The alpha-(2----8)-linked sialic acid oligosaccharides (NeuAc)n exhibit an unusual degree of heterogeneity in the conformation of their linkages. This was diagnosed by observation in their 13C NMR spectra of an equivalent and unique heterogeneity in the chemical shifts of their anomeric carbons and subsequently confirmed by more comprehensive 1H and 13C NMR studies. In these studies both one-dimensional and two-dimensional experiments were carried out on the trisaccharide (NeuAc)3 and colominic acid. In addition to the unambiguous assignment of the signals in the spectra, these experiments demonstrated that both linkages of (NeuAc)3 differed in conformation from each other and from the inner linkages of colominic acid. The NMR data indicate that these conformational differences extend to both terminal disaccharides of oligosaccharides larger than (NeuAc)5, a result that has considerable physical and biological significance. In the context of the group B meningococcal polysaccharide, it provides an explanation for the conformational epitope of the group B meningococcal polysaccharide, which was proposed on the evidence that (NeuAc)10, larger than the optimum size of an antibody site, was the smallest oligosaccharide able to bind to group B polysaccharide specific antibodies. Because the two terminal disaccharides of (NeuAc)10 differ in conformation to its inner residues, the immunologically functional part of (NeuAc)10 resides in its inner six residues. This number of residues is now consistent with the maximum size of an antibody site.  相似文献

Benzene, an accepted leukemogen, has been suggested to cause other hemato- and lymphopoietic cancers. Here we review the published literature for non-Hodgkin lymphoma (NHL) and occupational exposure to benzene. Six cohorts, sixteen case–control studies and two studies of other designs were identified through keyword searches of bibliographic databases. Twenty-two of twenty-four studies found no association between NHL and ever exposed to benzene compared to never; a random-effects meta-analysis gave a pooled risk estimate of 1.11 (95% confidence interval 0.94–1.30). Our finding of no effect agrees with one of two previous meta-analyses. The other meta-analysis examined if high benzene exposure increased NHL risk but a lack of consistent exposure categories within the same metric should have precluded pooling risks by exposure level. Instead, we reviewed whether dose–response relationships existed. The best available data came from six studies where exposure was estimated from historical measurements and on the whole, no trends in risks of NHL with rising cumulative, average, peak, or duration of benzene exposure were found. NHL is a heterogeneous group of malignancies and although less well-studied, benzene was not associated with any NHL subtype. In conclusion, benzene at either low or high doses does not increase the risk of NHL.  相似文献

The orientation of washed Escherichia coli bacteria in suspension induced by applied a.c. electric fields has been monitored by observing the accompanying changes in the intensity of light scattered by the suspension. The data enable the anisotropy of electrical polarisability deltaalpha to be determined. Changes in deltaalpha due to the addition of various antibiotics to the suspension have been measured as a function of both the antibiotic concentration and the temperature of the suspension. The results are taken to indicate that there is an accumulation of the antibiotic molecules at the bacterial surface.  相似文献

We have derived oligosaccharides from the capsular polysaccharide of type III group B Streptococcus by enzymatic hydrolysis of a specific backbone glycosidic bond utilizing an endo-beta-galactosidase from Flavobacterium keratolyticus. Enzymatic digestion of the polysaccharide produced oligosaccharide fragments of one or more pentasaccharide repeating units. On the basis of 13C NMR, 1H NMR, and methylation analyses, it was established that the smallest digestion fragment was alpha-D-NeupNAc-(2----3)-beta-D-Galp-(1----4)-[beta-D-Glcp-(1----6 )]- beta-D-GlcpNAc-(1----3)-beta-D-Gal. The isolation of this oligosaccharide is consistent with the susceptibility of the beta-D-Galp-(1----4)-beta-D-Glcp linkage in the backbone of the type III group B streptococcal polysaccharide and confirms that the polysaccharide is composed of a pentasaccharide repeating unit. High resolution 13C NMR spectroscopic studies indicated that, as in the case of the pentasaccharide, the terminal sialic acid residues of the type III group B streptococcal polysaccharide were linked to O-3 and not to O-6 of its branch beta-D-galactopyranosyl residues as had been previously reported (Jennings, H. J., Rosell, K.-G., and Kasper, D. L. (1980) Can. J. Chem. 58, 112-120). This linkage was confirmed in an independent methylation analysis of the type III group B streptococcal polysaccharide. Thin layer chromatogram binding assay and radioactive antigen binding assays with radiolabeled oligosaccharides demonstrated the single repeating unit pentasaccharide oligosaccharide to be poorly antigenic. Increasing oligosaccharide size to a decasaccharide consisting of two repeating units resulted in an 8-fold increase in antigen binding in the direct radioactive antigen binding assay. The results suggest that a region of the immunodeterminant site critical for antibody binding is located in the backbone of the polysaccharide and involves the beta-D-galactopyranose-(1----4) beta-D-glucopyranose bond.  相似文献

