Fluctuations of grass pollen content in the atmosphere of East Perugia and meteorological correlations (year 1989)

Summary Gramineae pollination from a pollen monitoring station located in the eastern suburb of Perugia and meteorological correlations are reported. The data refers to the year 1989. Grass pollen peak pollination was from May to July; in this period the influence of relative humidity and of temperature on pollen concentration was very high. Phenological observations, to identify the time of maximum stamen extension in the most common genera in the area, are also reported. During the period of investigation the counts of pollen grains over four-hour periods showed a regular diurnal rhythm with peaks of concentration in the four-hour period 16.00–20.00. Aerosporological data and meteorological data related to four-hour periods were correlated following different criteria.     

Observing the Temperature Dependent Transition of the GP2 Peptide Using Terahertz Spectroscopy

The GP2 peptide is derived from the Human Epidermal growth factor Receptor 2 (HER2/nue), a marker protein for breast cancer present in saliva. In this paper we study the temperature dependent behavior of hydrated GP2 at terahertz frequencies and find that the peptide undergoes a dynamic transition between 200 and 220 K. By fitting suitable molecular models to the frequency response we determine the molecular processes involved above and below the transition temperature (T _D). In particular, we show that below T _D the dynamic transition is dominated by a simple harmonic vibration with a slow and temperature dependent relaxation time constant and that above T _D, the dynamic behavior is governed by two oscillators, one of which has a fast and temperature independent relaxation time constant and the other of which is a heavily damped oscillator with a slow and temperature dependent time constant. Furthermore a red shifting of the characteristic frequency of the damped oscillator was observed, confirming the presence of a non-harmonic vibration potential. Our measurements and modeling of GP2 highlight the unique capabilities of THz spectroscopy for protein characterization.     

A selective differential medium for the isolation of Listeria monocytogenes

A new medium has been developed for the isolation of Listeria monocytogenes from clinical specimens with a mixed flora. Almost complete inhibition of unwanted organisms was achieved and recognition of colonies of Listeria spp. was usually possible after 24 h using the aesculin-ferric ammonium citrate indicator system. Compared to McBride agar the new medium was more inhibitory to representative contaminating species in pure culture and more successful in isolating small numbers of L. monocytogenes from artificially seeded clinical specimens.     

The role of air temperature in determining dormancy release and flowering ofCorylus avellana L.

Summary 7 years of airborne pollen monitoring in Perugia (central Italy) were used to determine the temperature requirements to break dormancy and to resume growth and bloom ofCorylus avellana L.,Corylus needs 1000 chill-units to complete its dormancy and this value, in the Perugian area, is met by the end of December or the first days of January. MoreoverCorylus trees require 220 growth degree hours before they are able to flower. If air temperature is high, this value can be achieved in only 10 days, but if the temperature remains too low, the heat accumulation can require up to 35 days. With these parameters it is possible to build a model to predict the date of the beginning ofCorylus avellana pollen season.     

Studies on human blood lymphocytes with iC3b (type 3) complement receptors. I. Granular, Fc-IgG receptor positive and negative subsets in healthy subjects and patients with systemic lupus erythematosus

By using the OKM1 monoclonal antibody and the fluorescence-activated cell sorter to identify lymphocytes bearing iC3b (type 3) complement receptors, two principal populations of OKM1+ lymphocytes have been identified in human peripheral blood. One subset exhibited azurophilic granules and Fc receptors for IgG stained by Leu-11. The other population did not display FcR, but was enriched in cells reacting with OKT3 and OKT8 (low intensity). In healthy subjects, approximately 60% of CR3+ lymphocytes were granular FcR-bearing cells and only 18% co-expressed OKT3 determinants. In patients with systemic lupus erythematosus (SLE), CR3+ lymphocytes were predominantly FcR negative cells and 71% lacked granules. Only 33% reacted with Leu-11, but 50% co-expressed OKT3, 44% reacted with OKT8+, and 15% were OKT4+. We tested the hypothesis that agranular OKT3+ Leu-11- lymphocytes, such as those found in SLE patients, contained the precursors of natural killer (NK) cells. Leu-11+ cells were removed from normal lymphocytes by complement lysis, and the remaining cells were treated with recombinant IFN-alpha, IFN-gamma, or IL 2. These procedures were ineffective in generating typical NK effector cells. Our studies do not support the hypothesis that CR3+ Leu-11- lymphocytes are the precursors of granular Leu-11+ NK cells.     

In vitro chondrogenesis and cell viability

Anterior somites cultured with (NSA) or without (SA) notochord, and posterior somites cultured with (NSP) or without notochord (SP) were compared with respect to changes in their DNA content, their potential to synthesize the active sulfate principle phosphoadenosine phosphosulfate (PAPS), and their ability to accumulate ³⁵S-sulfate.Chondrogenesis was observed in the NSA, NSP, and SP explants, but was rarely noted in the SA explants. A decrease in DNA content during the initial 48 hr of culture was common to all explants. After this initial decrease, DNA content increased most in those explants forming cartilage. The synthesis of PAPS by cell-free extracts of each type of somite explant also decreased during the initial period of culture. Only extracts of those explants undergoing chondrogenesis showed increases in PAPS synthesis with continued culture. Each type of somite explant accumulated ³⁵S-sulfate into chondroitin sulfate during the first hours of culture. The non-chondrogenic SA explants accumulated little ³⁵S-sulfate during the period of culture. At varying times after 24 hr the chondrifying explants (NSA, SP, and NSP) initiated an increased rate of accumulation of ³⁵S-sulfate.Cartilage nodules, increases in DNA content, PAPS synthesis and ³⁵S-sulfate accumulation occurred within the same 24 hr period, during the 2nd day in NSP explants, the 3rd day in NSA explants, and between the 3rd and 4th day for SP explants. A hypothesis of in vitro somite chondrogenesis based on differential cell viability is presented.     

The results of 2-year pollen monitoring of an urban network in Perugia,Central Italy

Continuous pollen monitoring of an urban network consisting of three stations has been undertaken for a period of 2 years in Perugia, central Italy. The aim has been to establish whether the Perugia pollen trap, active since 1983, is still representative of the area following recent urbanisation. Quantitative differences were found between the stations, reflecting different vegetational areas, but only slight differences were detected in relation to the timing of the principal period of pollination. Therefore, although individual pollen traps are necessary to characterize fully the different areas, one trap is sufficient to determine the key allergenic thresholds in the studied area.
