Hybrid protein, cancer necrosis factor thymosin-α1 (TNF-T), when synthesizing in strain-producer of Escherichia coli SG200-50 with plasmid pThy315, was a part of “inclusion bodies” mostly in the form of a high-molecular complex with other proteins due to the S-S bonds formation. An approach of purification of TNF-T has been proposed, which is based on the destruction of the complex in the presence of sodium dodecylsulfate (DDS-Na) and dithiotreitol (DDT) followed by gel-filtration on Sephadex G-100 and renaturation by ultrafiltration on hollow fibers. The method allows the isolation of electrophoretically homogeneous TNF-T containing no DDS-Na and having high cytotoxic activity against cancer cells of mouse adenocarcinome L-929. The yield of TNF-T achieved 80% relative its content in biomass and 30% relative the total protein. 相似文献

Atp-activated ionic permeability in smooth muscle cells isolated from the guinea pig urinary bladder
Smooth muscle cells from the guinea pig urinary bladder were investigated by voltage clamping at the plasma membrane and using an intracellular perfusion technique. Applying adenosine triphosphate (ATP) at a concentration greater than 3 × 10–8 M and at a membrane potential of –100 to –30 mV produced a rise in fast inward transmembrane current. A similar effect was exerted by adenosine diphosphate (ADP) and -, -, and ,-methylene ATP. Application of guanosine triphosphate, inosine triphosphate, adenosine monophosphate (AMP), and adenosine failed to activate this current. It was found that AMP blocks ATP receptors competitively. No pharmacological differences were found between the latter ATP receptors and those of rat sensory neurons. The ATP receptors were rapidly desensitized and recovered their sensitivity to agonists extremely slowly. Speed of desensitization was reduced by a decrease in ATP concentration.A. A. At high doses (200-400 Gy) radiation-induced changes played a major role in the development of the cerebral form of radiation sickness. It is shown that in case of antioxidant insufficiency (AOI) activation of NADPH- and ascorbate-dependent lipid peroxidation (LPO) in sarcoplasmic reticulum (SR) of skeletal muscles proceeds 1.7 and 4.1 times faster, respectively. Activation of lipid peroxidation in AOI leads to damage of Ca2+ transport processes in SR of skeletal muscles. Under these conditions ATP-dependent accumulation of 45Ca (by 88%) and Ca(2+)-ATPase (by 14%) activity in SR of skeletal muscles falls. In case of AOI a significant disturbance of passive Ca2+ transport in SR of skeletal muscles takes place, being characterized by an increased passive 45Ca output from vesicles due to breakage of the biomembrane permeability as a result of lipid peroxidation of membranes. Treatment of animals with ionol, a synthetic antioxidant, causes a decrease of activated NADPH- and ascorbate-dependent LPO in SR of skeletal muscles and stabilization of Ca2+ transport processes. 相似文献
