Nine yeast strains were isolated from spontaneous fermentations in the Alsace area of France, during the 1997, 1998 and 1999 grape harvests. Strains were characterized by pulsed-field gel electrophoresis, PCR-restriction fragment length polymorphism (RFLP) of the MET2 gene, delta-PCR, and microsatellite patterns. Karyotypes and MET2 fragments of the nine strains corresponded to mixed chromosomal bands and restriction patterns for both Saccharomyces cerevisiae and Saccharomyces bayanus var. uvarum. They also responded positively to amplification with microsatellite primers specific to both species and were demonstrated to be diploid. However, meiosis led to absolute nonviability of their spores on complete medium. All the results demonstrated that the nine yeast strains isolated were S. cerevisiaexS. bayanus var. uvarum diploid hybrids. Moreover, microsatellite DNA analysis identified strains isolated in the same cellar as potential parents belonging to S. bayanus var. uvarum and S. cerevisiae.  相似文献

Spatial variability of methane emissions from Swiss alpine fens   总被引:1,自引:0,他引:1  
Wetland ecosystems are a major natural source of the important greenhouse gas methane (CH4). Among these ecosystems, fens have been shown to release high quantities of CH4. Data on CH4 emissions from alpine fens are scarce and mainly limited to the United States and China. Therefore, static chambers were used to quantify CH4 emissions from 14 fens located in the Swiss Alps. The aims of this study were to determine the spatial variability of the emissions and to identify potential key factors which influence CH4 turnover. The fens were located at altitudes between 1,800 and 2,600 m a.s.l., the pore water varied from acidic to slightly acidic (pH 4.5–6.4) and the vegetation was dominated by plants of the genus Carex. In addition, the underlying bedrock was either siliceous or calcareous. Methane emissions ranged from 74 ± 43 to 711 ± 212 mg CH4 m?2 day?1. The type of bedrock, the plant biomass above the water table and the CH4 pore water concentrations at depths from 0 to 20 cm were the main factors influencing CH4 emissions. Detailed measurements in three selected fens suggested that more than 98 % of the total CH4 emissions are due to plant-mediated transport.  相似文献

Uncoupling effects of local anesthetics on rat liver mitochondria   总被引:10,自引:0,他引:10  
We demonstrate in this paper that bupivacaine, a local anesthetic, can act alone as an uncoupler of rat liver mitochondria. It stimulates state 4 respiration, induces a swelling in potassium acetate (in the presence of valinomycin), and collapses the transmembrane potential. Lidocaine, another local anesthetic, requires the presence of a lipophilic anion such as TPB- to produce the same effects. TPB- can also reinforce the action of bupivacaine. These differences in action of the two local anesthetics can be explained by the difference in the liposolubility.  相似文献

Alzheimer's disease (AD) is characterized by severe neuronal loss as well as the accumulation of amyloid‐β (Aβ), which ultimately leads to plaque formation. Although there is now a general agreement that the aggregation of Aβ can be initiated by prion‐like seeding, the impact and functional consequences of induced Aβ deposits (Aβ seeding) on neurons still remain open questions. Here, we find that Aβ seeding, representing early stages of plaque formation, leads to a dramatic decrease in proliferation and neurogenesis in two APP transgenic mouse models. We further demonstrate that neuronal cell death occurs primarily in the vicinity of induced Aβ deposits culminating in electrophysiological abnormalities. Notably, environmental enrichment and voluntary exercise not only revives adult neurogenesis and reverses memory deficits but, most importantly, prevents Aβ seeding by activated, phagocytic microglia cells. Our work expands the current knowledge regarding Aβ seeding and the consequences thereof and attributes microglia an important role in diminishing Aβ seeding by environmental enrichment.  相似文献

Forty-six Lecanicillium strains and one Verticillium strain were isolated from subterranean and epiphytic arthropods, soil, and other sources collected in Indonesia and Japan. These strains were identified as nine Lecanicillium and one Verticillium species including six undescribed species based on light microscopy and the sequences of the ITS-1 and ITS-2 regions including 5.8S ribosomal DNA. Four of the ten species (L. araneicola, L. kalimantanense, Lecanicillium sp. 4, and V. indonesiacum) were recovered from Indonesia, five of the ten (L. attenuatum, L. fusisporum, L. psalliotae, Lecanicillium sp. 1, and Lecanicillium sp. 3) were from Japan, and L. saksenae was from both countries. In this article, new species (L. araneicola, L. kalimantanense, and V. indonesiacum) and a new combination (L. saksenae) are proposed from the fungi isolated from epiphytic and subterranean arthropods collected in East Kalimantan.  相似文献
