Pythium oligandrum Drechsler bearing spherical sporangia with complex subglobose elements was isolated for the first time in Egypt from agricultural field soil cultivated with alfalfa (Trifolium alexandrinum) in El-Minia, Egypt. This fungus was found to be an active bio-control agent against P. ultimum var. ultimum, the damping-off organism of wheat. In agar plates, P. oligandrum parasitized P. ultimum var. ultimum hyphae with the aid of thin haustorial branches or infection pegs, eventually leading to host destruction. Incorporation of P. oligandrum into carboxymethylcellulose seed coating successfully eliminated pre-emergence damping-off of wheat caused by P. ultimum var. ultimum, whereas Post-emergence damping-off was prevented by adding inocula of P. oligandrum to the soil. Of 24 Trichoderma isolates, T harzianum Rifai (T24) showed a potential for control of the phytopathogenic basidiomycete Sclerotium rolfsii. When T24 was grown on different carbon sources, growth inhibition of S. rolfsii by the T24 culture filtrate correlated with the activity of extracellular chitinase and beta-1,3-glucanase. The 43-kilodalton (kDa) chitinase and the 74-kDa beta-1,3-glucanase were purified from the T24 culture filtrate in two and three steps, respectively, using ammonium sulphate precipitation followed by hydrophobic interaction chromatography (phenyl-Sepharose) and gel filtration (beta-1,3-glucanase). Km and Kcat were 3.8 g l(-1) and 0.71 s(-1) for the chitinase (chitin) and 1.1 g(-1) and 52 s(-1) for the beta-1,3-glucanase (laminarin). The chitinase showed higher activity on chitin than on less-acetylated substrate analogues (chitosan), while the beta-1,3-glucanase was specific for beta-1,3-linkages in polysaccharides. Both enzymes were stable at 30 degrees C, while at 60 degrees C the chitinase and the beta-1,3-glucanase were rapidly inactivated, showing half-lives of 15 and 20 min, respectively. The enzymes inhibited growth of S. rolfsii in an additive manner showing a promising ED50 (50% effective dose) value of 2.7 microg/ml.  相似文献

Twenty-five Penicillium species isolated from Egyptian soil were examined for their ability to produce gluconic acid in surface culture. Of the eight species capable of producing gluconic acid, Penicillium puberulum gave the maximum yield (91% gluconic acid from glucose after 7 days of fermentation with 3% CaCO3). Peptone was the best nitrogen source for acid fermentation and glucose was superior to sucrose. Addition of low concentrations of KH2PO4 and MgSO4 - 7 H2O stimulated acid production. An initial pH of 6.1 was most favourable for acid accumulation and addition of CaCO3 was necessary for maximum acid production.  相似文献

In wheat, corn and sorghum grains twenty-six genera and seventy-seven species including sixteen ofAspergillus and twenty-one ofPenicillium were identified.In grain samples adjusted to various moisture levels (up to 18.8 % on a dry-weight basis) and stored for 4 months at 8, 20 and 30 C seed-borne fungi were monthly identified and counted, and the germinability of the grains was tested. In the control samples (moisture content 7.1–8.2 %) temperature had no effect on the fungal counts and on the precentage germination. With the rise of temperature and moisture content the fungal counts markedly increased and the germinability declined.The list of fungi dominant in one or more of the experiments, included eight species ofAspergillus, six ofPenicillium, two ofFusarium and one each ofAlternaria andCurvularia. The order of dominance of these fungi varied according to the type of grain, the length of storage and the treatment.A. niger was the dominant organism in the control samples of the three grains. It could grow successfully at high moisture contents but above 15 %, it was usually overgrown by other fungi. In wheatP. citrinum andA. sydowii, in cornP. citrinum andA. terreus, and in sorghumA. terreus andA. niger were the dominant fungi at moisture contents above 15 % after four months storage at 30 C. When stored at 8 CPenicillium species tended to predominate over other fungi in grains with high moisture contents.In surface-sterilized grains adjusted to 15 % moisture content, inoculated with six dominant fungi separately and incubated at 30 C,A. niger, P. citrinum andP. variabile caused severe deterioration in the three grains;A. ochraceus in wheat only andF. moniliforme in wheat and corn;C. spicifer was slightly injurious to the three grains.  相似文献

Summary Extract from onion bulbs and diffusate from roots of onion seedlings were fractionated by column chromatography. The stimulatory effects of the different fractions of onion extract on sclerotial germination ofSclerotium cepivorum were studied. The sugar fraction was the most stimulatory, whereas, the amino acid fraction was not effective. Paper chromatographic analysis revealed the presence of glucose, fructose and no amino acids in the root diffusate. These two sugars and 13 amino acids were identified in the onion extract. When various sugars and amino acids were supplied individually to autoclaved soil, only glucose, fructose, mannose and maltose effectively induced sclerotial germination. Partial stimulation occured in nonsterile soil amended with high glucose concentrations. Studies on the antibiotic effect of the different fractions against some soil fungi by the spore germination method showed that, the sugar fraction inhibits completely the spore germination of all the fungi, tested, whereas, the amino acid fraction was non-inhibitory. Both fractions did not show antibiotic activity when tested by the filter paper disc method. Attempts to extract inhibitory substances from soil which inhibit sclerotial germination were unsuccessful. It was suggested that onion extract plays a twofold role in stimulating sclerotial germination in natural soil: (a) a direct nutritional influence; (b) an antibiotic effect on soil mycoflora which reduces competition for nutrients.  相似文献

Oospore germination occurred over a temperature ranging of 15–35°C forPythium coloratum, 10–35°C forP. diclinum, 15–30°C forP. dissotocum, 7–30°C forP. monospermum, and 10–30°C forP. pleroticum. Optimum temperature was 25°C for all species tested. In case of pH, oospore germination occurred over a range of 4.76–8.55 with an optimum of 6.40–7.40. The least germination occurred at pH 4.76 forP. coloratum, P. diclinum, P. monospermum andP. pleroticum, whileP. dissotocum germinated from pH 5.02. Oospores of the all tested pythia were able to germinate at –0.13 to –1.65 MPa and could not germinate at –3.40 MPa, with the highest germination rate at –0.27 to –0.47 MPa. The effect of temperature, pH and osmotic potential on oospore germination was discussed in relation to pollution of pond water.  相似文献

Pythium fluminum produced zoospores most abundantly at 15°C, whereas the optima forPythium group F andP. marsipium were 20 and 25°C, respectively. Increasing the incubation temperature above the optimum resulted in the decrease of the duration of zoospore production. InPythium group F the ability to produce zoospores was not lost even after incubation at 40°C for 24 h. On the other hand,P. marsipium andP. fluminum lost the ability under these conditions. Zoospore production was inhibited at pH 4.5 and 10.5 in all the species tested.Pythium fluminum andP. marsipium were found to have two pH optima for zoospore production (7.5 and 9.5 for the former and 5.5 and 8.5 for the latter). The optimum pH for zoospore production byPythium group F was 6.5–7.5. Moderate osmotic potentials (–0.27–0.47 MPa) appeared to favor zoospore production by the pythia tested. The effect of temperature, pH and osmotic potential on zoospore production was discussed in relation to pollution of pond water.  相似文献
