Four-hourly volumetric measurements of airborne grass-pollen concentrations at Leiden, near the west coast of the Netherlands, were analyzed according to wet-versus-dry meteorological conditions in the pollen source area, and according to the distance of the over-land advection to the pollen sampler. Airborne pollen concentrations appear to be low when the source surface is wet by past or present rain, fog, or dew; and high when the source area is dry. Air adverted over long over-land distances from dry source areas contains much pollen, especially in the afternoon, due to pollen release and decreasing air turbulence. High nightly pollen concentrations are observed after a warm and dry day with much pollen release in distant source areas when the nocturnal meteorological conditions stimulate concentration of pollen grains into the lower layers of the atmosphere.Presented at the Sixth International Palynological Conference, 26 August – 1 September 1984, Calgary, Canada.  相似文献

Résumé Par un jeûne de trois jours ou par l'administration per os de réserpine(2 à 4 mg/kg) durant 15 à 24 jours, on a provoqué des ulcères d'estomac chez des rats. Une partie des bÊtes ainsi traitées fut exposée durant cette période et pendant 2 heures par jour à un air ionisé contenant 5.000 à 15.000 petits ions par cm3 d'air. Pour les animaux soumis au jeûne, la proportion d'ulcères se monte à 93% chez les bÊtes de contrÔle, alors qu'elle n'est que de 59% chez les bÊtes traÎtées par l'air ionisé. Pour les rats traÎtés à la réserpine, les proportions sont de 83,respectivement de 45%. On a en outre exposé 66 patients atteints d'ulcères d'estomac et d'intestins durant 12 à 15 jours et pendant 15 à 45 minutes par jour à un air contenant 5.000 à 10.000 petits ions négatifs par cm3 d'air. Pour 38 d'entre eux, la douleur et les autres symptÔmes avaient diminué ou disparu après le traitement.
Gastric ulcers were produced in rats by starvation for 3 days or the administration of reserpine (2 to 4 mg/kg) for 15 to 24 days. Some of the animals were exposed to ionized air for 2 hours daily during these periods(5,000 to 15,000 small negative ions/m3 air). The incidence of ulcer development was 93% in the untreated starved rats and 59% in those treated with ionized air; 83% in the animals treated with reserpine and 45% in those with additional ionized air treatment. In a clinical investigation 66 patients with peptic ulcer were exposed for 12 to 15 days to 5,000 to 10,000 small negative ions/m3 air daily. Pain and other symptoms were diminished or abolished after this treatment in 38 patients.

Zusammenfassung Bei Ratten wurden durch 3 Tage Hungern oder Behandlung mit Reserpin (2–4 mg/kg) wÄhrend 15 bis 24 Tagen Magenulcera provoziert. Ein Teil der Tiere wurde wÄhrend dieser Perioden 2 Std. tÄglich ionisierter Luft exponiert (5.000–15.000 kleine Ionen/m3 Luft). Die Ulcusrate betrug bei den unbehandelten Hungertieren 93%, bei den mit ionisierter Luft behandelten 59%; bei den Reserpin behandelten Tieren betrug sie 83% und bei den zusÄtzlich mit ionisierter Luft behandelten 45%. In der Klinik wurden 66 Patienten mit Magen-Darmulcera 12–15 Tage lang 15–45 min tÄglich mit 5.000 –10.000 neg. kleinen Ionen/m3 Luft behandelt. In 38 Patienten waren nach der Behandlung der Schmerz und die anderen Symptome abgeschwÄcht oder verschwunden.

