Current advances in genetic analysis are opening up our knowledge of the genetics of species differences, but challenges remain, particularly for out‐bred natural populations. We constructed a microsatellite‐based linkage map for two out‐bred lines of Drosophila montana derived from divergent populations by taking advantage of the Drosophila virilis genome and available cytological maps of both species. Although the placement of markers was quite consistent with cytological predictions, the map indicated large heterogeneity in recombination rates along chromosomes. We also performed a quantitative trait locus (QTL) analysis on a courtship song character (carrier frequency), which differs between populations and is subject to strong sexual selection. Linkage mapping yielded two significant QTLs, which explained 3% and 14% of the variation in carrier frequency, respectively. Interestingly, as in other recent studies of traits which can influence speciation, the strongest QTL mapped to a genomic region partly covered by an inversion polymorphism.  相似文献

Abstract: We tested the hypothesis that piping plover (Charadrius melodus) habitat quality and chick survival on the Missouri River, USA, were lower on a cold-water reservoir and downstream from a hypolimnetic (cold-water) release dam with diel water fluctuations (Garrison Dam) than downstream from an epilimnetic dam (Gavins Point Dam). Plovers in adjacent alkali wetlands provided an index to the maximum reproductive potential in the region. Chicks gained weight more rapidly in the alkali wetlands than on epilimnetic and hypolimnetic river reaches. Invertebrate numbers and biomass were higher in the wetlands and epilimnetic reach, but chick survival was lower on the epilimnetic reach. Thus, piping plovers adapted to a variety of prey densities, and other factors, likely predation, reduced survival rates in the epilimnetic reach. Temporal and spatial variability in site quality indices suggests the need for a regional management strategy with different strategies at each site. Managers can minimize effects of local fluctuations in resource abundance and predators by ensuring protection of or creating geographically dispersed habitat. 1. Two senior ecologists summarised their experience of the scientific publication process ( Statzner & Resh, Freshwater Biology, 2010 ; 55 , 2639) to generate discussion, particularly among early career researchers (ECRs). As a group of eight ECRs, we comment on the six trends they described. 2. We generally agree with most of the trends identified by Statzner & Resh (2010) , but also highlight a number of divergent perspectives and provide recommendations for change. Trends of particular concern are the use of inappropriate metrics to evaluate research quality (e.g. impact factor) and the salami slicing of papers to increase paper count. We advocate a transparent and comprehensive system for evaluating the research. 3. We stress the importance of impartiality and independence in the peer review process. We therefore suggest implementation of double‐blind review and quality control measures for reviewers and possibly editors. Besides such structural changes, editors should be confident to overrule biased reviewer recommendations, while reviewers should provide helpful reviews but be explicit if a submission does not meet quality standards. Authors should always conduct a thorough literature search and acknowledge historical scientific ideas and methods. Additionally, authors should report low‐quality copy editing and reviews to the editors. 4. Both early and late career researchers should jointly implement these recommendations to reverse the negative trends identified by Statzner & Resh (2010) . However, more senior scientists will always have to take the lead with respect to structural changes in the publication system given that they occupy the majority of decision‐making positions.  相似文献

Abstract. We recently discussed a method for measuring optical properties of light scattering and absorbing plant tissue ( Seyfried, Fukshansky & Schafer, 1983 ). This method has been used to measure the changes in optical properties of cotyledons between 360 and 1000 run during the first 7d of development. The seedlings were either etiolated or grown under continuous white light, the latter either herbicide-treated (SAN 9789 = Norflurazon) or untreated. Some of the observed changes in seedlings grown under white light are due to chlorophyll accumulation. This accumulation leads to an increase in absorption coefficients at all wavelengths except in the 750 to 850 nm region. Reflectance, transmittance, and the scattering coefficient decreased markedly. Other changes seem to be independent of light conditions since they occur in much the same way under all treatments. These are a generally decreasing reflectance and scattering coefficient and an even stronger decrease of reflectance from the upper face of the cotyledon as compared to the reflectance from the lower face, in particular in the blue region of the spectrum. The observed changes are discussed in terms of light gradients and the resulting problems for in vivo spectroscopy.  相似文献

Recently, the systematics and biogeography of the Mediterranean biota have received much attention. This paper deals with Eupholidoptera Ma?an, a Mediterranean lineage of Tettigoniidae. The genus is restricted to the northern and eastern basin of the Mediterranean, with a significant number of species found on the Aegean islands. To produce a phylogeny and use it to make assumptions about the historical biogeography of Eupholidoptera, material of 46 species from several collections was studied. A phylogenetic analysis based mainly on morphological characters suggested two lineages in the genus: the E. chabrieri and the E. prasina groups. Based on the consistency between historical geographical events and branching events on the phylogenetic tree, Eupholidoptera is assumed to have evolved from an ancestor present in the Aegeid plate in the Mid‐Miocene. The division of the Aegeid plate into Anatolia and Greece in the Tortonian, the reoccurrence of terrestrial corridors between these mainlands in the Messinian, the regression of the Aegean area in the Pliocene and sea level changes in the Pleistocene are assumed to have been the main palaeogeographical events directing speciation in Eupholidoptera. As most of the species are allopatric, vicariance is suggested to be the main pattern. By combining the nature of the characters used in the phylogenetic analysis, the phylogenetic tree produced and the biogeographical assumptions, four tentative conclusions can be made: (i) radiation in the genus is a result of divergence in morphology; (ii) because the main character source is male genitalia, there has possibly been intensive sexual selection, which leads to morphological speciation; (iii) as the difference in temporal parameters of the song is prominent in sympatric/parapatric species pairs only, co‐occurrence is suggested to be the main reason driving divergence in the song; (iv) there seems to be a negative correlation between the size of the distribution range and the evolutionary rate in speciation; this may be the reason why the E. prasina group (restricted to a small part of the range of the genus) is more diverse than the E. chabrieri group, which is distributed over the entire range.  相似文献

