We investigated adaptive responses of the photosynthetic electron transport to a decline in the carbon assimilation capacity. Leaves of different ages from wild-type tobacco (Nicotiana tabacum) L. var Samsun NN and young mature leaves of tobacco transformants with impaired photoassimilate export were used. The assimilation rate decreased from 280 in young mature wild-type leaves to below 50 mmol electrons mol chlorophyll(-1) s(-1) in older wild-type leaves or in transformants. The electron transport capacity, measured in thylakoids isolated from the different leaves, closely matched the leaf assimilation rate. The numbers of cytochrome (cyt)-bf complexes and plastocyanin (PC) decreased with the electron transport and assimilation capacity, while the numbers of photosystem I (PSI), photosystem II, and plastoquinone remained constant. The PC to PSI ratio decreased from five in leaves with high assimilation rates, to values below one in leaves with low assimilation rates, and the PC versus flux correlation was strictly proportional. Redox kinetics of cyt-f, PC, and P700 suggest that in leaves with low electron fluxes, PC is out of the equilibrium with P700 and cyt-f and the cyt-f reoxidation rate is restricted. It is concluded that the electron flux is sensitive to variations in the number of PC, relative to PSI and cyt-bf, and PC, in concert with cyt-bf, is a key component that adjusts to control the electron transport rate. PC dependent flux control may serve to adjust the electron transport rate under conditions where the carbon assimilation is diminished and thereby protects PSI against over-reduction and reactive oxygen production. 相似文献

Previous investigations of exposure to electric, magnetic, or electromagnetic fields (EMF) in households were either about electricity supply EMFs or radio frequency EMFs (RF‐EMFs). We report results from spot measurements at the bedside that comprise electrostatic fields, extremely low‐frequency electric fields (ELF‐EFs), extremely low‐frequency magnetic fields (ELF‐MFs), and RF‐EMFs. Measurements were taken in 226 households throughout Lower Austria. In addition, effects of simple reduction measures (e.g., removal of clock radios or increasing their distance from the bed, turning off Digital Enhanced Cordless Telecommunication (DECT) telephone base stations) were assessed. All measurements were well below International Commission on Non‐Ionizing Radiation Protection (ICNIRP) guideline levels. Average night‐time ELF‐MFs (long‐term measurement from 10 pm to 6 am, geometric mean over households) above 100 nT were obtained in 2.3%, and RF‐EMFs above 1000 µW/m2 in 7.1% of households. Highest ELF‐EFs were primarily due to lamps beside the bed (max = 166 V/m), and highest ELF‐MFs because of transformers of devices (max = 1030 nT) or high current of power lines (max = 380 nT). The highest values of RF‐EMFs were caused by DECT telephone base stations (max = 28979 µW/m2) and mobile phone base stations (max = 4872 µW/m2). Simple reduction measures resulted in an average decrease of 23 nT for ELF‐MFs, 23 V/m for ELF‐EFs, and 246 µW/m2 for RF‐EMFs. A small but statistically significant correlation between ELF‐MF exposure and overall RF‐EMF levels of R = 0.16 (P = 0.008) was computed that was independent of type (flat, single family) and location (urban, rural) of houses. Fcgamma receptors (FcgammaR) and the C5a receptor (C5aR) are key effectors of the acute inflammatory response to IgG immune complexes (IC). Their coordinated activation is critical in IC-induced diseases, although the significance of combined signaling by these two different receptor classes in tissue injury is unclear. Here we used the mouse model of the passive reverse lung Arthus reaction to define their requirements for distinct phosphoinositide 3-kinase (PI3K) activities in vivo. We show that genetic deletion of class IB PI3Kgamma abrogates C5aR signaling that is crucial for FcgammaR-mediated activation of lung macrophages. Thus, in PI3Kgamma(-/-) mice, IgG IC-induced FcgammaR regulation, cytokine release, and neutrophil recruitment were blunted. Notably, however, C5a production occurred normally in PI3Kgamma(-/-) mice but was impaired in PI3Kdelta(-/-) mice. Consequently, class IA PI3Kdelta deficiency caused resistance to acute IC lung injury. These results demonstrate that PI3Kgamma and PI3Kdelta coordinate the inflammatory effects of C5aR and FcgammaR and define PI3Kdelta as a novel and essential element of FcgammaR signaling in the generation of C5a in IC disease. 相似文献

Interleukin-6 (IL-6) expression and secretion, induced by inflammatory processes, stimulate the acute phase response cascade. The overexpression of IL-6 contributes to a variety of inflammatory diseases, e.g. rheumatoid arthritis, Castleman’s disease, multiple myeloma, and prostate cancer. Screening for high amounts of IL-6 in the patients’ blood serum can be crucial for an adequate treatment. In this study, five novel murine monoclonal antibodies (mAbs) reactive to human IL-6 were generated. The mAbs were characterized for potential diagnostic purposes and recombinant antibodies were derived thereof. Initial epitope mapping using a combination of blocking experiments and Hyper-IL-6, a fusion protein consisting of IL-6 and the soluble IL-6 receptor revealed distinct but overlapping binding sites. At least one of the mAbs was found to interact with the region of IL-6/ IL-R complex formation. Three mAbs were applied successfully in intracellular staining by flow cytometry, whereas one of the mAbs showed comparable binding as a reference reagent. Furthermore, the mAbs were tested for applications in various immunological assays such as ELISA, Western blot and surface plasmon resonance spectroscopy (SPR), using IL-6 from commercial sources as well as in-house produced protein (IL-6_IME). The limit of detection was determined by sandwich ELISA (0.5 ng/mL, SD ±0.005). Our results also demonstrated that the recombinant IL- 6 produced was functional and correctly folded. In midday ginkgo ( Ginkgo biloba L. ) leaves have to bear photon flux density over 1 400 μmol·m-2·s-l in combination with high temperatures around 35℃ at natural habitat. They show typical midday depression of stomatal conductance and of CO2 assimilation rate. The zeaxanthin changes with light intensity during the day. The influence of the combination of strong light and temperature on photoinhibition was also examined in the laboratory. A low CO2 internal conductance (31 mmol· m- 2·s- 1 ) was found in ginkgo leaves, which had been exposed to excessive light at temperature between 15 ℃ and 35 ℃ with reduced CO2 (80 μL·L-l) or oxygen (2%) for 2 h, causing a low CO2 concentration at the carboxylation site and a high proportion of photorespimtion. The ratio of electron transport to CO2 fixation was rather high in ginkgo ( 16 e- /CO2 at 25 ℃ ) as compared with other plants. It increased with temperature also in 2% 02 which could not be explained solely as due to change of photorespimtion. The reduction of oxygen in 340 or 80 μL·L- 1 CO2 had no effect on the extent of photoinhibition at all temperatures, which indicated that eleetron flow caused by photorespiration in excess light was negligible in protective effect in ginkgo leaves. However, a decreased CO2 coneentration increased photoinhibition, especially at high temperature. It is concluded that the dissipation of excessive excitation energy in the PS II antennae through the xanthophyll cycle may be the major protective mechanism to preventing from the deteriorated effects of strong light in ginkgo leaves. 相似文献
