Data on immuno- and biochemical identification, genetic control and phylogenesis of new allotype Lpm13 of the Lpm system in domestic mink are presented. This allotype is encountered in mink populations with the frequency 0.9 and higher. The availability of Lpm13 genetic marker permitted another haplotype to be revealed, in addition to the eight known Lpm haplotypes by means of genetic analysis. It was established that, alongside with the earlier described haplotype Lpm3,4,6,8,9,10,11 (abbreviation H3), there exists a similar haplotype, Lpm3,4,6,8,9,10,11,13 (abbreviation H3.13), containing the Lpm13 gene. Of the rest seven haplotypes, five have the Lpm13 gene and two do not. Taking into account this gene and corresponding antigenic marker, the differentiation of 28, instead of 25, phenotypes and 45, instead of 36, genotypes for the Lpm system became possible. Lpm13 antigenic specificity was found with no exception in all individual serum samples taken from ten species and interspecific hybrids of Mustelidae which are closely related to domestic mink. The data obtained give grounds to refer the newly identified Lpm13 gene to the first evolutionary conservative category of genes of the multigenic Lpm system which is also represented by the Lpm6, Lpm9, Lpm10 and Lpm11 genes. The hypotheses of instantaneous formation of polymorphism of the Lpm system in domestic mink are briefly regarded.  相似文献

The results of genetic study on linkage of Lpm locus with peptidase B gene are presented. Investigation of 111 offspring back-crosses shows that Lpm allotypes and allelic variants of peptidase B are inherited in concert. The frequency of recombination between the Lpm locus and peptidase B gene is 11 +/- 3% in male. Since it was earlier established that peptidase B gene is a marker of chromosome 9, our data indicate that the Lpm loci family is situated in the chromosome 9 of domestic mink.  相似文献

Distribution in the organs and tissues of two proteins of alpha-macroglobulin fraction, that differ in their antigenic structures, has been studied in the American mink. The both proteins (alpha 2M and Lpm) are present in hepatocytes, in cells of the follicular epithelium of the ovary, in the thymic bodies, in the alveolar macrophages of the lungs and in the splenic lymphoid nodes. Joint localization of alpha 2M and Lpm is revealed in the connective tissue of all the organs examined. The exception make the stomach and the uterine, where alpha 2M is revealed but not Lpm. The results obtained demonstrate a similar distribution of the two alpha-macroglobulins in the mink organism. They correspond to the literature data on morphofunctional topography of alpha 2M in the man. Certain individual differences in alpha 2M and Lpm localization can reflect peculiarities inherent in each of these alpha-macroglobulins of the American mink.  相似文献

Background

Frequent painful vaso-occlusive crises (VOCs) were associated with mortality in the Cooperative Study of Sickle Cell Disease (CSSCD) over twenty years ago. Modern therapies for sickle cell anemia (SCA) like hydroxyurea are believed to have improved overall patient survival. The current study sought to determine the relevance of the association between more frequent VOCs and death and its relative impact upon overall mortality compared to other known risk factors in a contemporary adult SCA cohort.

Methods

Two hundred sixty four SCA adults were assigned into two groups based on patient reported outcomes for emergency department (ED) visits or hospitalizations for painful VOC treatment during the 12 months prior to evaluation.

Results

Higher baseline hematocrit (p = 0.0008), ferritin (p = 0.005), and HDL cholesterol (p = 0.01) were independently associated with 1 or more painful VOCs requiring an ED visit or hospitalization for acute pain. During a median follow-up of 5 years, mortality was higher in the ED visit/hospitalization group (relative risk [RR] 2.68, 95% CI 1.1-6.5, p = 0.03). Higher tricuspid regurgitatant jet velocity (TRV) (RR 2.41, 95% CI 1.5-3.9, p < 0.0001), elevated ferritin (RR 4.00, 95% CI 1.8-9.0, p = 0.001) and lower glomerular filtration rate (RR=2.73, 95% CI 1.6-4.6, p < 0.0001) were also independent risk factors for mortality.

Conclusions

Severe painful VOCs remain a marker for SCA disease severity and premature mortality in a modern cohort along with other known risk factors for death including high TRV, high ferritin and lower renal function. The number of patient reported pain crises requiring healthcare utilization is an easily obtained outcome that could help to identify high risk patients for disease modifying therapies.

Abstract

The molecular basis of the marked structure-activity relationship for a homologous series of DNA-binding phenoxazone drugs (ActII-ActIV) has been investigated by NMR spectroscopy and molecular mechanics. The spatial structures of the complexes between the drugs and a model deoxytetranucleotide, 5′-d(TpGpCpA), have been determined by molecular mechanics methods using homonuclear 1H-1H 2D-NOESY and heteronuclear 1H-31P (HMBC) NMR spectroscopic data. Observed intermolecular NOE contacts and equilibrium binding studies confirm that the binding affinity of the synthetic phenoxazone derivatives with d(TGCA) decreases with an increase in the number of CH2 groups in the dimethylami- noalkyl side chains, i.e., ActII > ActIII > ActIV, in agreement with the observed biological activity of these compounds. Molecular mechanics calculations of the spatial structures of the intercalated complexes of ActII-ActIV with d(TGCA) indicate that the different binding constants of the phenoxazone derivatives with the DNA oligomer are due to the different degrees of intercalation of the chromophore and the different steric arrangements of aminoalkyl side chains in the minor groove of the tetramer duplex; this results in different distances between the negatively-charged phosphates of the DNA duplex and the terminal positively-charged N(CH3)2 groups of the side chains.  相似文献

The results of genetic studies of transformation of alpha-macroglobullin (AM) allogroups (i.e., fixed allotype combinations) in pigs are presented. Population and hybridization studies showed that the phenomenon discovered was based on activation/inactivation of the AMI allotype expression, which depended on the genetic environment and interlocus relationships. Genetic and selection bases, as well as the significance of the transformation phenomenon described, are discussed.  相似文献

Effects of medium viscosity on kinetic parameters of poly(U) hydrolysis catalyzed by RNase from Bac. intermedius 7P (binase) were studied in solutions of sucrose (4-50 wt. %) and glycerol (35-62 wt. %) in Tris--sodium acetate buffer (pH 7.5) at 25 degreesC. The rate constant of reaction kcat was practically unchanged over a wide range of viscosities (1-15 cP for sucrose and 2.5-3 cP for glycerol). In glycerol solutions, kcat slightly increased with viscosity increase from 4 to 10 cP. Addition of NaCl to the buffer medium resulted in an inhibitory effect of Na+ on kcat, prevented by 50% sucrose or 60% glycerol. It is concluded that binase-catalyzed poly(U) cleavage occurs through a "tense"-substrate mechanism, similarly to reactions catalyzed by alpha-chymotrypsin, trypsin, and laccase.  相似文献

The specificity of phenotypic expression and inheritance of immunoglobulin allotypes IgG1a and IgG2b in piglets are discussed. It was shown that a negative state by both these allotypes is repeatedly found among newborn piglets but is extremely rare in pigs older than one month. A model, which simultaneously describes the genetic determination of allogroups formed by allotypes IgG1a and IgG2b and the dynamics of ontogenetic expression of individual genotype by systemIgG with a result of a visually registered phenotype, was developed. Allele frequencies by system IgG were assessed in populations of domestic pigs of commercial breeds, laboratory miniature pigs, and Eurasian wild boar.  相似文献
