This work describes a simple and reproducible method of determining the fructose content of sperm in order to assess the impregnation of the genital tract with testosterone in men in whom spermatogenesis is blocked by a combination of progesterone and androgens. The colorimetric method of Karvonen and Malm was used with certain modification. The fructose content of sperm is colorimetrically determined by the indol in a concentrated chlorhydric milieu, after deproteinization by cadmium hydroxide. Instructions are given for preparation of the aqueous solution of indol, the sulfuric solution of cadmium sulfate and the fructose solution, as well as for the procedure itself and calculation of the fructose concentration. The method was designed to monitor men taking 20 mg/daily of medroxyprogesterone acetate and applying testosterone daily to the torso. The progesterone inhibits spermatogenesis in inhibiting follicle stimulating hormone and luteinizing hormone and also blocks testosterone secretion by the testicles. The addition of testosterone is designed to maintain a normal level of testosterone to avoid feminization.  相似文献

We discuss the statistics of spikes trains for different types of integrate-and-fire neurons and different types of synaptic noise models. In contrast with the usual approaches in neuroscience, mainly based on statistical physics methods such as the Fokker-Planck equation or the mean-field theory, we chose the point of the view of the stochastic calculus theory to characterize neurons in noisy environments. We present four stochastic calculus techniques that can be used to find the probability distributions attached to the spikes trains. We illustrate the power of these techniques for four types of widely used neuron models. Despite the fact that these techniques are mathematically intricate we believe that they can be useful for answering questions in neuroscience that naturally arise from the variability of neuronal activity. For each technique we indicate its range of applicability and its limitations.  相似文献

This special issue of the Journal of Physiology, Paris, is an outcome of NeuroComp'06, the first French conference in Computational Neuroscience. The preparation for this conference, held at Pont-à-Mousson in October 2006, was accompanied by a survey which has resulted in an up-to-date inventory of human resources and labs in France concerned with this emerging new field of research (see team directory in http://neurocomp.risc.cnrs.fr/). This thematic JPP issue gathers some of the key scientific presentations made on the occasion of this first interdisciplinary meeting, which should soon become recognized as a yearly national conference representative of a new scientific community. The present introductory paper presents the general scientific context of the conference and reviews some of the historical and conceptual foundations of Systems and Computational Neuroscience in France.  相似文献

Background  

Neutrophils infiltrate the endometrium pre-menstrually and after long-term progestin only-contraceptive (LTPOC) treatment. Trafficking of neutrophils involves endothelial cell-expressed intercellular adhesion molecule (ICAM-1). Previous studies observed that ICAM-1 was immunolocalized to the endothelium of endometrial specimens across the menstrual cycle, but disagreed as to whether extra-endothelial cell types express ICAM-1 and whether ICAM-1 expression varies across the menstrual cycle.  相似文献

A project of QTL detection was carried out in the French Holstein, Normande, and Montbéliarde dairy cattle breeds. This granddaughter design included 1 548 artificial insemination bulls distributed in 14 sire families and evaluated after a progeny-test for 24 traits (production, milk composition, persistency, type, fertility, mastitis resistance, and milking ease). These bulls were also genotyped for 169 genetic markers, mostly microsatellites. The QTL were analysed by within-sire linear regression of daughter yield deviations or deregressed proofs on the probability that the son receives one or the other paternal QTL allele, given the marker information. QTL were detected for all traits, including those with a low heritability. One hundred and twenty QTL with a chromosome-wise significance lower than 3% were tabulated. This threshold corresponded to a 15% false discovery rate. Amongst them, 32 were genome-wise significant. Estimates of their contribution to genetic variance ranged from 6 to 40%. Most substitution effects ranged from 0.6 to 1.0 genetic standard deviation. For a given QTL, only 1 to 5 families out of 14 were informative. The confidence intervals of the QTL locations were large and always greater than 20 cM. This experiment confirmed several already published QTL but most of them were original, particularly for non-production traits.  相似文献

The human endometrium is an important site of innate immune defence, giving protection against uterine infection. Such protection is critical to successful implantation and pregnancy. Infection is a major cause of preterm birth and can also cause infertility and ectopic pregnancy. Natural anti-microbial peptides are key mediators of the innate immune system. These peptides, between them, have anti-bacterial, anti-fungal and anti-viral activity and are expressed at epithelial surfaces throughout the female genital tract. Two families of natural anti-microbials, the defensins and the whey acidic protein (WAP) motif proteins, appear to be prominent in endometrium. The human endometrial epithelium expresses beta-defensins 1–4 and the WAP motif protein, secretory leukocyte protease inhibitor. Each beta-defensin has a different expression profile in relation to the stage of the menstrual cycle, providing potential protection throughout the cycle. Secretory leukocyte protease inhibitor is expressed during the secretory phase of the cycle and has a range of possible roles including anti-protease and anti-microbial activity as well as having effects on epithelial cell growth. The leukocyte populations in the endometrium are also a source of anti-microbial production. Neutrophils are a particularly rich source of alpha-defensins, lactoferrin, lysozyme and the WAP motif protein, elafin. The presence of neutrophils during menstruation will enhance anti-microbial protection at a time when the epithelial barrier is disrupted. Several other anti-microbials including the natural killer cell product, granulysin, are likely to have a role in endometrium. The sequential production of natural anti-microbial peptides by the endometrium throughout the menstrual cycle and at other sites in the female genital tract will offer protection from many pathogens, including those that are sexually transmitted.  相似文献

A computational study into the motion perception dynamics of a multistable psychophysics stimulus is presented. A diagonally drifting grating viewed through a square aperture is perceived as moving in the actual grating direction or in line with the aperture edges (horizontally or vertically). The different percepts are the product of interplay between ambiguous contour cues and specific terminator cues. We present a dynamical model of motion integration that performs direction selection for such a stimulus and link the different percepts to coexisting steady states of the underlying equations. We apply the powerful tools of bifurcation analysis and numerical continuation to study changes to the model’s solution structure under the variation of parameters. Indeed, we apply these tools in a systematic way, taking into account biological and mathematical constraints, in order to fix model parameters. A region of parameter space is identified for which the model reproduces the qualitative behaviour observed in experiments. The temporal dynamics of motion integration are studied within this region; specifically, the effect of varying the stimulus gain is studied, which allows for qualitative predictions to be made.  相似文献
