Marsh.) stand in southern Quebec. We hypothesized that winter soil frost would induce some of the decline symptoms observed in sugar maple stands in southern Quebec in the early 1980's. Snow was continuously removed from around trees for a one week (partial removal) or for a four-month period (complete removal) during the 1990–1991 winter. Foliage and soils were sampled periodically during the summer of 1991. The complete snow removal treated trees showed decreased leaf water potential and increased peroxidase activity over most of the growing season. Foliar Ca was reduced in both snow removal treatments early in the growing season while foliar N was reduced in the complete snow removal trees late in the growing season. Soil NO 3 and K+ were elevated in both snow removal treatments at various times throughout the growing season. Prolonged soil frost in a sugar maple stand can induce lower leaf water potential, higher leaf peroxidase activity and early leaf senescence during the following growing season. Soil frost may have reduced nutrient uptake without affecting significantly the leaf nutrient status.  相似文献

A proof-of-principle experiment to inject a sub-attomole amount of a channel compound into a bilayer membrane is described. The system is based on reductive cleavage of a self-assembled gold-thiol monolayer. In 'macroscopic' experiments, 11-biphenyloxyundecane thiol formed well-ordered monolayers by open-circuit or controlled potential deposition. The products of reductive release were determined by chromatographic analysis. In DMF, the sole reduction product is the corresponding disulfide. In acetonitrile and water, only the thiol is detected. The current efficiency is low due to competing electrolysis of water, and to the low solubility of the released thiol or disulfide layer. On a 'microscopic' scale, the half ester of dithiodibutyric acid with gramicidin was deposited on a gold microelectrode under open circuit conditions. The thoroughly washed microelectrode, placed in proximity to a bilayer, released gramicidin only following a 100 ms pulse of reducing potential. The transfer efficiency of this method for controlled positioning of ion channels is estimated to be better than 1 part in 10(5).  相似文献

Ecosystem responses to current global climate change can be predicted through experimental climate simulations. One such simulation method is the open-top chamber (OTC). The effects of OTCs on environmental factors are potentially complex, and recognizing the numerous interactions among these factors is crucial for the proper use of chambers. We studied the effects of OTCs on microclimatic factors including ambient temperature, relative humidity, soil temperature, and soil moisture. Plant abundance responses were also assessed. Our study involved the construction of 20 OTCs (1 m in diameter and 0.75 m in height; made of clear acrylic plastic) and 20 control plots on substrates with and without Sphagnum moss, at post-fire and logging sites of the transitional mixedwood-boreal forest in the southern part of James Bay region, Quebec. Experimental trials were also conducted to test the effects of OTCs on snowmelt in the Montreal region. Our results suggest that OTC treatment is most evident in terms of increased daytime maximum temperatures (2°C to 3°C), and cooler (up to ~2.4°C), drier (up to 10% volumetric moisture content) soils. Advanced thawing of the insulating snow cover and exposure of soil in the OTCs to low spring temperatures appeared to prolong soil freeze and result in cooler soils. Earlier snowmelt probably also led to earlier onset and overall increased evaporation of meltwater in the OTCs, leading to drier soils. Plant abundance responses to OTC treatment differed depending on plant species. Overall, open-top chambers provide an effective and simple method of climate change simulation, but it is highly advisable that the complex interactive effects, both desired and undesired, are well understood and appreciated before using OTCs for experimental climate simulation.  相似文献

Understanding the links among plant genotype, plant chemistry, and food selection by vertebrate herbivores is critical to assess the role of herbivores in the evolution of plant secondary chemistry. Some specialized vertebrate herbivores have been shown to select plants differentially according to plant genotype, but examples from generalists, which constitute the vast majority of vertebrate herbivores, are few, especially in natural conditions. We examined the relationship between the North American porcupine (Erethizon dorsatum), a generalist mammalian herbivore, and clonal trembling aspen (Populus tremuloides), a preferred food source of porcupines. We determined preference for certain aspen trees through visual examination of porcupine climbing scars left on tree bark, and through a controlled feeding experiment. We used genetic and biochemical analyses to link the behavioral archives (climbing scars) left by porcupines on aspen trunks to the clonal structure and chemical composition of trees. We show that two phenolic glycosides (tremulacin and salicortin), which are under a high degree of genetic control and thus vary in concentration across clones, are the chemical variables that most influence (deter) feeding choices by porcupines. Using behavioral archives left by a wild herbivore on a natural stand of plants thus allowed us to demonstrate that a generalist vertebrate herbivore can choose plants according to their clonal structure and genetically based chemical composition. Our results contribute to extending previous findings obtained with generalist herbivores studied in controlled conditions, and with specialist herbivores studied in the field. Rapid anthropogenic habitat changes can lead to non‐ideal habitat use by animals, often resulting in lower fitness and population declines. An extreme case of use and fitness mismatch is an ecological trap where habitat quality cues are disjointed from the true quality of the habitat. Species primarily associated with anthropogenically altered habitat, such as red‐headed woodpeckers (Melanerpes erythrocephalus), may be especially vulnerable to use and fitness mismatch as they encounter novel environmental challenges. We investigated multi‐scale habitat use and nesting success of red‐headed woodpeckers to assess their vulnerability to mismatches between use and fitness as a result of non‐ideal habitat use across multiple scales. We found that habitat characteristics that promote feeding potential such as canopy openness and greater dead limb length appeared paramount and were consistent in use across several spatial scales although reproductive fitness suffered. This contrasts with the assumption that habitat use by nesting birds should instead favor predation avoidance at smaller scales to improve reproductive fitness and suggests that maladaptive, food‐based habitat use by red‐headed woodpeckers in southern Ontario may result in ecological traps for the species. Whether due to poor habitat choices or costly ones in favor of feeding potential, it is vital to consider this behavior in conservation and management plans for this and similar species. We suggest multi‐scale habitat use studies that consider fitness outcomes are critical for species‐at‐risk in human‐modified landscapes.  相似文献

