Respiration of the air breathing fishPiabucina festae   总被引:1,自引:1,他引:0  
Summary Piabucina festae, a Central American stream fish, breathes air frequently, even in air saturated water, however, is not an obligate air breather. Without access to air, it can maintain routine by aquatic respiration down to aP wO2 of about 70 Torr which is its critical O2 tension (P cO2, Fig. 5). Aerial respiration averages 10% of total in air saturated water and 70% in hypoxic water (Fig. 4). At lowP wO2 air breathing is more frequent (Fig. 1), and more O2 is utilized from each air breath (Table 3), and tidal volume may increase (Fig. 7). Vascularized respiratory compartments or cells (Fig. 6), located in the second chamber of the physostomus gas bladder, function for aerial respiration. In ventilation air is gulped and forced through a large pneumatic duct into the gas bladder, excess gas is then released through opercula. Inspiration always precedes expiration and tidal volume is small, keeping gas bladderP O 2 low (Table 4). Major differences in the air breathing physiology ofP. festae and other species are its higherP cO2, a low aerial in normoxic water, even though air gulps are frequent, and its pattern of inhalation prior to expiration. The interrelationship and optimization of the three gas bladder functions (buoyancy, sound reception, and air breathing) inP. festae is discussed. Aerial respiration may have evolved secondarily to the gas bladder's function in buoyancy control.  相似文献

Accelerated molecular evolution in halophilic crustaceans   总被引:8,自引:0,他引:8  
In contrast to the stable ionic composition of the oceans, inland waters show striking diversity, possessing salt concentrations varying from I mM to 5 M. Although species diversity is highest in fresh water, some lineages have colonized hypersaline environments where they encounter elevated levels of both ultraviolet (UV) radiation and osmotic stress. This study compares rates of evolution in halophilic and freshwater taxa for two groups of microcrustaceans, anostracans and daphniids, from Australia and North America. The results establish that halophilic species show consistent rate acceleration, involving elevated levels of both insertion/deletion events and of nucleotide substitutions. The elevated pace of molecular evolution does not appear to be linked to selection or to other agents that are known to influence the supply rate of mutations, such as UV exposure, generation length, or shifts in metabolic rate. However, variance in ionic strength, which is known to have potent effects on DNA-protein interactions as well as on the structural properties of DNA and proteins, might account for the lowered fidelity of DNA replication in life from hypersaline settings. Regardless of its cause, the consistent rate acceleration in halophiles suggests that past efforts to employ sequence divergences to date events, such as the age of asexual lineages in Artemia, have resulted in serious overestimates. More generally, the results indicate that coordinated shifts in rates of molecular evolution may occur in lineages exposed to extreme environmental conditions.  相似文献

The study of venom components is an important step toward understanding the mechanism of action of such venoms and is indispensable for the development of new therapies. This work aimed to investigate the venom of Lachesis muta rhombeata and evaluate enzymes related to its toxicity. Phospholipase A2 (PLA2), l ‐amino acid oxidase (LAAO), and proteinase activities were measured, and the molecular weights were estimated. We found the venom to contain one PLA2 (17 kDa), one LAAO (132 kDa), and three serine proteinases (40, 31, and 20 kDa). Although only serine proteinases were observed in the zymogram, metalloproteinases were found to contribute more to the total proteolytic activity than did serine proteinases. The work confirmed the presence of highly active enzymes; and, moreover, we proposed a novel method for confirming the presence of LAAOs by zymography. We also suggested a simple step to increase the sensitivity of proteinase assays. © 2012 Wiley Periodicals, Inc. Three DNA polymerase activities, named 1, 2 and 3 were purified from maize embryo axes and were compared in terms of ion requirements, optimal pH, temperature and KCl for activity, response to specific inhibitors and use of templates. All three enzymes require a divalent cation for activity, but main differences were observed in sensitivity to inhibitors and template usage: while DNA polymerases 1 and 2 were inhibited by N-ethyl maleimide and aphidicolin, inhibitors of replicative-type enzymes, DNA polymerase 3 was only marginally or not affected at all. In contrast, DNA polymerase 3 was highly inhibited by very low concentrations of ddTTP, an inhibitor of repair-type enzymes, and a 100-fold higher concentration of the drug was needed to inhibit DNA polymerases 1 and 2. Additionally, DNA polymerases 1 and 2 used equally or more efficiently the synthetic template polydA-oligodT, as compared to activated DNA, while polymerase 3 used it very poorly. Whereas DNA polymerases 1 and 2 shared properties of replicative-type enzymes, DNA polymerase 3 could be a repair-type enzyme. Moreover, a DNA primase activity copurified with the 8000-fold purified DNA polymerase 2, strenghtening the suggestion that polymerase 2 is a replicative enzyme, of the -type. This DNA primase activity was also partially characterized. The results are discussed in terms of relevant data about other plant DNA polymerases and primases reported in the literature.  相似文献

