Lake Rogoznica, near ibenik, Croatia, is a salt coastal lakewith a maximum depth of about 15 m. During most of the year,this small, naturally eutrophied lake is highly stratified,with hypoxia/anoxia occurring in the bottom layer. Total anoxia,with the presence of hydrogen sulphide, was recorded on September27, 1997. At that moment, massive mortality of all planktonicand benthic organisms was observed. After the anoxia, Acartiaitalica, the only planktonic copepod species of the lake, re-establishedquickly. Before anoxia, the population dynamics of the A.italicapopulation depended primarily on predator/prey relationships.However, in the post-anoxic period, nutrient–phytoplankton–copepodrelationships became more important, as there was no longerany predation pressure. From all accounts, it would appear thatonly A.italica is adapted to the extreme conditions which appearfrom time to time in the lake, and that this species has animportant role in the functioning of the lake ecosystem. Acartiaitalica was very important for the gradual normalization oflife in the lake after total anoxia.  相似文献

Background

Population-based biorepositories are important resources, but sample handling can affect data quality.

Objective

Identify metabolites of value for clinical investigations despite extended postcollection freezing delays, using protocols representing a California mid-term pregnancy biobank.

Methods

Blood collected from non-pregnant healthy female volunteers (n?=?20) underwent three handling protocols after 30 min clotting at room temperature: (1) ideal—samples frozen (??80 °C) within 2 h of collection; (2) delayed freezing—samples held at room temperature for 3 days, then 4 °C for 9 days, the median times for biobank samples, and then frozen; (3) delayed freezing with freeze–thaw—the delayed freezing protocol with a freeze–thaw cycle simulating retrieved sample sub-aliquoting. Mass spectrometry-based untargeted metabolomic analyses of primary metabolism and complex lipids and targeted profiling of oxylipins, endocannabinoids, ceramides/sphingoid-bases, and bile acids were performed. Metabolite concentrations and intraclass correlation coefficients (ICC) were compared, with the ideal protocol as the reference.

Results

Sixty-two percent of 428 identified compounds had good to excellent ICCs, a metric of concordance between measurements of samples handled with the different protocols. Sphingomyelins, phosphatidylcholines, cholesteryl esters, triacylglycerols, bile acids and fatty acid diols were the least affected by non-ideal handling, while sugars, organic acids, amino acids, monoacylglycerols, lysophospholipids, N-acylethanolamides, polyunsaturated fatty acids, and numerous oxylipins were altered by delayed freezing. Freeze–thaw effects were assay-specific with lipids being most stable.

Conclusions

Despite extended post-collection freezing delays characteristic of some biobanks of opportunistically collected clinical samples, numerous metabolomic compounds had both stable levels and good concordance.

Background

Low back pain (LBP) is the symptom of a group of syndromes with heterogeneous underlying mechanisms and molecular pathologies, making treatment selection and patient prognosis very challenging. Moreover, symptoms and prognosis of LBP are influenced by age, gender, occupation, habits, and psychological factors. LBP may be characterized by an underlying inflammatory process. Previous studies indicated a connection between inflammatory response and total plasma N-glycosylation. We wanted to identify potential changes in total plasma N-glycosylation pattern connected with chronic low back pain (CLBP), which could give an insight into the pathogenic mechanisms of the disease.

Methods

Plasma samples of 1128 CLBP patients and 760 healthy controls were collected in clinical centers in Italy, Belgium and Croatia and used for N-glycosylation profiling by hydrophilic interaction ultra-performance liquid chromatography (HILIC-UPLC) after N-glycans release, fluorescent labeling and clean-up. Observed N-glycosylation profiles have been compared with a cohort of 126 patients with acute inflammation that underwent abdominal surgery.

Results

We have found a statistically significant increase in the relative amount of high-branched (tri-antennary and tetra-antennary) N-glycan structures on CLBP patients' plasma glycoproteins compared to healthy controls. Furthermore, relative amounts of disialylated and trisialylated glycan structures were increased, while high-mannose and glycans containing bisecting N-acetylglucosamine decreased in CLBP.

