Degradation of elastin leads to the production of elastin-derived peptides (EDP), which exhibit several biological effects, such as cell proliferation or protease secretion. Binding of EDP on the elastin receptor complex (ERC) triggers lactosylceramide (LacCer) production and ERK1/2 activation following ERC Neu-1 subunit activation. The ability for ERC to transduce signals is lost during aging, but the mechanism involved is still unknown. In this study, we characterized an in vitro model of aging by subculturing human dermal fibroblasts. This model was used to understand the loss of EDP biological activities during aging. Our results show that ERC uncoupling does not rely on Neu-1 or PPCA mRNA or protein level changes. Furthermore, we observe that the membrane targeting of these subunits is not affected with aging. However, we evidence that Neu-1 activity and LacCer production are altered. Basal Neu-1 catalytic activity is strongly increased in aged cells. Consequently, EDP fail to promote Neu-1 catalytic activity and LacCer production in these cells. In conclusion, we propose, for the first time, an explanation for ERC uncoupling based on the age-related alterations of Neu-1 activity and LacCer production that may explain the loss of EDP-mediated effects occurring during aging.  相似文献

Polyphenisms—the expression of discrete phenotypic morphs in response to environmental variation—are examples of phenotypic plasticity that may potentially be adaptive in the face of predictable environmental heterogeneity. In the butterfly Bicyclus anynana, we examine the hormonal regulation of phenotypic plasticity that involves divergent developmental trajectories into distinct adult morphs for a suite of traits as an adaptation to contrasting seasonal environments. This polyphenism is induced by temperature during development and mediated by ecdysteroid hormones. We reared larvae at separate temperatures spanning the natural range of seasonal environments and measured reaction norms for ecdysteroids, juvenile hormones (JHs) and adult fitness traits. Timing of peak ecdysteroid, but not JH titres, showed a binary response to the linear temperature gradient. Several adult traits (e.g. relative abdomen mass) responded in a similar, dimorphic manner, while others (e.g. wing pattern) showed a linear response. This study demonstrates that hormone dynamics can translate a linear environmental gradient into a discrete signal and, thus, that polyphenic differences between adult morphs can already be programmed at the stage of hormone signalling during development. The range of phenotypic responses observed within the suite of traits indicates both shared regulation and independent, trait-specific sensitivity to the hormone signal.  相似文献

Abstract Cyclic AMP-induced cAMP and cGMP responses during development of Dictyostelium discoideum were investigated. The cAMP-induced cGMP response is maximal when aggregation is in full progress, and then decreases to about 10% of the maximal level during further multicellular development. The cAMP response increases upon starvation, reaches its maximum at the onset of aggregation, and then decreases to about 8% of the maximum level. The dynamics of the post-aggregative cAMP response are in qualitative agreement with the dynamics of the cAMP relay response in aggregation-competent cells.  相似文献

Surveys and monitoring of 37 caves in and around the Ankarana Special Reserve, northern Madagascar, yielded evidence of hunting of bats and potential disturbance of bats by miners and tourists, and colony counts for several bat species of potential conservation concern. Colony size decreased by 95% and 14% for a colony of Hipposideros commersoni and a colony of Eidolon dupreanum, respectively, when recent evidence of hunting occurred at those colonies and those declines are probably attributable to hunting. Evidence of hunting occurred commonly at the roosts of those species and most commonly at the roosts of Rousettus madagascariensis. Hunting of pteropodids was associated with high vulnerability of roosts to hunters, little forest buffer between the cave and open savannah and the absence of tombs in the cave. Roost sites of the hunted species persisted for at least several years and this regularity may facilitate hunting. This work supports the ranking of E. dupreanum, R. madagascariensis and H. commersoni as species of conservation concern. Managers should consider the impact of tourist visits on bats and of increasing access to caves for tourism. Conservation efforts for the hunted species should also seek to protect vulnerable and unprotected cave roosts.  相似文献

