Chlorophylls c1 and c2 have been separated from total pigment extracts of the alga Pavlova gyrans Butcher using a reversed-phase high-performance liquid chromatography system. Pigments were separated on a 5 micrometer C18 column (25 centimeters × 4.6 millimeters) using a gradient of methanol-acetonitrile-water. Other photosynthetic pigments were also well resolved by the system used. The separation system described may replace current thin layer chromatography methods for qualitative and quantitative determination of chlorophyll c species.  相似文献

A light-harvesting fucoxanthin-chlorophyll a/c-protein complex has been isolated from the diatom Phaeodactylum tricornutum by detergent extraction of thylakoid membranes coupled with sucrose density gradient centrifugation. The isolated complex was devoid of photochemical activity and displayed spectral characteristics consistent with light harvesting function. It has three major polypeptides of apparent molecular weights 18,000, 19,000, and 19,500 as determined by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Using protein synthesis inhibitors, these polypeptides were shown to be synthesized on 80S cytoplasmic ribosomes. Antibodies raised to a mixture of the 19,000 and 19,500 dalton components of the complex were used to demonstrate structural similarity among the three polypeptide components. Immunoprecipitation from primary translation products synthesized in a reticulocyte lysate system primed with P. tricornutum poly(A) RNA, indicates that the polypeptide components are synthesized as precursors 3,000 to 5,000 daltons larger than the mature polypeptides.  相似文献

Photosystem II light-harvesting complexes were isolated from a number of ulvophycean algae. Some of these light-harvesting complexes displayed unusual features, most notably a high apparent molecular weight (ca. 58,000) when isolated by lithium doderyl sulfate polyarrylamide gel electrophoresis. Other ulvophycean light-harvesting complexes had a low-molecular weight (ca. 30,000). The distribution of the high-molecular weight complex was limited to certain members of the Caulerpales and Blastophysa rhizopus (Siphanocladales). Within the Caulerpales, there were also spectral differences between the high-molecular weight and low-molecular weight light-harvesting complex types. The differences in light-harvesting complexes in the Ulvophyceae suggest that there are two lines of evolution in the Caulerpales and that Blastophysa may be an intermediate between the Siphon-ocladales and the Caulerpales.  相似文献

Introduction

Ultrasonography (US) might have an added value to clinical examination in diagnosing early rheumatoid arthritis (RA) and assessing remission of RA. We aimed to clarify the added value of US in RA in these situations performing a systematic review.

Methods

A systematic literature search was performed for RA, US, diagnosis and remission. Methodological quality was assessed; the wide variability in the design of studies prohibited pooling of results.

Results

Six papers on the added value of US diagnosing early RA were found, in which at least bilateral metacarpophalangeal (MCP), wrists and metatarsophalangeal (MTP) joints were scanned. Compared to clinical examination, US was superior with regard to detecting synovitis and predicting progression to persistent arthritis or RA. Eleven papers on assessing remission were identified, in which at least the wrist and the MCP joints of the dominant hand were scanned. Often US detected inflammation in patients clinically in remission, irrespective of the remission criteria used. Power Doppler signs of synovitis predicted X-ray progression and future flare in patients clinically in remission.

Conclusions

US appears to have added value to clinical examination for diagnosing of RA when scanning at least MCP, wrist and MTP joints, and, when evaluating remission of RA, scanning at least wrist and MCP joints of the dominant hand. For both purposes primarily power Doppler US might be used since its results are less equivocal than those of greyscale US.  相似文献

The class Eustigmatophyceae includes mostly coccoid, freshwater algae, although some genera are common in terrestrial habitats and two are primarily marine. The formal classification of the class, developed decades ago, does not fit the diversity and phylogeny of the group as presently known and is in urgent need of revision. This study concerns a clade informally known as the Pseudellipsoidion group of the order Eustigmatales, which was initially known to comprise seven strains with oval to ellipsoidal cells, some bearing a stipe. We examined those strains as well as 10 new ones and obtained 18S rDNA and rbcL gene sequences. The results from phylogenetic analyses of the sequence data were integrated with morphological data of vegetative and motile cells. Monophyly of the Pseudellipsoidion group is supported in both 18S rDNA and rbcL trees. The group is formalized as the new family Neomonodaceae comprising, in addition to Pseudellipsoidion, three newly erected genera. By establishing Neomonodus gen. nov. (with type species Neomonodus ovalis comb. nov.), we finally resolve the intricate taxonomic history of a species originally described as Monodus ovalis and later moved to the genera Characiopsis and Pseudocharaciopsis. Characiopsiella gen. nov. (with the type species Characiopsiella minima comb. nov.) and Munda gen. nov. (with the type species Munda aquilonaris) are established to accommodate additional representatives of the polyphyletic genus Characiopsis. A morphological feature common to all examined Neomonodaceae is the absence of a pyrenoid in the chloroplasts, which discriminates them from other morphologically similar yet unrelated eustigmatophytes (including other Characiopsis-like species).  相似文献

