Key message

The drought tolerance in young oil palm plants is related to greater efficiency in preventing oxidative damage by activating enzymatic and non-enzymatic antioxidant strategies simultaneously.

Abstract

Drought is a major environmental constraint limiting growth and yield of oil palm trees. In this study, two oil palm hybrids (BRS Manicoré and BRS C 2501) were grown in large containers and subjected to a water deficit during 57 days. Leaf gas exchange analysis was combined with an in-depth assessment of the antioxidant system over the drought imposition. Under drought, leaf water potential at predawn (Ψ pd) decreased similarly in both hybrids. In parallel, there were decreases in the net CO2 assimilation rate (A), chlorophyll concentrations and Rubisco total activity. Overall, these decreases were more pronounced in BRS C 2501 than in BRS Manicoré. BRS C 2501 plants triggered more markedly its enzymatic antioxidant system earlier (Ψ pd = ?2.1 MPa) than did BRS Manicoré, but these responses were accompanied by higher concentrations of H2O2 and malondialdehyde in BRS C 2510 than in BRS Manicoré. With the progress of drought stress (Ψ pd = ?2.9 MPa and below), BRS Manicoré was better able to cope with oxidative stress through a more robust antioxidant system. In addition, significant decreases in drought-induced NAD+-malate dehydrogenase activities were only observed in stressed BRS C 2501 plants. Regardless of watering regimes, the total carotenoid, ascorbate and glutathione concentrations were higher in BRS Manicoré than in BRS C 2501. In conclusion, BRS Manicoré is better able to tolerate drought than BRS C 2501 by triggering multiple antioxidant strategies involved both in reactive oxygen species scavenging and dissipation of excess energy and/or reducing equivalents particularly under severe drought stress.

In earlier-maturing coffee clones, owing to the shorter time required for fruit filling and ripening, photo-assimilates should be transported to fruits in a period shorter than that found in intermediate- or late-maturing clones. We hypothesised that at a given source-to-sink ratio, a presumably greater sink strength in early-maturing clones relative to intermediate- and late-maturing individuals should be correlated to increased rate of net carbon assimilation (A) and greater photo-assimilate transport to the fruits. Overall, earlier-maturing clones displayed greater A rates than the intermediate-maturing clones, which, in turn, had higher A than their late counterparts. Changes in A were largely associated with changes in stomatal conductance. Only marginal alterations occurred in the internal-to-ambient CO2 concentration ratio, the carbon isotope composition ratio, soluble sugars and chlorophyll a fluorescence parameters. Some changes in starch pools were detected among treatments. To the best of our knowledge, this is the first report showing evidence that increased precociousness of fruit growth and maturation results in higher A and thus increased source strength, a fact associated to a large degree with higher stomatal aperture.  相似文献

Background

Beef cows herd accounts for 70% of the total energy used in the beef production system. However, there are still limited studies regarding improvement of production efficiency in this category, mainly in developing countries and in tropical areas. One of the limiting factors is the difficulty to obtain reliable estimates of weight variation in mature cows. This occurs due to the interaction of weight of maternal tissues with specific physiological stages such as pregnancy. Moreover, variation in gastrointestinal contents due to feeding status in ruminant animals is a major source of error in body weight measurements.

Objectives

Develop approaches to estimate the individual proportion of weight from maternal tissues and from gestation in pregnant cows, adjusting for feeding status and stage of gestation.

Methods and Findings

Dataset of 49 multiparous non-lactating Nellore cows (32 pregnant and 17 non-pregnant) were used. To establish the relationships between the body weight, depending on the feeding status of pregnant and non-pregnant cows as a function of days of pregnancy, a set of general equations was tested, based on theoretical suppositions. We proposed the concept of pregnant compound (PREG), which represents the weight that is genuinely related to pregnancy. The PREG includes the gravid uterus minus the non-pregnant uterus plus the accretion in udder related to pregnancy. There was no accretion in udder weight up to 238 days of pregnancy. By subtracting the PREG from live weight of a pregnant cow, we obtained estimates of the weight of only maternal tissues in pregnant cows. Non-linear functions were adjusted to estimate the relationship between fasted, non-fasted and empty body weight, for pregnant and non-pregnant cows.