The identification of CYP2C9 and VKORC1 genes has strongly stimulated the research on pharmacogenetics of coumarins in the last decade. We assessed the combined influence of CYP2C9 *2 and *3, and VKORC1 c.-1639G>A, 497C>G, and 1173C>T variants, on acenocoumarol dosage using a novel algorithm approach, in 193 outpatients who had achieved stable anticoagulation. We constructed an “acenocoumarol-dose genotype score” (AGS, maximum score = 100) based on the number of alleles associated with higher acenocoumarol dosage carried by each subject for each polymorphism. The mean AGS was higher in the high-dose (>28mg/week) compared with the low-dose (<7mg/week) group (mean(SEM) of 84.1±3.4 vs. 62.2±4.8, P = 0.008). An AGS>70 was associated with an increased odds ratio (OR) of requiring high acenocoumarol dosage (OR: 3.347; 95%CI: 1.112–10.075; P = 0.032). In summary, although more research is necessary in other patient cohorts, and this algorithm should be replicated in an independent sample, our data suggest that the AGS algorithm could be used to help discriminating patients requiring high acenocoumarol doses to achieve stable anti-coagulation.  相似文献

Probing the relationships of the branchiopod crustaceans   总被引:4,自引:0,他引:4  
The Branchiopoda display extraordinary variation in body form, even within the morphologically diverse crustaceans. To fully understand the origin and evolution of these morphological reconfigurations, a robust phylogeny of the group is essential. To infer the affinities among branchiopods, we employed two approaches to taxon and gene sampling, presented new sequence data from three genes, incorporated previously published sequence data from three additional genes, and utilized comprehensive techniques of phylogeny reconstruction. The results provided support for a number of longstanding hypotheses concerning the relationships among the orders. For example, we obtained support for the Cladoceromorpha and Gymnomera, and favoured a unique arrangement of the cladoceran orders. A few affinities remain to be resolved, particularly at the base of the Phyllopoda and within the Anomopoda. However, the results suggest that increased gene sampling is recommended for future investigations of branchiopod systematics.  相似文献

Maize CycD3;1 associates to CDKA or CDKB1;1 proteins during germination and the complexes formed develop kinase activity. These complexes appear to vary in size as germination proceeds, suggesting association to different sets of proteins. CycD3;1 and associated CDK proteins respond to phytohormones and sucrose. Results revealed a reduction in the CycD3;1 protein amount along germination in the presence of indoleacetic acid (IAA) or abscisic acid (ABA), although in the latter protein levels recover at the end of germination. While the levels of CDKA increase with IAA, they decrease with ABA. Both phytohormones, IAA and ABA, increase levels of CDKB1;1 only during the early germination times. CycD3;1 associated kinase activity is only reduced by both phytohormones towards the end of the germination period. On the other hand, lack of sucrose in the imbibition medium strongly reduces CycD3;1 protein levels without affecting the levels of neither CDKA nor CDKB1;1. The corresponding CycD3;1 associated kinase activity is also severely decreased. The presence of sucrose in the medium appears to stabilize the CycD3;1 protein levels.  相似文献