Noradrenaline (1-10 microM) inhibited Ca2+-induced insulin secretion from electrically permeabilised islets of Langerhans with an efficacy similar to that for inhibition of glucose-induced insulin secretion from intact islets. The inhibition of insulin secretion from permeabilised islets was blocked by the alpha 2-adrenoreceptor antagonist, yohimbine. Adenosine 3',5'-cyclic monophosphate (cAMP) did not relieve the noradrenaline inhibition of Ca2+-induced secretion from the permeabilised islets, although noradrenaline did not affect the secretory responses to cAMP at substimulatory (50 nM) concentrations of Ca2+. These results suggest that catecholamines do not inhibit insulin secretion solely by reducing B-cell adenylate cyclase activity, and imply that one site of action of noradrenaline is at a late stage in the secretory process.  相似文献

Adenosine 3',5-cyclic monophosphate (cAMP) was shown to stimulate insulin secretion from electrically permeabilised islets of Langerhans incubated in Ca2+/EGTA buffers. cAMP-induced insulin secretion occurred in the presence of either sub-stimulatory (50 nM) or stimulatory (greater than 100 nM) concentrations of Ca2+. Similar effects on secretion were obtained in response to 8-bromo-cAMP (8-Br-cAMP) or the phosphodiesterase inhibitor, 3-isobutyl-1-methylxanthine. Forskolin (0.2-20 microM) increased adenylate cyclase activity and enhanced insulin secretion from the permeabilised islets. These results suggest that, in electrically permeabilised islets, cAMP-induced insulin secretion is not dependent on changes in cytosolic Ca2+.  相似文献

1. At least two distinct G-proteins are activated by light in squid photoreceptor membranes, a 45,000 mol. wt cholera toxin substrate and a 40,000 mol. wt pertussis toxin substrate. 2. The light-stimulated GTPase activity is partially inhibited by pretreatment with either toxin and abolished by treatment with both toxins. 3. At 24 degrees C, a free calcium ion concentration of 1 microM inhibits GTPase activity of both toxin substrates and ADP ribosylation by pertussis toxin. 4. This calcium sensitivity of squid G-proteins may be important in interpretation of experimental results on the phosphoinositide or other signalling pathways in squid visual transduction.  相似文献

The influence of tillage practices on native arbuscular mycorrhizal fungi (AMF) was studied in two, consecutive years in eastern Canada, in two 11 year-old long-term tillage-fertilizer experimental field soils, a sandy loam and a clay, growing corn in monoculture. The three tillage practices were: 1) conventional tillage (CT; fall plowing plus spring disking), reduced tillage (RT; spring disking) and no-till (NT). The corn crop received either inorganic (N and K) or organic (liquid dairy manure) fertilizers. Mycorrhizal hyphal density was estimated from soil samples obtained in early spring (before disking), at the 12–14 leaf stage, at silking, and at harvest. The percentage of corn root colonization by AMF at the 12–14 leaf stage, at silking and at harvest was also determined. The sandy loam was sampled over two consecutive seasons and the clay soil over one season.Densities of total and metabolically active soil hyphae, and mycorrhizal root colonization were significantly lower in CT soil than in RT and NT soil. Lowest soil hyphal densities were observed in early spring. The levels of intra- and extraradical fungal colonization always increased from spring to silking and decreased thereafter. Spring disking had only a small and transient negative effect on hyphal abundance in soil. Fertilization did not influence mycorrhizal colonization of corn or abundance of soil hyphae in the sandy loam soil, but in the clay soil metabolically active hyphae were more abundant with manure application than with mineral fertilization. In 1992, in both soils different tillage systems had same grain yield, however, in 1993, corn yield was higher in NT compared to CT system.  相似文献

A two-year study was undertaken in a maple-dominated watershed of southern Québec, Canada, to examine relationships between trends in fine root growth, stem diameter growth, soil moisture, soil temperature, mineralized-N and extractable-P. Until September, soil temperature was consistently higher in 1995 than in 1994. Apart from the first sampling in mid-May, soil moisture was higher in 1994 than in 1995. In 1994, most fine roots were produced before leaf expansion, whereas in 1995, fine root production peaked in July. Annual fine root production was estimated to be 2.7 times higher in 1994 than in 1995. Stem growth was strongly associated with the seasonal and annual variation in soil temperature. Root and diameter growth were asynchronous in 1994 but not in 1995. Fine root production was associated with two groups of variables: a soil fertility (mineralized-N and extractable-P) group and a physical soil environment (moisture and temperature) group. Our results are consistent with the negative effect of high soil-N fertility on fine root production but are inconclusive as to the positive effect of high soil-P fertility. Soil conditions that are detrimental to root growth such as high N availability and anaerobiosis could modify the normal dynamics of fine root growth.  相似文献