Summary Airborne grass-pollen concentrations in six cities in the EEC are compared, based on observations from five years, 1982–1986. Results show that there are quantitative differences both between the monitoring stations and between the years. Very provisionally, the average seasonal total of the European urban airborne grass-pollen concentration can be put at 4 to 5000 per m3 of air. Also qualitatively, regarding the seasonal fluctuations, there are differences between the stations, and between the years. Generally, the results confirm that June is the most typical grass-pollen month in northwestern Europe, whereas in mediterranean Europe May is the more prominent grass-pollen period. There seems to be little coherence between the starting dates of the grass-pollen season even at relatively nearby stations in northwestern Europe, suggestive for a great influence of the actual weather situation.  相似文献

Dilution culture, a method for growing the typical small bacteria from natural aquatic assemblages, has been developed. Each of 11 experimental trials of the technique was successful. Populations are measured, diluted to a small and known number of cells, inoculated into unamended sterilized seawater, and examined three times for the presence of 104 or more cells per ml over a 9-week interval. Mean viability for assemblage members is obtained from the frequency of growth, and many of the cultures produced are pure. Statistical formulations for determining viability and the frequency of pure culture production are derived. Formulations for associated errors are derived as well. Computer simulations of experiments agreed with computed values within the expected error, which verified the formulations. These led to strategies for optimizing viability determinations and pure culture production. Viabilities were usually between 2 and 60% and decreased with >5 mg of amino acids per liter as carbon. In view of difficulties in growing marine oligobacteria, these high values are noteworthy. Significant differences in population characteristics during growth, observed by high-resolution flow cytometry, suggested substantial population diversity. Growth of total populations as well as of cytometry-resolved subpopulations sometimes were truncated at levels of near 104 cells per ml, showing that viable cells could escape detection. Viability is therefore defined as the ability to grow to that population; true viabilities could be even higher. Doubling times, based on whole populations as well as individual subpopulations, were in the 1-day to 1-week range. Data were examined for changes in viability with dilution suggesting cell-cell interactions, but none could be confirmed. The frequency of pure culture production can be adjusted by inoculum size if the viability is known. These apparently pure cultures produced retained the size and apparent DNA-content characteristic of the bulk of the organisms in the parent seawater. Three cultures are now available, two of which have been carried for 3 years. The method is thus seen as a useful step for improving our understanding of typical aquatic organisms.  相似文献

Abstract: Galactosylceramide ("galactocerebroside"; GalC) is a major glycolipid in the myelin sheath of the CNS and the PNS. The enzyme UDP-galactose:ceramide galactosyltransferase (CGalT) catalyzes the final step of the synthesis of GalC: the transfer of galactose to ceramide. By a differential screening approach, we have isolated a cDNA, the sequence of which is identical to the recently isolated cDNA clones for CGalT. By northern analysis and in situ hybridization we demonstrated that CGalT mRNA is expressed at birth in oligodendrocytes and Schwann cells, an expression pattern corresponding to the onset of myelination. In addition to the high expression levels of CGalT in oligodendrocytes and Schwann cells, in situ hybridization also showed expression in subtypes of neurons in spinal cord, cerebellum, and brainstem in the adult CNS, but at a much lower level than in oligodendrocytes. Expression of CGalT in COS cells demonstrated that CGalT has a preference for hydroxyceramide as a substrate. CGalT-expressing COS cells synthesize and transport GalC to their cell surface as shown by immunofluorescence and by lipid analysis of living cells. Our results suggested that the CGalT specifically uses hydroxyceramide for the synthesis of GalC and that separate (co)enzymes are not needed.  相似文献

In gram-negative bacteria only few proteins are exported across both the cytoplasmic membrane and the outer membrane which forms an extra barrier for protein excretion. In this review we describe the mechanisms of production and export of two types of plasmid-encoded proteins inEscherichia coli. These proteins are the bacteriocin cloacin DF13 and the K88ab and K99 fimbrial subunits. Specific so-called helper proteins located at different positions in the cell envelope play an essential role in the export of these proteins. The genetic organization, subcellular location and functions of these helper proteins, as well as the effects of mutations and culture conditions on the export of the proteins are described. Models for the export mechanisms are presented and future application possibilities for engineering foreign protein excretion inE. coli with these export systems are discussed.  相似文献