Conclusions

Observed changes in CLBP on the plasma N-glycome level are consistent with N-glycosylation changes usually seen in chronic inflammation.

General significance

To our knowledge, this is a first large clinical study on CLBP patients and plasma N-glycome providing a new glycomics perspective on potential disease pathology.  相似文献

Introduction

Neonatal pulmonary hypertension (PH) is a common manifestation of bronchopulmonary dysplasia (BPD) and contributes to increased morbidity and mortality of preterm birth. Postnatal growth restriction (PNGR) and hyperoxia are independent contributors to PH development, as indicated by our previous work in a rat model of BPD.

Objective

To explore the metabolic consequences of induction of PH with hyperoxia and PNGR in a rat model of BPD.

Methods

Sprague–Dawley rat pups (n?=?4/group) underwent three modes of PH induction: (1) growth restriction-induced by larger litter size; (2) hyperoxia-induced by 75% oxygen exposure; (3) combined growth restriction and hyperoxia. Primary metabolism, complex lipids, biogenic amines, and lipid mediators were characterized in plasma and lung tissue using GC- and LC-MS technologies.

Results

Specific to hyperoxic induction, pulmonary metabolomics suggested increased reactive oxygen species (ROS) generation as indicated by: (1) increased indicators of β-oxidation and mitochondrial respiration; (2) changes in ROS-sensitive pathway activity and metabolites including the polyol pathway and xanthine oxidase pathways, and reduced glutathione; (3) decreased plasmalogens. Unlike the lung, circulating metabolite changes were induction mode-specific or additive in the combined modes (e.g. 1) growth-restriction reduced phosphatidylcholine; (2) hyperoxia increased oxylipins and trimethylamine-N-oxide (TMAO); (3) additive effects on 3-hydroxybutyric acid and arginine.

Conclusion

The present study highlights the variety of metabolic changes that occur due to PNGR- and hyperoxia-induced PH, identifying numerous metabolites and pathways influenced by treatment-specific or combined effects. The rat model used in this study presents a robust means of uncovering the mechanisms that contribute to the pathology of PH.

Dichlorodiphenyltrichloroethane (DDT) has been used extensively to control malaria, typhus, body lice and bubonic plague worldwide, until countries began restricting its use in the 1970s. Its use in malaria control continues in some countries according to recommendation by the World Health Organization. Individuals exposed to elevated levels of DDT and its metabolite dichlorodiphenyldichloroethylene (DDE) have an increased prevalence of diabetes and insulin resistance. Here we hypothesize that perinatal exposure to DDT disrupts metabolic programming leading to impaired metabolism in adult offspring. To test this, we administered DDT to C57BL/6J mice from gestational day 11.5 to postnatal day 5 and studied their metabolic phenotype at several ages up to nine months. Perinatal DDT exposure reduced core body temperature, impaired cold tolerance, decreased energy expenditure, and produced a transient early-life increase in body fat in female offspring. When challenged with a high fat diet for 12 weeks in adulthood, female offspring perinatally exposed to DDT developed glucose intolerance, hyperinsulinemia, dyslipidemia, and altered bile acid metabolism. Perinatal DDT exposure combined with high fat feeding in adulthood further impaired thermogenesis as evidenced by reductions in core temperature and in the expression of numerous RNA that promote thermogenesis and substrate utilization in the brown adipose tissue of adult female mice. These observations suggest that perinatal DDT exposure impairs thermogenesis and the metabolism of carbohydrates and lipids which may increase susceptibility to the metabolic syndrome in adult female offspring.  相似文献

