A spin labeled fatty acid (16-doxylstearic acid) was linked to a photochemical reacting group (azido derivative). When the molecule is introduced, at a low concentration, into rabbit sarcoplasmic reticulum membranes, the spectrum before illumination is identical to the spectrum obtained with the corresponding spin labeled fatty acid. After illumination, a large immobilized components is seen. It corresponds to about 70% of the ESR signal of the effectively bound label, at room temperature. The fraction of immobilized component varies with temperature, from 100% at 0°C to 50% at 35°C. Addition of a small amount of detergent (dodecyl octaethylene glycol monoether), under non solubilizing conditions, decreases the fraction of signal due to a strongly immobilized probe. A possible interpretation is that the immobilized signal reflects protein bound spin labels trapped in Ca2+ ATPase oligomers, which are partially dissociated by detergent addition or temperature increase. 相似文献

Immunocytological investigations have been performed on semi-thin sections of human fetal pituitaries ranging in fetal age from 6 to 26 weeks. Corticotrophs can be revealed by anti-ACTH (1-24), anti-ACTH (17-39) and anti-beta MSH but not by anti-alpha MSH immunesera from the 8th week. Somatotrophs are revealed with anti-human STH from 9th week. Differentiating cells containing only alpha subunits of glycoproteic hormones are present from 8th to 12th week. At 13th week beta subunits of TSH can be revealed immunocytologically in thyreotrophs. Beta subunits of LH or FSH can be detected in same gonadotrophic cells only from 15th week. 相似文献

Phenotypic analysis with monoclonal antibodies showed that OKT8+ T cells represent the main cell population of the periparasitic mononuclear cell infiltrate in the liver of 7 patients with alveolar echinococcosis. A significant decrease of the OKT8+ subpopulation had been previously demonstrated in the peripheral blood mononuclear cells of these patients. The results obtained in this study suggest an intrahepatic homing for these particular T cells. We have cloned and sequenced DNAs complementary to the mRNA encoding the precursor of the rat melanin-concentrating hormone. This allowed us to elucidate the primary structure of the 96 C-terminal residues of this precursor. It contains three possible sites for enzymatic cleavage enabling the generation of MCH and of two additional neuropeptides. These three peptides can bind one of our antisera raised against human somatocrinin 1-37, alpha-melanotropin and salmon melanin-concentrating hormone, which immunocytochemically stain the same neuron population in the hypothalamus. 相似文献

They trained on a cycloergometer 3 times/wk for 16 wk(50-80%O2 max,then 80-85%O2 max). They werechecked at 8 wk, 16 wk, and 4 mo after detraining. Training improvedO2 max (+16%) andinduced plasma volume expansion (+11%). No change in total body water,extracellular fluid, interstitial and intracellular fluid volumes,fat-free mass, and body weight was detected in this small sample withtraining. Body fat mass decreased (2.1 ± 2.2 kg).Echocardiography at rest showed increased fractional shortening andejection fraction and decreased left ventricular end-systolic dimension(P < 0.05). Blood volume expansioncorrelates with cardiac contractility and has an impact on cardiacfunction. In the external layer of the median eminence of the fox, the somatostatin-containing fibers and neurophysin-containing fibers of the hypothalamo-infundibular tract are located in distinct areas. In the neural lobe, somatostatin-positive areas are simultaneously neurophysin-positive. Outside the SON and PVN, some somatostatin-positive and neurophysin-negative perikarya are scattered close to the third ventricle. These facts suggest the existence of two somatostatin systems: a hypothalamo-infundibular (neurophysin-negative) one and a hypothalamo-neurohypophyseal (neurophysin-positive) one. 相似文献

On paraffin or semi-thin sections various anti-LH or anti-TSH sera stain indifferently all the thyrotrophs and the gonadotrophs. Inversely anti-beta-TSH, anti-beta-LH or anti-beta-FSH purified sera permit the discrimination of these two cell populations. The constancy of fixation of the anti-beta-LH and anti-beta-FSH sera on all the gonadotrophs gives evidence of their ability to produce both FSH and LH. However in a few female rats the central gonadotrophs are stained more weakly by anti-beta-FSH serum than by anti-beta-LH serum. The purification of the antisera by adjunction of hormonal antigens (alpha subunits or heterologous hormone) does not enable, with the PAP technique on thin sections, a selective staining of the secretory granules of the thyrotrophs or of the gonadotrophs. 相似文献
