The pathogenicity of three isolates of Beauveria bassiana (Bals.) Vuil. and one isolate of Metarhizium anisopliae (Metsch.) Sorok. (Deuteromycotina: Hyphomycetes) was assessed in the laboratory against adults of the leaf-footed bug Leptoglossus zonatus (Dallas) (Heteroptera: Coreidae) and the shield-backed bug Pachycoris klugii Burmeister (Heteroptera: Scutelleridae), the two most frequent pest species in physic nut ( Jatropha curcas L., Euphorbiaceae) plantations in Nicaragua. In a dipping bioassay, the median lethal concentration (LC) of the most efficient strain, M. anisopliae NB, was determined as 4.34 106 50 conidia/ml for adult P. klugii . In a field trial, a scheduled high-volume spray regime using B. bassiana increased fruit yield by 28%, and was more effective than malathion or an aqueous extract of ground neem seeds. The effectiveness of M. anisopliae was further tested in field cages covering entire trees and containing a predetermined number of insects. Mineral oilbased ultra-low volume controlled droplet applications of M. anisopliae at a rate of 1 1010 conidia/tree were made using hand-held Micron ULVA + sprayers. Tetrahydrofuran/toluene solutions of (nacnac)TiCl2 (nacnac− = [ArNC(tBu)]2CH, Ar = 2,6-iPr2C6H3) react readily with KC8 to afford the titanium imide (ArN(tBu)CCHC(tBu))TiNAr(THF)Cl (1) in 67% isolated yield. Complex 1 forms from the two-electron reductive C-N bond cleavage of the β-diketiminate ligand. Likewise, reduction of (nacnac)TiCl(NHAr) (2), prepared in 85% yield from (nacnac)TiCl2 and LiNHAr, with KC8 results in formation of the imide-anilide analogue (ArN(tBu)CCHC(tBu))TiNAr(NHAr) (3) in 88% yield. Another reductant such as LitBu (3 equiv.) reacts cleanly with the precursor (nacnac)TiCl2 to afford the alkylidene-ate complex [Li(Et2O)][(ArN(tBu)CCHC(tBu))TiNAr(Et2O)] (4), in 81% yield. Complexes 1-4 have been characterized by 1H and 13C NMR spectra as well as single-crystal X-ray diffraction analysis. Mannitol is a naturally occurring low calorie sweetener, widely used in the food, pharmaceutical, medicine and chemical industries.
In this study mannitol producing strains of Leuconostoc spp. (210) were isolated from a wide array of sources such as raw milk, fermented milks, fermented cereal foods, fruits,
vegetables and sugar factory syrup. During initial screening, half of the population of these isolates (105) exhibited ability
to produce mannitol to a variable extent. Only 11.4% isolate produced mannitol yield of above 80% (when fructose used @ 50 g/l).
Cultural and environmental factors affecting growth and mannitol production were studied for four high mannitol producing
isolates. High mannitol production was favored by high temperature and high pH. Isolates had high osmotic tolerance as these
could use fructose concentration as high as 100 g/l in batch culture. Sequencing of 16S rRNA genes of the strains revealed
that Ln27, Ln104 and Ln206 were Leuconostoc mesenteroides and Ln92 was Leuconostoc fallax. 相似文献

The majority of bacteria isolated from rhizospheres of Arachis hypogea (Groundnut) and Vigna radiata (Mung bean) predominantly produced catechol-type siderophores except for a few fluorescent pseudomonads that produced hydroxamates
in addition to catecholates. The rhizospheric isolates differed in their ability to cross-utilize siderophores produced by
other rhizospheric isolates (heterologous); some were highly proficient at utilizing heterologous siderophores, while others
were poor cross-utilizers. Isolate G9, which utilized hydroxamate as well as catecholate siderophores, was found to be an
efficient siderophore cross-utilizer, while isolates G2 and G6 were poor-utilizers of catecholate and non-utilizers of hydroxamate
siderophores. Growth stimulation of two isolates G9 and G6 was seen when grown in the presence of externally supplied heterologous
siderophores, which they cross-utilized. The iron-regulated outer membrane protein (IROMP) profiles differed for the most
cross-utilizer and the least cross-utilizer strains, but in both the cases no new outer membrane proteins (OMP) were induced
in response to the exogenous siderophores supplied. The growth of the organisms in the presence of heterologous siderophores
that they failed to cross-utilize led to growth inhibition in the case of isolate G9. This appears to be due to a lower affinity
of the siderophore of G9 as compared to the exogenously supplied G6 siderophore. A simple method was devised to measure relative
affinities of respective siderophores for iron based on CAS solution decolorization by the siderophore preparations. The effect
on the growth of the differential affinities of the siderophores for iron and the interactions of the organisms through cross-utilization