Conclusions

Our results allow for estimating the actual live weight of pregnant cows and their body constituents, and subsequent comparison as a function of days of gestation and feeding status.  相似文献

This study aimed to evaluate under photoautotrophic conditions the effect of CO2-enriched atmosphere (360 or 1,000 μmol CO2 mol?1 air) combined with two substrate types (agar or Florialite®) in vitro on plants of Pfaffia glomerata, an endangered medicinal species with promising applications in phytotherapy and phytomedicine. The effects of the treatments on the growth, stomatal density, Rubisco activity, carbon isotopic discrimination, metabolite accumulation, photosynthetic pigments and ultrastructural characteristics were investigated. After a 35-day cultivation period, the in vitro-growth of P. glomerata nodal segments under the different treatments resulted in plants with substantial differences in relation to their growth, photosynthetic pigments, stomatal density and leaf ultrastructural characteristics. The enrichment with CO2 coupled with a porous substrate increased the growth of P. glomerata. The stomatal density in the abaxial epidermis more than doubled in response to the high CO2 supply in both supporting types, whereas the Rubisco activity and activation state were both unresponsive to the treatments. Regardless of the CO2 supply, the plants grown in agar displayed higher carbon isotope discrimination than their counterparts grown in Florialite®. We propose that the long-term photosynthetic performance was improved using Florialite® as a growth support in combination with a high CO2 supply. No apparent signs of photosynthetic down-regulation could be found under elevated CO2 conditions. The enrichment of in vitro atmospheres with CO2 coupled with a porous substrate offers new possibilities for improving the growth and production on a commercial scale of high morphological and physiological quality Pfaffia plants.  相似文献

The objective of this study was to evaluate ether extract (EE) concentrations, pigments, and wax in forages (n = 14), concentrates (n = 23), and cattle feces (n = 100) using extraction methods recommended by Association of Official Analytical Chemists (AOAC; method 920.39) and American Oil Chemist's Society (AOCS; method Am 5-04). The EE contents were compared by adjusting a linear regression model for each sample type. For the feces and forage samples, the EE contents produced by the AOCS method were greater (P<0.05) compared to those obtained using the AOAC method. No differences between methods were observed in EE content of concentrates (P>0.05). Concentration of vegetable pigments and wax were evaluated by using analysis for variance. Vegetable pigments were lower (P<0.05) in the post-extraction residues using the AOCS method, than the AOAC method, indicating greater participation of vegetable pigments in the EE. No differences were observed between the methods in wax concentration of the post-extraction residues (P>0.05). The quantification method of the EE content that is recommended by AOCS is not suggested for analyses of forage and feces of ruminants because it possibly increases the removal of non-fatty material, mostly pigments, in comparison to the method recommended by AOAC.  相似文献