The qualitative composition, numerical abundance and verticaldistribution of radiolarians, tintinnines, nauplii and smallcopepod assemblages were studied in the central area of theSouthern Adriatic Pit at three stations during 10 cruises fromOctober 1985 to May 1990. The samples were collected with aplankton net of 53 µm mesh size equipped with a closingsystem in eight vertical layers. Data are presented for 53 radiolarians,61tintinnines and for the first time for 22 poecilostomatoidspecies in the Adriatic Sea. According to the numerical abundanceof assemblages and the frequency of occurrence of the bulk ofthe individual species population in the water column, fourcommunities could be defined: the surface (0–50 m), thesubsurface (50–100m), the midwater (100–600 m) andthe deep-sea community (below 600 m). The euphotic layer wascharacterized by tintinnines, copepod nauplii, cyclopoids andjuvenile calanoid copepods, while radiolarians and poecilostomatoidcopepods dominated in the deeper layers. The highest numericaldifferences between stations and seasons were noted only onthe surface. Towards the deeper layers, the differences wereconstantly smaller, and below400 m there was a uniform distributionof all assemblages. This research, on the basis of the numericalabundance of protozoans and micrometazoans, shows that the centralpart of the South Adriatic Pit is considerably richer than mentionedby earlier authors.  相似文献

The crystal structure of α-D-Manp-(1→3)-β-D-Manp-(1→4)-α-D-GlcNAcp has been determined by the direct method using the multi-solution, tangent formula, and “magic integer” procedures. The space group is P22, and 2 molecules are in the unit cell with a  9.894 (5), b  10.372 (6), c  11.816 (6) Å, and β  95.03° (6). The structure was refined to R 0.059 for 2099 reflections measured with Mo Kα radiation. Difference synthesis showed all the hydrogen atoms, and indicated a partial (~30%) substitution of the α-anomer molecules by the β-anomer molecules. The D-mannopyranose and the D-glucopyranose have the normal 4C1 conformation; an intramolecular hydrogen-bond O-3″-H.....O-5′ (2.703 Å) stabilises the GlcNAc in relation to β-D-mannopyranose.  相似文献

A new species of Mesaiokeras is described from the hyperbenthosof Veliko Jezero, a marine lake on the island of Mljet, Croatia.This is the first species of this genus and family (Mesaiokeratidae)reported from the Adriatic and Mediterranean seas. It is characterizedby small body length (an average 361 µm for females and337 µm for males), the low number of free segments (21)and the armature of the antennules, very long spine (a2) ofexopodite I of female right leg 4, the presence of long processeson the ventroposterior area of the female genital double-somite,the form and surface ornamentation of the genital area, theform and ornamentation of male and female leg 5, and the formand armature of caudal rami. This species was found in shallowwater with very stable hydrographic conditions and at a populationdensity from 1 to 26 ind. m-3.  相似文献

A new copepod genus and species, Speleohvarella gamulini, collectedfrom the anchialine cave iva Voda on Hvar Island (Croatia),is described from both sexes. This is the first record of acalanoid copepod found in an Adriatic anchialine cave. The newgenus is distinguished from other Stephidae by a combinationof the following features: the cephalosome is separate fromthe first pedigerous somite, and the fourth and fifth pedigeroussomites are fused; the female urosome has 4 segments, the genitaldouble-somite is symmetrical and the caudal rami is asymmetricalwith the right longer than the left; antennules of both sexesare symmetrical and 24-segmented, with the fusion of ancestralsegments II–IV, X–XI and XXVII–XXVIII; theexopod of antenna is 7-segmented; 2 setae are present on thebasal exite of maxillule; in the male the right leg 5 is shortand 3-segmented, and the left leg elongate and 5-segmented.  相似文献

During an infection, one of the principal challenges for the host is to detect the pathogen and activate a rapid defensive response. The Toll-like family of receptors (TLRs), among other pattern recognition receptors (PRR), performs this detection process in vertebrate and invertebrate organisms. These type I transmembrane receptors identify microbial conserved structures or pathogen-associated molecular patterns (PAMPs). Recognition of microbial components by TLRs initiates signaling transduction pathways that induce gene expression. These gene products regulate innate immune responses and further develop an antigen-specific acquired immunity. TLR signaling pathways are regulated by intracellular adaptor molecules, such as MyD88, TIRAP/Mal, between others that provide specificity of individual TLR- mediated signaling pathways. TLR-mediated activation of innate immunity is involved not only in host defense against pathogens but also in immune disorders. The involvement of TLR-mediated pathways in auto-immune and inflammatory diseases is described in this review article.  相似文献