Current hypotheses on the distribution of freshwater microchlorophytes lead to predictions of low diversity and wide environmental tolerances. Thus, the same few species should be found worldwide in many different habitats. However, these hypotheses are based on a morphospecies concept, which precludes the possibility of numerous cryptic species among these organisms. In this study, we examined the diversity of coccoid green microalgae and chlamydomonads (Chlorophyta) isolated from sites in Minnesota and North Dakota (USA) using techniques of 18S rDNA sequence analysis. Of 93 distinct 18S rDNA sequences identified from among 273 isolates examined by molecular techniques, all but four are new to science. The spatial distribution of organisms represented by these 18S rDNA sequences was not uniform, because some lakes and ponds yielded distinct 18S rDNA types not found at other sites. In addition, organisms generally considered to be cosmopolitan, such as Chlamydomonas reinhardtii and Chlorella vulgaris, were not found. These results challenge predictions of low species number and wide environmental tolerances among these eukaryotic microorganisms.  相似文献

Many shallow lakes in north temperate zones experience reduced dissolved oxygen concentration under ice. However, some shallow lakes display supersaturated dissolved oxygen concentrations (>20 mg·L ? 1) in late winter under conditions of maximum ice thickness. During the winters of 1996, 1997, and 1999, we collected phytoplankton samples from Arrowwood Lake near Pingree, North Dakota to determine whether a specific alga was involved in dissolved oxygen supersaturation in this lake. Although dissolved oxygen supersaturation was not observed during this period, we did observe an increase in dissolved oxygen concentration that was associated with a phytoplankton bloom during late February and early March in both 1996 and 1997. In 1996, the bloom was composed of the dinoflagellate, Peridinium aciculiferum (Lemm.) Lemm. and several species of cryptomonads. A similar bloom of P. aciculiferum was followed by a bloom of several species of euglenoids in 1997. In contrast, P. aciculiferum was only a minor component of the winter phytoplankton, dissolved oxygen concentrations remained low, and no bloom event was observed in 1999. Statistical analyses indicated a significant relationship (rs = 0.57, P = 0.019) between dissolved oxygen levels and the density of the dinoflagellate, P. aciculiferum, but no significant relationship between dissolved oxygen levels and densities of other phytoplankton. These results suggest that the elevated levels of dissolved oxygen are associated with the dinoflagellate, P. aciculiferum. This bloom was most likely the result of an excystment event rather than a general growth response.  相似文献

The general model of seasonal phytoplankton succession in temperate lakes suggests that winter phytoplankton growth is minimal under ice-cover. However, some studies have found diverse phytoplankton communities during winter. The primary objectives of this study were to determine the species composition and the changes in the winter phytoplankton community structure under the ice. For 2 consecutive winters, phytoplankton samples were collected under ice-cover at 4 sites on 3 lakes in Arrowwood National Wildlife Refuge (ANWR), near Pingree, North Dakota. Ninety taxa were identified and enumerated. Densities of several of these taxa frequently exceeded 106 cells l–1. The winter phytoplankton communities in these lakes were dominated by flagellates, principally cryptomonads, a synurophyte (Synura uvella), small chrysophytes (Chrysococcus spp., Kephyrion spp.) and a dinoflagellate (Peridinium aciculiferum), as well as non-flagellate microchlorophytes (Monoraphidium spp., Ankistrodesmus spp., and Pseudodictyosphaerium sp.), a cyanobacterium (Gloeocapsa aeruginosa) and centric diatoms (Stephanodiscus minutulus, S. parvus and Cyclotella meneghiniana).  相似文献

The Sheyenne River is a third order stream in central and eastern North Dakota. The algae in this river had not been previously studied. For this study, periphytic algal samples were collected Fall 1997, April–November 1998, and May–October 1999. Two hundred sixteen periphytic algal taxa were identified. These taxa included 137 Bacillariophyceae, 33 Cyanophyta, 40 Chlorophyta, 8 Xanthophyceae, and 3 Euglenophyta. Several species of algae found in the river appear to be previously undescribed taxa. Two of these organisms were described as new species, Nitzschia sheyennensis and Nitzschia dakotensis (Bacillariophyceae). The distribution of the periphytic algae was correlated to changes in the selected environmental variables using canonical correspondence analysis (CCA) and mult-dimensional scaling (MDS). The statistically significant environmental variables were nitrite-nitrate, pH, arsenic, potassium, magnesium, manganese, and total dissolved solids. A reservoir, Lake Ashtabula on the river was formed by a dam in 1954. In the initial statistical analysis, the effects of this reservoir on the periphytic algae downstream were so great that an upstream/downstream covariable had to be used. Further statistical analysis was used to determined that this effect could be correlated to changes in the nutrient levels rather than hydrologic changes.  相似文献

Choricystis (Trebouxiphyceae, Chlorophyta) is considered a very common member of the freshwater picoplankton. We have established a culture collection from Itasca State Park (ISP), Minnesota, and Arrowwood National Wildlife Refuge (ANWR), North Dakota, that includes many Choricystis isolates. We examined 109 Choricystis isolates from ISP and 19 isolates from ANWR using PCR‐RFLP of the rbcL gene. Twelve types for ISP and 7 types forANWR were distinguished by this technique. Sequence analysis revealed additional diversity within some of the RFLP types. Moreover, none of the Choricystis isolates from ANWR possessed rbcL sequences identical to any isolate from ISP. Phylogenetic analysis of the sequence data revealed at least 2 major lineages. These results indicate that Choricystis is much more diverse than previously thought.