Summary Goldfish, acclimated to 20°C and normal = 130 mmHg) and low ( = 19 mmHg) oxygen levels, were exposed to different periods of hypoxia and anoxia. Experiments were carried out at night. ATP, ADP, AMP, IMP, CrP, glycogen and lactate were determined in red muscle, white muscle and liver. Acclimation to hypoxia resulted in a marked increase of the energy charge of liver and red muscle and of the glycogen content of red and white muscle, indicating an increased anaerobic capacity. Short exposures to anoxia, up to 1 h, had little influence on the value of the measured parameters. Long-term exposures (12 h) to anoxia caused a significant decrease of CrP and glycogen levels in all tissues examined. The energy-charge of red and white muscle was hardly affected by a 12 h exposure to anoxia, but in liver tissue the energy charge decreased from 0.60 to 0.32. It is concluded that during anoxia muscle tissues are able to maintain high energy-charges, probably by means of a yet unknown anaerobic energy-producing system.Abbreviations CrP creatine phosphate - EC energy charge - IMP inosine monophosphate - I.U. Abstract

The genus Coleura contains two species: C. seychellensis restricted to the Seychelles Archipelago and C. afra broadly distributed on the African continent and the Arabian Peninsula. We recently captured and collected specimens of Coleura from Madagascar, an island from which it was previously unknown. Using museum specimens and the new material from Madagascar, we review morphological variation in members of this genus and present characters for species identification. On the basis of these comparisons, specimens from Madagascar are referable to C. afra. We present information on the natural history of the recently discovered Malagasy population.  相似文献

Although the land mammals of Madagascar have been the subject of many studies, the island’s bats have yet to feature prominently on the research or conservation agenda. In this study we used mist nets, acoustic sampling and cave surveys to assess habitat use, seasonality and roost selection. Four microchiropteran species (Triaenops rufus, T. furculus, Miniopterus manavi and Myotis goudoti) appeared to be strongly associated with the forest interior based on trapping, but analysis of time-expanded echolocation recordings revealed that T. rufus and M.␣manavi were frequently recorded in forest edges and clearings. Bat activity was significantly lower inside the forest than at the interface between agricultural land and forest. The caves visited most often by tourists were low in bat abundance and species richness. Anjohikinakina Cave, which was visited infrequently by people, was used by five species and contained between 54% (winter) and 99% (summer) of bats counted in 16 caves and is a site of national importance for bat conservation. Hipposideros commersoni was only netted in our study area during October and may be a migrant to the site or present but inactive during the austral winter. The forest surrounding the caves is therefore important because it provides cover for emerging bats and a potential source of invertebrate prey whilst the forest edge is important to foraging bats.  相似文献

Using measures of gas exchange and photosynthetic chain activity, we found some differences between grapevine inflorescence and leaf in terms of photosynthetic activity and photosynthesis regulations. Generally, the leaf showed the higher net photosynthesis (P N) and lower dark respiration than that of the inflorescence until the beginning of the flowering process. The lower (and negative) P N indicated prevailing respiration over photosynthesis and could result from a higher metabolic activity rather than from a lower activity of the photosynthetic apparatus. Considerable differences were observed between both organs in the functioning and regulation of PSI and PSII. Indeed, in our conditions, the quantum yield efficiency and electron transport rate of PSI and PSII were higher in the inflorescence compared to that of the leaf; nevertheless, protective regulatory mechanisms of the photosynthetic chain were clearly more efficient in the leaf. This was in accordance with the major function of this organ in grapevine, but it highlighted also that inflorescence seems to be implied in the whole carbon balance of plant. During inflorescence development, the global PSII activity decreased and PSI regulation tended to be similar to the leaf, where photosynthetic activity and regulations remained more stable. Finally, during flowering, cyclic electron flow (CEF) around PSI was activated in parallel to the decline in the thylakoid linear electron flow. Inflorescence CEF was double compared to the leaf; it might contribute to photoprotection, could promote ATP synthesis and the recovery of PSII.  相似文献