Jerusalem artichoke DNA, extracted from resting rhizomes, wasanalysed by the reassociation kinetics of sheared denaturedfragments. Various fractions were isolated according to theirreassociation rates. The slowest fractions consist of unrepeatedor single-copy DNA which makes up 55 per cent of the total DNA.The total sequence length of the haploid genome is estimatedas corresponding to 0.23?1012 daltons. Among the fractions ofrepeated DNA isolated, two were relatively homogeneous withan average complexity of 2?108 and 109 daltons respectively.  相似文献

The photocontrol of chalcone synthase gene expression was studied by means of promoter analyses, in vitro systems, photoreceptor mutants and microinjections, and pharmacological approaches. A 52 bp element of the promoter is necessary and sufficient to transfer light regulation. Chalcone synthase expression is primarily under the control of phyA and blue/UV photo receptors; the latter are functional even in the absence of phyA and phyB. Phytochromes seem to be soluble proteins and, within seconds of irradiation, light-dependent phosphorrylations were observed in membrane-depleted cytosol preparations, indicating very early processes of signal transduction. Microinjection and pharmacological experiments reveal that, in the phyA pathway, heterotrimeric G-proteins, cGMP and a genistein-sensitive protein kinase are involved, whereas the UV pathway includes several trimeric G-proteins and Ca/calmodulin-dependent steps.  相似文献

Females of many species mate multiple times and store transferred sperm in storage organs. The mechanisms underlying sperm release from the stores at fertilization remain poorly understood, although they are central to an understanding of the female influence on post-copulatory male competition. Using double-mated females of the yellow dung fly, we counted the sperm sticking to the surface of deposited eggs of two successive clutches to obtain insight into the physiological processes associated with fertilization. The number of sperm released to fertilize an egg decreased between the first and second clutches, as well as within clutches from early to late eggs. These results indicate that: (1) sperm are lost from the stores over time independent of egg laying and (2) the number of sperm released depends on the amount of sperm stored. The lower number of sperm on eggs of the second clutches was accompanied by a strong increase of the proportion of sperm adhering to the micropyle region, suggesting that sperm use is more efficient and sperm release better controlled when sperm supply is substantially reduced. Disturbances such as fire, hurricanes, and herbivory often result in the net release of CO2 from forests to the atmosphere, but the magnitude of carbon (C) loss is poorly quantified and difficult to predict. Here, we investigate the carbon balance of an oak/pine forest in the New Jersey Pine Barrens using the Canopy Conductance Constrained Carbon Assimilation (4C‐A) model. The 4C‐A model utilizes whole‐tree sap‐flux and leaf‐level photosynthetic gas exchange measurements at distinct canopy levels to estimate canopy assimilation. After model parameterization, sensitivity analyses, and evaluation against eddy flux measurements made in 2006, the model was used to predict C assimilation for an undisturbed year in 2005, and in 2007 when the stand was completely defoliated for 2–3 weeks during an infestation of gypsy moths (Lymantria dispar L.). Following defoliation, only 50% of the foliage reemerged in a second flush. In 2007, canopy net assimilation (AnC), as modeled with the 4C‐A, was reduced to approximately 75% of AnC in 2006 (940 vs. 1240 g C m?2 a?1). Overall, net primary production (NPP) in 2007 was approximately 240 g C m?2 a?1 (vs. 250 g C m?2 a?1 in 2006), with 60% of NPP allocated to foliage production, a short‐term carbon pool. Woody biomass accumulation, a long‐term carbon pool, was reduced by 20% compared with the previous year (72 vs. 57 g C m?2 a?1 in 2006 and 2007, respectively). The overall impact of the defoliation spanned 21% of upland forests (320 km2) in the New Jersey Pine Barrens, representing a significant amount of overall C not being taken up from the atmosphere by the forest, thus not accumulated in the biosphere.  相似文献

Abstract Mustard seedlings were light treated at 24 h after sowing (25°C) to induce phytochrome-mediated anthocyanin synthesis in cotyledons and hypocotylar hook. All light treatments were performed within the range of the reciprocity law. The in situ photoconversion kinetics of phytochrome (Pr→ Pfr) were measured under the same light treatment. It was found that between 0.4 and 1.0 relative Pfr level the amount of anthocyanin extracted from the organs at 52 h after sowing was linearily correlated with the amount of Pfr produced at 24 h in cotyledons and hypocotylar hook. It is concluded that an explanation of the fluence response function for red light mediated anthocyanin synthesis in the mustard seedling does not require the concept of active vs. bulk phytochrome.  相似文献
