The reactivity of 100 sera taken from patients with different blood diseases and donors with respect to synthesized peptides in the variable area of protein NS4 of hepatitis C virus was studied. The presence of type-specific antibodies in the blood sera of patients with hepatitis C was shown. Two antigenic determinant corresponding to 1683-1705 and 1711-1732 amino acid residues in the protein area under study were detected. In hematological patients undergoing frequent blood transfusions mixed infection with different types of hepatitis C virus was registered; these types could be reliably determined with the use of synthetic peptides. The serotype determined with the use of peptides corresponded to the type of the circulating virus.  相似文献

A shortening of the lag phase in dichloromethane (DCM) consumption was observed in the methylobacteria Methylopila helvetica DM6 and Albibacter methylovorans DM10 after prior growth on methanol with the presence of 1.5% NaCI. Neither heat nor acid stress accelerated methylobacterium adaptation to DCM consumption. Sodium azide (1 mM) and potassium cyanide (1 mM) inhibited consumption of DCM by these degraders but not by transconjugants Methylobacterium extorquens AM1, expressing DCM dehalogenase but unable to grow on DCM. This indicates that the degrader strains possess energy-dependent systems of transport of DCM or chloride anions produced during DCM dehalogenation. Inducible proteins were found in the membrane fraction of A. methylovorans DM10 cells adapted to DCM and elevated NaCl concentration.  相似文献

The expression of β-III tubulin, a marker protein of early neuronal cells, was studied by molecular genetic and immunochemical techniques. The study was performed with the eyes of human fetuses on weeks 8.5 to 27–28 of intrauterine development. Expression of β-III tubulin was detected immunochemically in the retina and lens fibers of fetuses at stages of 8.5 to 22–23 weeks. By means of PCR, a strong expression of the β-III tubulin gene was revealed in the retina of 9.5-week fetuses. Its level remained high until week 18, became slightly lower by week 24, and decreased to zero by week 27–28. The expression of this gene was also revealed in the lens of 9.5-week fetuses. Its level at stages of 15 to 24 weeks was very low, and no expression could be detected in 27-to 28-week fetuses. The results of PCR analysis were consistent with immunochemical data.  相似文献

The effect of malonate and sedaxane, a compound with the fungicidal effect which act as succinate dehydrogenase inhibitors, on the resistance of etiolated wheat seedlings (Triticum aestivum L.) to osmotic stress caused by 12% PEG 6000 solution, was studied. The presowing treatment of seeds with 0.3 mM sedaxane solution significantly reduced the inhibitory effect of osmotic stress on seedling growth. The protective effect of 10 mM malonate was significant when it was added to the incubation medium of the roots; the effect of preseeding treatment with malonate was less significant. Unlike malonate, malate had no positive effect on seedling growth under osmotic stress. The activity of succinate dehydrogenase and the hydrogen peroxide content decreased in seedlings after the treatment of roots with malonate and sedaxane. Pretreatment with sedaxane and the addition of malonate to the incubation medium of roots prevented the accumulation of a lipid peroxidation product, malondialdehyde, which is caused by osmotic stress, and increased peroxidase activity. It was concluded that the stress-protective effect of sedaxane and malonate on wheat seedlings might be due to the inhibition of succinate dehydrogenase-dependent formation of reactive oxygen species and the prevention of oxidative cell damage.  相似文献

The effect of hydrogen sulfide (H2S) donor sodium hydrosulfide (NaHS) on the heat resistance of wheat (Triticum aestivum L.) coleoptile cells, the formation of reactive oxygen species (ROS), and the activity of the antioxidant enzymes in them was investigated. The treatment of coleoptiles with 100 µM NaHS caused transient enhancement of the generation of the superoxide anion radical (O2 ?) and an increased hydrogen peroxide content. The activities of antioxidant enzymes—superoxide dismutase, catalase, and guaiacol peroxidase— and coleoptile resistance to damaging heat was later found to have increased. The biochemical and physiological effects of the hydrogen sulfide donor described above were inhibited by the treatment of wheat coleoptiles with the hydrogen peroxide scavenger dimethylthiourea, the NADPH oxidase inhibitor imidazole, the extracellular calcium chelator EGTA, and the phosphatidylinositol-specific phospholipase C inhibitor neomycin. A conclusion was made on the role of ROS generation, which is dependent on the activity of NADPH oxidase and calcium homeostasis, in the transduction of the H2S signal, which induces antioxidant enzymes and the development of plant cell heat resistance.  相似文献

The transformants of Methylobacterium dichloromethanicum DM4 (DM4-2cr-/pME8220 and DM4-2cr-/pME8221) and of Methylobacterium extorquens AM1 (AM1/pME8220 and AM1/pME8221) that express the dcm A gene of dichloromethane dehalogenase undergo lysis when incubated in the presence of dichloromethane and are sensitive to acidic shock. The lysis of the transformants was found to be related neither to the accumulation of Cl- ions, CH2O, and HCOOH, nor to the impairment of glutathione synthesis or to the maintenance of intracellular pH. The (exo-) Klenow fragment-mediated incorporation of [alpha-32P]dATP into the DNA of the transformants DM4-2cr-/pME8220 and AM1/pME8220 was considerably greater when the transformed cells were incubated with CH2Cl2 than when they were incubated with CH3OH, indicating the occurrence of a significant increase in the total length of gaps. At the same time, the strain AM1 (which lacks dichloromethane dehalogenase) and the dichloromethane-degrading strain DM4 incubated with CH2Cl2 showed an insignificant increase in the total length of the gaps. The transformed cells are likely to lyse due to the relatively inefficient repair of DNA lesions that are induced in response to the alkylating action of S-chloromethylglutathione, an intermediate product of CH2Cl2 degradation. The data obtained suggest that the bacterial mineralization of dichloromethane requires an efficient DNA repair system.  相似文献
