Several workers have suggested that cell membranes have a high proton conductance. Our interest in this concept arose from the possibility that the nutrient (submucosal-facing) membrane of the gastric mucosa may have a high proton or hydroxyl ion conductance which would play a role in the regulation of the acid-base balance of the cell. We found that wide changes in the H+ concentration of the fluid bathing the nutrient side of the in vitro frog gastric mucosa did not result in significant changes in p.d. However, a maintained change of the H+ concentration of the bathing fluid would be expected to produce only a temporary change in p.d. Since a diffusion barrier is present on the nutrient side the temporary change in p.d. might be masked. An analysis of this possibility was made on the basis of a conceptual model and as a result of the analysis it is concluded that the proton (and/or OH?) conductance of the nutrient membrane of the frog gastric mucosa is not a significant fraction of its total conductance. The present status of the proton conductance hypothesis with respect to striated muscle and to the secretory membrane of the gastric mucosa is reviewed. 相似文献

Comparison of models for nucleotide substitution used in maximum- likelihood phylogenetic estimation 总被引:31,自引:15,他引:16
Using real sequence data, we evaluate the adequacy of assumptions made in
evolutionary models of nucleotide substitution and the effects that these
assumptions have on estimation of evolutionary trees. Two aspects of the
assumptions are evaluated. The first concerns the pattern of nucleotide
substitution, including equilibrium base frequencies and the
transition/transversion-rate ratio. The second concerns the variation of
substitution rates over sites. The maximum-likelihood estimate of tree
topology appears quite robust to both these aspects of the assumptions of
the models, but evaluation of the reliability of the estimated tree by
using simpler, less realistic models can be misleading. Branch lengths are
underestimated when simpler models of substitution are used, but the
underestimation caused by ignoring rate variation over nucleotide sites is
much more serious. The goodness of fit of a model is reduced by ignoring
spatial rate variation, but unrealistic assumptions about the pattern of
nucleotide substitution can lead to an extraordinary reduction in the
likelihood. It seems that evolutionary biologists can obtain accurate
estimates of certain evolutionary parameters even with an incorrect
phylogeny, while systematists cannot get the right tree with confidence
even when a realistic, and more complex, model of evolution is assumed.

We consider sex differences in human facial morphology in the context of developmental change. We show that at puberty, the height of the upper face, between the lip and the brow, develops differently in males and females, and that these differences are not explicable in terms of sex differences in body size. We find the same dimorphism in the faces of human ancestors. We propose that the relative shortening in men and lengthening in women of the anterior upper face at puberty is the mechanistic consequence of extreme maxillary rotation during ontogeny. A link between this developmental model and sexual dimorphism is made for the first time, and provides a new set of morphological criteria to sex human crania. This finding has important implications for the role of sexual selection in the evolution of anthropoid faces and for theories of human facial attractiveness. 相似文献

Interaction of sauger Sander canadensis and walleye Sander vitreus in a large,shallow northern river
The objectives of this study were to: (1) determine the relative abundance of sauger Sander canadensis and walleye S. vitreus within the Rainy River using a standardized index netting technique; (2) assess the life-history characteristics of the two species in a northern river; (3) examine the spatial distribution of the two species; (4) assess year-class synchrony. At a larger scale, relative abundance of S. canadensis and S. vitreus were similar among sections of the river. However, at a finer scale, S. canadensis were the dominant species over more area (24%) of the river than S. vitreus (12%). Life-history traits for both species were within the range reported for North American. Mortality rates were similar, suggesting that anglers were not affecting one species more than the other. Year-class strengths were not synchronous between S. canadensis and S. vitreus (r = 0.07). There was evidence of S. canadensis potentially outcompeting S. vitreus with a strong year class of S. canadensis followed by a very weak class of S. vitreus. Additionally, S. canadensis were larger than S. vitreus in most sections of the river when adjusted for mean size. However, the potential interspecific competition was not to the exclusion of S. vitreus. Turbidity was probably the factor that enable S. canadensis to survive sympatrically with S. vitreus given their inability to segregate by depth within the river. 相似文献

In this study we investigated the effect of glucose on GSK3β and β-catenin expression and the involvement of the N-linked glycosylation and hexosamine pathways in the Wnt canonical pathway in response to in vitro conditions resembling normoglycemia (5 mmol) and hyperglycemia (20 mmol) in the metastatic breast cancer-derived cell line MDA-MB-231. We also investigated the relationship between this circuitry and the thioredoxin-interacting protein (TXNIP) regulation that seems to be related. MDA-MB-231 cells were grown either in 5 or 20 mM glucose chronically prior to plating. For glucose shift (5/20), cells were plated in 5 mM glucose and shifted to 20 mM at time 0. Both protein and mRNA levels for GSK3β but only the protein expression for β-catenin, were increased in response to high glucose. Furthermore, we assessed the response of GSK3β, β-catenin, and TXNIP to inhibition of the N-linked glycosylation, hexosamine, and Wnt pathways. Wnt signaling pathway activation was validated by specific reporter assay. We show that high levels of glucose regulate mRNA and protein expression of GSK3β, and consequently higher levels of activated β-catenin protein, which locates to the nucleus and is associated with increased levels of cyclin D1 expression. This event coincides with increased level of N-terminal Ser 9 phosphorylation of GSK3β protein. The inhibition of both the hexosamine pathway and N-linked glycosylation along with Wnt signaling pathway by sFRP1 and DKK1 is associated with significant decrease of the protein levels of GSK3β, β-catenin, and TXNIP RNA. Our work illuminates a novel and never described before function of this signaling pathway that relates glucose metabolism with redox regulation mechanism. 相似文献

Crown rot and head blight of wheat are caused by the same Fusarium species. To better understand their biology, this study has compared 30 isolates of the three dominant species using 13 pathogenic and saprophytic fitness measures including aggressiveness for the two diseases, saprophytic growth and fecundity and deoxynivalenol (DON) production from saprophytic colonization of grain and straw. Pathogenic fitness was generally linked to DON production in infected tissue. The superior crown rot fitness of Fusarium pseudograminearum was linked to high DON production in the stem base tissue, while Fusarium culmorum and Fusarium graminearum had superior head blight fitness with high DON production in grains. Within each species, some isolates had similar aggressiveness for both diseases but differed in DON production in infected tissue to indicate that more than one mechanism controlled aggressiveness. All three species produced more DON when infecting living host tissue compared with saprophytic colonization of grain or straw, but there were significant links between these saprophytic fitness components and aggressiveness. As necrotrophic pathogens spend a part of their life cycle on dead organic matter, saprophytic fitness is an important component of their overall fitness. Any management strategy must target weaknesses in both pathogenic fitness and saprophytic fitness. 相似文献

An isolated occurrence of Fusarium head blight (FHB) of wheat was detected in the south-west region of Western Australia during
the 2003 harvest season. The molecular identity of 23 isolates of Fusarium spp. collected from this region during the FHB outbreak confirmed the associated pathogens to be F. graminearum, F. acuminatum or F. tricinctum. Moreover, the toxicity of their crude extracts from Czapek-Dox liquid broth and millet seed cultures to brine shrimp (Artemia franciscana) was associated with high mortality levels. The main mycotoxins detected were type B trichothecenes (deoxynivalenol and 3-acetyldeoxynivalenol),
enniatins, chlamydosporol and zearalenone. This study is the first report on the mycotoxin profiles of Fusarium spp. associated with FHB of wheat in Western Australia. This study highlights the need for monitoring not just for the presence
of the specific Fusarium spp. present in any affected grain but also for their potential mycotoxin and other toxic secondary metabolites. 相似文献