Three unsprayed coffee farms (farm 1, 2 and 3) were studied for the natural occurrence of the insect pathogenic fungus Beauveria bassiana in Hypothenemus hampei populations throughout the rainy season of 2004 (July-November) and 2005 (July-December). B. bassiana infections were found during most sampling dates in both years, on all three farms. The B. bassiana infection levels were higher in 2005 than in 2004 with mean prevalence of 12.1% and 2.7%, respectively. No consistent significant differences in infection level between farms were found in any of the years. B. bassiana infection levels fluctuated widely throughout the season, and peaked at 13.5% on farm 3 in 2004 and at 44.0% on farm 1 in 2005. The H. hampei population was significantly higher in 2004 than in 2005, with 6.9% of the berries infested in 2004 and only 0.7% in 2005. In both years, the H. hampei infestation level was significantly higher on farm 2. No consistent significant differences in H. hampei infestation levels were found between sampling dates on any of the farms. H. hampei infestation levels fluctuated throughout both seasons, and peaked at 15.3% on farm 2 in 2004 and 2.2% on farm 2 in 2005. No consistent density dependent correlation between H. hampei infestation level and B. bassiana infection level was found. Endophytic bacteria which are known to reside in plant tissues have often been shown to promote plant growth. Present study deals with the isolation of putative endophytes from the surface sterilized root nodules of pigeon pea (Cajanus cajan) designated as non-rhizobial (NR) isolates. Three of these non-rhizobial isolates called NR2, NR4 and NR6 showed plant growth promotion with respect to increase in plant fresh weight, chlorophyll content, nodule number and nodule fresh weight when co-inoculated with the rhizobial bioinoculant strain IC3123. The three isolates were neither able to nodulate C. cajan nor did they show significant plant growth promotion when inoculated alone without Rhizobium spp. IC3123. All the three isolates were gram positive rods with NR2 and NR4 showing endospore formation and formed one single cluster in Amplified Ribosomal DNA Restriction Analysis (ARDRA). Partial sequences of 16S rRNA genes of NR4 and NR6 showed 97% similarity to Bacillus megaterium. The Bacillus strains NR4 and NR6 were able to produce siderophores which the rhizobial bioinoculant IC3123 was able to cross-utilize. Under iron starved conditions IC3123 showed enhanced growth in the presence of the Bacillus isolates indicating that siderophore mediated interactions may be underlying mechanism of beneficial effect of the NR isolates on nodulation by IC3123. 相似文献

Hyperglycemia induces a wide array of signaling pathways in the kidney that lead to hypertrophy and matrix expansion, eventually culminating in progressive kidney failure. High glucose-induced reduction of the tumor suppressor protein phosphatase and tensin homolog deleted in chromosome 10 (PTEN) contributes to renal cell hypertrophy and matrix expansion. We identified microRNA-21 (miR-21) as the molecular link between high glucose and PTEN suppression. Renal cortices from OVE26 type 1 diabetic mice showed significantly elevated levels of miR-21 associated with reduced PTEN and increased fibronectin content. In renal mesangial cells, high glucose increased the expression of miR-21, which targeted the 3'-UTR of PTEN mRNA to inhibit PTEN protein expression. Overexpression of miR-21 mimicked the action of high glucose, which included a reduction in PTEN expression and a concomitant increase in Akt phosphorylation. In contrast, expression of miR-21 Sponge, to inhibit endogenous miR-21, prevented down-regulation of PTEN and phosphorylation of Akt induced by high glucose. Interestingly, high glucose-stimulated miR-21 inactivated PRAS40, a negative regulator of TORC1. Finally, miR-21 enhanced high glucose-induced TORC1 activity, resulting in renal cell hypertrophy and fibronectin expression. Thus, our results identify a previously unrecognized function of miR-21 that is the reciprocal regulation of PTEN levels and Akt/TORC1 activity that mediate critical pathologic features of diabetic kidney disease. 相似文献

Nitrogen fixation, the biological reduction of dinitrogen gas (N2) to ammonium (NH4+), is quantitatively the most important external source of new nitrogen (N) to the open ocean. Classically, the ecological niche of oceanic N2 fixers (diazotrophs) is ascribed to tropical oligotrophic surface waters, often depleted in fixed N, with a diazotrophic community dominated by cyanobacteria. Although this applies for large areas of the ocean, biogeochemical models and phylogenetic studies suggest that the oceanic diazotrophic niche may be much broader than previously considered, resulting in major implications for the global N-budget. Here, we report on the composition, distribution and abundance of nifH, the functional gene marker for N2 fixation. Our results show the presence of eight clades of diazotrophs in the oxygen minimum zone (OMZ) off Peru. Although proteobacterial clades dominated overall, two clusters affiliated to spirochaeta and archaea were identified. N2 fixation was detected within OMZ waters and was stimulated by the addition of organic carbon sources supporting the view that non-phototrophic diazotrophs were actively fixing dinitrogen. The observed co-occurrence of key functional genes for N2 fixation, nitrification, anammox and denitrification suggests that a close spatial coupling of N-input and N-loss processes exists in the OMZ off Peru. The wide distribution of diazotrophs throughout the water column adds to the emerging view that the habitat of marine diazotrophs can be extended to low oxygen/high nitrate areas. Furthermore, our statistical analysis suggests that NO2− and PO43− are the major factors affecting diazotrophic distribution throughout the OMZ. In view of the predicted increase in ocean deoxygenation resulting from global warming, our findings indicate that the importance of OMZs as niches for N2 fixation may increase in the future. 相似文献

Two Brucella abortus S19 commercial vaccine strains used for vaccination against brucellosis in India and three S19 strains available as international reference were examined by microbiological assays and molecular analysis of the ery loci involved in erythritol metabolism, and tested for residual virulence in BALB/c mice. According to the sensitivity to penicillin and i-erythritol, the five strains tested had the phenotypic characteristics of strain S19. However, on culture medium containing i-erythritol, all strains developed spontaneous i-erythritol resistant colonies at mutation rates ranging from 1.42x10(-2) to 1.33x10(-6). The S19 characteristic 702 bp deletion in the erythrulose 1-phosphate dehydrogenase gene of the ery locus was present only in the three reference strains but not in the two commercial vaccines. Both commercial strains and one of the reference strains showed reduced virulence in BALB/c mice. The presence or absence in S19 strains of the 702 bp deletion in the ery locus had no correlation with either the rates of spontaneous mutation to erythritol resistance or the residual virulence in mice. 相似文献