Hemodynamic effects of anti-G suit inflation in a 1-G environment   总被引:1,自引:0,他引:1  
This study evaluated effects of various anti-G inflation pressures on cardiac volumes and the relationship of these volume changes to mean arterial pressure changes. Ventricular volumes were calculated using two-dimensional echocardiography. An anti-G suit was inflated to 2, 4, and 6 psi in the standing and supine positions for 10 male subjects. In the supine position, mean arterial pressure increased from base line for all three inflation pressures (P = 0.05). The end-diastolic volume increased after 2-psi inflation (P = 0.03). Cardiac output or stroke volume did not change. After standing, mean arterial pressure (P = 0.002), end-diastolic volume (P = 0.002), and stroke volume (P = 0.05) fell after suit deflation. Peripheral vascular resistance fell in the 2- and 4-psi inflation profiles. In the standing protocol, mean arterial pressure, end-diastolic volume, stroke volume, and cardiac output rose with all three inflation pressures (P less than 0.05). After reclining, heart rate increased (P = 0.02) and mean arterial pressure fell (P less than 0.05) in the 4- and 6-psi inflation profiles after suit deflation. Increases in mean arterial pressure are caused by increases in cardiac preload and cardiac output after inflation of the anti-G suit while subjects were standing. Increased cardiac preload was not consistently seen after inflation while subjects were supine. Changes in end-diastolic volume and mean arterial pressure were dependent on the pressure used to inflate the anti-G suit.  相似文献

Conclusion Though relatively little direct attention has been given by Marxist anthropologists to a theory of individual behavior and thought in relation to societal processes, the above partial summary indicates the wealth of insight that is available for elaborating such a theory. In addition, there are of course significant developments in the field of psychology, notably the attention to activity as central to individual personality [], the renewed concern with levels of integration theory [], and the burgeoning interest in Vygotsky's writings on language, thought, and culture []. There are also the continuing attempts to locate Freud's positive contributions in a historical materialist frame [].Where, then, is psychological anthropology? As always, producing a richness of suggestive materials — such as those on varying conceptions of the self — but, as a glance at the pages of Ethos will show, unfortunately not engaged in fundamental theoretical innovation. By way of illustration, let me cite Spindler's The Making of Psychological Anthropology, a collection of articles by major figures, past and present. Spindler's introductory essay is thoughtful; always the teacher, he presents with consideration and modesty the history of the field, the disfavor into which it fell (in large part due to the methodlogical travesties of national character studies), and its stubborn persistence (following from the pervasive interest in the psychological dimension that has always characterized cultural anthropology). He summarizes ongoing problems as perceived from within the field, and as the major difficulty to be overcome pinpoints cultural overdeterminism and its inadequacy for explaining variations. The solution? Not the necessity of respecting history and focussing on how individuals variously understand and relate (according to their individual histories) to the established structures whereby particular societies produce, allocate, and consume basic goods, and how they variously respond to disjunctions in these structures even as they reproduce them. Instead Spindler writes, ... if we are to escape the double bind of our cultural overdeterminism, we are going to have to go beyond culture and even ecology, to biochemistry, to physiology and neurology, to genetics — to biology in the broadest sense of the term [].In closing, let me say to the reader, do not simply turn away in dismay. One must conclude that it is imperative to continue to build a solid alternative theory of relations among individual behaviors, individual understandings, cultural values, and societal processes, or, in other words, to replace a non-historical and essentially biological paradigm with a dialectical and materialist view of human action.Eleanor Leacock is Professor of Anthropology and Chair at the City College, City University of New York.
