A Bioluminescence Method for the Measurement of l-Glutamate: Applications to the Study of Changes in the Release of l-Glutamate from Lateral Geniculate Nucleus and Superior Colliculus After Visual Cortex Ablation in Rats

We have developed a rapid, simple, specific, and very sensitive bioluminescence method for the measurement of L-glutamate (L-Glu). Oxidation of L-Glu by glutamate dehydrogenase has been coupled with bacterial FMN reductase and luciferase. Light production (i.e., peak height or integral) was linear from less than 0.5 to 500 pmol of L-Glu. Potential interfering substances that may be encountered in brain tissue have been identified. The most potent inhibitors were ascorbate and the biogenic amines. Procedures that conferred long-term stability of the reagent mixture (greater than 8 h) were established. Bioluminescence analysis of L-Glu content in brain tissue extracts, fractions from release experiments, and human CSF corroborated respective results obtained by HPLC analysis. In this study, we have applied the method to monitor changes in the KCl-evoked release of endogenous L-Glu from milligram amounts of brain tissue, i.e., from lateral geniculate nucleus and superior colliculus after visual cortex ablation.     

Effect of neurotensin on pancreatic function in man

The effect of neurotensin on submaximally-stimulated hepatobiliary and pancreatic secretion was studied in 6 healthy subjects. An intravenous infusion of neurotensin 1.4 ± 0.3 pmol/kg/min, designed to reproduce plasma neurotensin immunoreactivity levels within the physiological range, produced a significant increase in pancreatic bicarbonate output. Plasma concentrations of pancreatic polypeptide rose by 83 ± 16 pmol/l and were associated with a small reduction in trypsin, but no significant change in bilirubin outputs.     

Différenciation des mécanismes induisant la segmentation et l'asymétrie des chromatides, après traitement par le 5-bromodéoxyuridine

5-Bromodeoxyuridine (BUdR)-induced segmentation and BUdR-induced asymmetry of the chromatids probably reflect a two-stage mechanism. The first stage consists of a BUdR substitution of the thymidine in the DNA during S period. The second stage, less obviously demonstrated, involves either alteration of chromosomal proteins or an imperfect association between substituted DNA and normal or abnormal proteins. This second stage takes place at different times, according to the type of chromatid modification: during S or G 2 period in the case of segmentation, and during G 1 in the case of asymmetry. One important implication of our experimental results is that the appearance of metaphasic chromatids is at least partly determined by the time of the G 1 period.L'induction d'une asymétrie, ou d'une segmentation des chromatides par un traitement au BUdR, résulte probablement d'un mécanisme à deux étapes distinctes. La première consiste en une substitution de la thymidine par le BUdR survenant lors de la replication de l'ADN. La seconde, plus difficile à mettre en évidence, consiste en une modification des protéines chromosomiques, ou en une mauvaise association entre les protéines, modifiées ou non, et l'ADN ayant incorporé le BUdR. Cette seconde étape est nécessairement différente selon la modification chromatidienne induite: elle se déroule en phase S ou G 2, en cas de segmentation, et en phase G 1 surtout, en cas d'asymétrie. Dans l'un et l'autre cas, la modification de l'association ADN-protéines pourrait être en rapport avec la régulation chromosomique. Une déduction importante de nos résultats expérimentaux est que l'aspect des deux chromatides métaphasiques dépend, au moins en partie, de la constitution de l'ADN dès la phase G 1.     

A possible effect on the Shwartzman reaction in the rabbit caused by fentanyl-fluanisone tranquillization

Shwartzman's reaction was elicited in four Chinchilla rabbits. Two of the animals were tranquillized with fentanyl-fluanisone (Hypnorm: Janssen Pharmaceuticals). There was a reduced response in the tranquillized rabbits while the untranquillized animals developed the typical dose-responsive skin reaction. Fentanyl-fluanisone tranquillization may interfere with the development of the Shwartzman reaction in Chinchilla rabbits.     

Effect of Metalaxyl on the incidence and severity of Pearl millet downy mildew disease in Northern Nigeria

Pearl millet downy mildew (DM) incidence, severity and yield losses of two pearl millet varieties (local and improved) due to the disease were determined in the field. Significant differences in the disease incidence and severity were recorded in the plots sown with metalaxyl-treated seeds and those sown with non-treated seeds, indicating the efficacy of the fungicide on the fungus. Yield losses due to non-treatment of seeds with metalaxyl was 40.88 and 45.39% in a local variety and 43.00 and 18.60% in an improved variety in the 2000 and 2001 cropping seasons respectively. Significant differences between plots sown with metalaxyl-treated and those sown with non-treated seeds were obtained for other yield components such as 1000-grains weight, panicle length and weight.     

Ostéomyélite de l’apophyse odontoïde : intérêt de la TEP/TDM au 18F-FDG dans le bilan d’extension infectieux à distance

Odontoid process is an atypical and very rare localization of osteomyelitis. We reported the case of a 72-year-old hemodialysed man with methicillin-sensitive Staphylococcus aureus osteomyelitis of the odontoid process. Osteomyelitis was diagnosed at MRI and ¹⁸F-FDG PET/CT. While clinical examination and conventional radiographs were non contributive, ¹⁸F-FDG PET/CT also allowed the diagnosis of right foot osteomylitis and multiple vertebral septic localizations. ¹⁸F-FDG PET/CT done at month 3 demonstrated a regression of the odontoid and foot hypermetabolic activity. This case illustrates the atypical presentation of this septic localization and the usefulness of ¹⁸F-FDG PET/CT to perform whole body screening and detect septic metastasis.