Summary A patient with the full clinical expression of the classical Lesch-Nyhan syndrome is presented with a residual hypoxanthine-guanine phosphoribosyl transferase (HGPRT) activity of 5–10% in erythrocyte lysate and about 30% in fibroblast lysate. The activities of other erythrocyte enzymes of purine metabolism were typical for a classical Lesch-Nyhan patient. The effects of allopurinol therapy on the excretion of urinary purine metabolites were studied by a newly developed isotachophoretic technique.The unusually high residual activity of HGPRT in erythrodytes and fibroblasts of the patient enabled the enzymologic characterization of the mutant enzyme: in fibroblasts the affinities for the substrates hypoxanthine and guanine were normal. However, there was an increased apparent K m for phosphoribosylpyrophosphate (PRPP), a complete absence of product inhibition by IMP and GMP, and a decreased heat stability. Addition of PRPP did not stabilize the mutant enzyme. In addition to the altered properties of the fibroblast enzyme, the K m of the erythrocyte enzyme for hypoxanthine was also increased.Immunoprecipitation experiments revealed the presence of an approximately normal amount of material cross-reacting with anti-human HGPRT antiserum. However, it appeared that this cross-reacting material had a decreased stability. When intact erythrocytes were incubated with radiolabeled purine bases, no formation of IMP or GMP could be detected, despite the relatively high residual activity of HGPRT in the hemolysate. The results fit the following hypothesis: as a consequence of a structural mutation affecting the PRPP-site of the enzyme and a decreased heat stability, the activity of the mutant enzyme under in vivo conditions is virtually zero.In the erythrocytes of the patient's mother a normal HGPRT-activity was found. However, the activity in her fibroblasts was lower than normal, while a decreased heat stability and an intermediate behavior towards IMP could be shown.Hair root analysis of several members of the patient's family confirmed the heterozygosity of the mother, whereas no other heterozygotes could be detected. The family anamnesis did not show other cases of Lesch-Nyhan syndrome. These findings were taken as evidence that the patient described in this paper might represent a mutation orginating from the gametes in either of the maternal grandparents.  相似文献

The synthesis of ketone bodies by intact isolated rat-liver mitochondria has been studied at varying rates of acetyl-CoA production and of acetyl-CoA utilization in the Krebs cycle. Factors which enhanced the rate of acetyl-CoA production caused an increase in the fraction of acetyl-CoA which was incorporated into ketone bodies. On the other hand, it was found that factors which stimulated the formation of citrate lowered the relative rate of ketogenesis. It is concluded that acetyl-CoA is preferentially used for citrate synthesis, if the level of oxaloacetate in the mitochondrial matrix space is adequate. The intramitochondrial level of oxaloacetate, which is determined by the malate concentration and the ratio of NADH over NAD+, is the main factor controlling the rate of citrate synthesis. The ATP/ADP ratio per se does not affect the activity of citrate synthase in this in vitro system. Ketogenesis can be described as an overflow of acetyl-groups: Ketone-body formation is stimulated only when the rate of acetyl-CoA production increases beyond the capacity for citrate synthesis. The interaction between fatty acid oxidation and pyruvate metabolism and the effects of long-chain acyl-CoA on mitochondrial metabolism are discussed. Ketone bodies which were generated during the oxidation of [1-14C] fatty acids were preferentially labelled in their carboxyl group. This carboxyl group had the same specific activity as the acetyl-CoA pool, whereas the specific activity of the acetone moiety of acetoacetate was much lower, especially at low rates of ketone-body formation. The activities of acetoacetyl-CoA deacylase and the hydroxymethylglutaryl-CoA (HMG-CoA) pathway were compared in soluble and mitochondrial fractions of rat- and cow-liver in different ketotic states. In rat-liver mitochondria, both pathways of acetoacetate synthesis were stimulated upon starvation or in alloxan diabetes. In cow liver, only the HMG-CoA pathway was increased during ketosis in the mitochondrial as well as in the soluble fraction.  相似文献
