Unique Pattern of Gene Expression in the Erythroid Precursor Cells

Erythroid cells were fractionated by preformed Percoll density gradient from livers of 12.5 day old mouse fetuses. With combination of lysing of mature erythroid cells, the CFU-E (colony forming unit of erythroid) was enriched as high as 30% pure. The mRNA levels of the rt-genes previously cloned as genes expressed in the reticulocytes are estimated in the fractionated erythroid cells. These rt-genes show a drastic change in expression during erythroid differentiation; Their expression was not detectable at the CFU-E cell stage. But it reached to maximum at the polychromatic erythroblast (stage I) and then decreases with maturation. The result suggests that mRNA synthesis of these rt-genes may be induced after the stimulation of erythropoietin.     

阳性脂质体介导反义寡聚核苷酸转染HeLa细胞的效果研究

采用FITC标记的未经修饰的和经过修饰的两种19-mer反义寡聚核苷酸序列(ODN19和S-ODN19)作为转染物质,用流式细胞技术(FCM)研究比较几种常用阳性脂质体介导的寡聚核苷酸转染HeLa细胞的效果及适宜的转染时间。未经化学修饰的ODN19转染结果显示,LipofectAmine和DM-RIE-C增强转染的作用相对较强,而其他两种脂质体的作用并不明显。对于经过修饰的S-ODN19转染而言,四种阳性脂质体均具有增强S-ODN19转染作用,但以LipofectAmine的效果最为明显,其转染效果(FITC均值为5203.11)为无脂质体介导对照的数十倍。四种阳性脂质体的增强转染作用排序为:LipofectAmine>FuGENE6>Lipofectin>DM-RIR-C。另外,在用FuGENE6介导寡聚核苷酸转染时,采用4小时转染时间可获较好转染效果。     

Macrochelid mite fauna in the eastern part of the Lesser Sunda Islands, with description of two new species

Thirteen species in four genera of mites of the family Macrochelidae phoretic on dung beetles were collected in the eastern part of the Lesser Sunda Islands, Indonesia, providing the first record of the family for that area. Of these, two species, Macrocheles entetiensis Hartini and Takaku, sp. nov. and Macrocheles sumbaensis Hartini and Takaku, sp. nov., are new to science. The remaining 11 species are Glyptholaspis fimicola, Holostaspella bifoliata, Macrocheles baliensis, Macrocheles sp. aff. glaber, M. hallidayi, M. kraepelini, M. krantzi, M. limue, M. merdarius, M. oigru and Neopodocinum sinicum.     

Mites of family Macrochelidae (Acari: Gamasida) associated with dung beetles in Mt Merapi National Park,Jogyakarta, Java,Indonesia

Fourteen mite species in three genera of the family Macrochelidae were collected from the body surface of dung beetles (Scarabaeidae) in Mt Merapi National Park, Jogyakarta, Indonesia. Of these, three species, Macrocheles turgoensis n. sp., M. pumilus n. sp. and Glyptholaspis merapiensis n. sp., were described as new to science. The remaining 11 species were Glyptholaspis fimicola (Sellnick, 1931), Neopodocinum spinirostris (Berlese, 1910), M. dispar (Berlese, 1910), M. entetiensis Hartini and Takaku, 2005, M. hallidayi Walter and Krantz, 1986, M. jabarensis Hartini and Takaku, 2003, M. merdarius (Berlese, 1889), M. muscaedomesticae (Scopoli, 1772), M. oigru Walter and Krantz, 1986, M. sukaramiensis Takaku, 2001 and M. sp. aff. glaber (Müller, 1860).     

Mites of the genus Holostaspella (Acari: Gamasida: Macrochelidae) in Indonesia

The genus Holostaspella of the family Macrochelidae consists of more than 30 species. Of these, seven species have been recorded from Indonesia. In the present study, we review the genus in Indonesia, record two species ( H. pulchella and H. similiornata ) for the first time from Indonesia, and describe two new species ( H. oblonga n. sp. and H. villosa n. sp.) on the basis of the specimens collected in Java and Sulawesi. A key to the species of the genus in Indonesia is also provided.