Determination of the energetic contribution of different chemical components of cattle diets based on the content of total digestible nutrients (TDN) has been suggested by several authors and was adopted by the NRC [National Research Council (NRC), 2001. Nutrient Requirements of Dairy Cattle, seventh ed. Academic Press, Washington]. The main objective of that proposal is to overcome the high costs and the extended time of traditional in vivo digestion assays. Although theoretically based, the summative system adopted by the NRC [National Research Council (NRC), 2001. Nutrient Requirements of Dairy Cattle, seventh ed. Academic Press, Washington] has been shown to be inaccurate in predicting the TDN content of individual feeds or diets under tropical conditions. Therefore, a new model, that also uses a summative approach, was developed but it was based on data obtained in the tropics. The new model consists of equations that predict the digestible fractions of ether extract (n = 108), crude protein (n = 93), non-fibrous carbohydrates (n = 84), and neutral detergent fibre (n = 164). Non-fibrous components were evaluated by the Lucas [Lucas, H.L., 1960. Relations Between Apparent Digestibility and the Composition of Feed and Feces. 1: A Quantitative Theory. North Caroline State College, Raleigh] test to obtain true digestibility coefficients, and by the factorial system to distinguish between truly digested and metabolic fractions. Fibre components were evaluated using an adaptation of the surface law to obtain the indigestible fraction and a meta-analysis approach to obtain digestibility coefficients of the potentially digestible fraction. The estimates obtained with the new model were validated and compared to those produced with the model adopted by the NRC [National Research Council (NRC), 2001. Nutrient Requirements of Dairy Cattle, seventh ed. Academic Press, Washington] for cattle managed under tropical conditions. Data from four trials, two with lactating cows (n = 63) and two with growing and finishing cattle (n = 44) were used. The estimates produced with the two models were compared based on the decomposition of the mean square prediction error. It can be concluded that the new model was more accurate in predicting TDN values than the NRC [National Research Council (NRC), 2001. Nutrient Requirements of Dairy Cattle, seventh ed. Drought tolerance was examined in two oil palm hybrids (BRS Manicoré and BRS C 2501) grown in large pots and subjected to long-term drought (57 days) and rehydration. Regardless of hybrids, predawn water potential (Ψ pd), net photosynthesis rates, and stomatal conductance decreased similarly upon drought imposition, but the absolute values of these variables were lower in BRS C 2501 than in BRS Manicoré. Overall, drought-induced decreases in activities of key enzymes associated with carbon metabolism, including Rubisco, ADP-glucose pyrophosphorylase, and sucrose-phosphate synthase, were stronger in BRS C 2501 than in BRS Manicoré. Our data suggest that synthesis of starch (and possibly sucrose) was fundamentally limited by a lower substrate availability rather than by enzymatic constraints, particularly until day 34 after suspending watering. Drought stress provoked similar decreases in biomass accumulation in either hybrid; however, BRS Manicoré plants displayed higher total leaf area and root length coupled with greater relative decreases in dry matter of above-ground parts than of roots as compared to BRS C 2501, thereby leading to an increased root-to-above ground ratio in the former. Upon resuming irrigation, the slow recovery of Ψ pd preceded the recovery of stomatal conductance and photosynthesis irrespective of hybrid. BRS Manicoré was better able to recover enzyme activities and carbohydrate status than BRS C 2501. In conclusion, both hybrids could tolerate (or survive) the severe drought conditions, but, overall, BRS Manicoré plants were better able to adjust its physiological, morphological, and biochemical traits to cope with drought than did BRS C 2501.  相似文献

These experiments were carried out to evaluate, using in vitro and in situ techniques, the effects of three inclusion levels of calcium oxide (0, 5, and 10 g/kg of sugarcane fresh matter) and four exposure times (0, 24, 48, and 72 h) of sugarcane to calcium oxide on the chemical composition and digestive kinetic parameters of sugarcane. The treatments were arranged in a 3 by 4 factorial design. Freshly-cut sugarcane (whole plant) was treated with calcium oxide and separated into 12 piles inside a barn to prevent direct exposure to sunlight, rain, and wind. Every day, before and after animal feeding, the calcium oxide was proportionally hand-mixed with approximately 150 kg of freshly-cut sugarcane to make up the dietary treatments. The lowest (Ti) and greatest (Ts) temperature and pH of the treated sugarcane piles were measured immediately before and after sampling, respectively. The ether extract (EE) and DM were not affected (P>0.05) by either exposure time or inclusion level. However, CP increased linearly (P=0.01) and OM decreased linearly (P<0.0001) as the exposure time and calcium oxide inclusion level increased. Interactions between inclusion level and exposure time on DM, OM, CP, EE, Ti, and Ts were not observed. However, significant interactions were detected for non-fibre carbohydrate (NFC), neutral detergent fibre (aNDF), and pH. A quadratic effect of exposure time on the Ti and Ts was observed (P=0.001 and P=0.001, respectively). The maximum temperature was reached with approximately 51 h of exposure time. Calcium oxide positively affected the insoluble potentially digestible fraction of sugarcane DM and aNDF (P=0.001 and P=0.001, respectively), and the indigestible fraction of sugarcane aNDF (P=0.001). Interactions between inclusion level and exposure time on soluble and indigestible fractions of sugarcane DM (P=0.0001 and P=0.01, respectively) were found. However, no interactions (P>0.27) were found between inclusion level and exposure time on aNDF digestive kinetic parameters. The fractional digestion rate (kd) of sugarcane DM and aNDF was not influenced by treatments (P>0.05). The mean values of kd for sugarcane DM and aNDF were 0.0235 and 0.0215/h, respectively. The gas production kinetics parameters were not affected (P>0.05) by treatments. In conclusion, the inclusion of calcium oxide improved the in situ potentially digestible fraction of sugarcane DM and aNDF; however, it did not influence the fractional digestion rate. No effects were observed on the in vitro digestive kinetic parameters.  相似文献
