Background
The Eurasian grapevine (Vitis vinifera L.) is the most widely cultivated and economically important horticultural crop in the world. As a one of the origin area, Anatolia played an important role in the diversification and spread of the cultivated form V. vinifera ssp. vinifera cultivars and also the wild form V. vinifera ssp. sylvestris ecotypes. Although several biodiversity studies have been conducted with local cultivars in different regions of Anatolia, no information has been reported so far on the biochemical (organic acids, sugars, phenolic acids, vitamin C) and antioxidant diversity of local historical table V. vinifera cultivars grown in Igdir province. In this work, we studied these traits in nine local table grape cultivars viz. ‘Beyaz Kismis’ (synonym name of Sultanina or Thompson seedless), ‘Askeri’, ‘El Hakki’, ‘Kirmizi Kismis’, ‘Inek Emcegi’, ‘Hacabas’, ‘Kerim Gandi’, ‘Yazen Dayi’, and ‘Miskali’ spread in the Igdir province of Eastern part of Turkey.Results
Variability of all studied parameters is strongly influenced by cultivars (P < 0.01). Among the cultivars investigated, ‘Miskali’ showed the highest citric acid content (0.959 g/l) while ‘Kirmizi Kismis’ produced predominant contents in tartaric acid (12.71 g/l). The highest glucose (16.47 g/100 g) and fructose (15.55 g/100 g) contents were provided with ‘Beyaz Kismis’. ‘Kirmizi Kismis’ cultivar had also the highest quercetin (0.55 mg/l), o-coumaric acid (1.90 mg/l), and caffeic acid (2.73 mg/l) content. The highest ferulic acid (0.94 mg/l), and syringic acid (2.00 mg/l) contents were observed with ‘Beyaz Kismis’ cultivar. The highest antioxidant capacity was obtained as 9.09 μmol TE g-1 from ‘Inek Emcegi’ in TEAC (Trolox equivalent Antioxidant Capacity) assay. ‘Hacabas’ cultivar had the highest vitamin C content of 35.74 mg/100 g.Conclusions
Present results illustrated that the historical table grape cultivars grown in Igdir province of Eastern part of Turkey contained diverse and valuable sugars, organic acids, phenolic acids, Vitamin C values and demonstrated important antioxidant capacity for human health benefits. Further preservation and use of this gene pool will be helpful to avoid genetic erosion and to promote continued agriculture in the region. 相似文献2.

The aim of this study was to determine the effects of raffinose and hypotaurine on sperm parameters after the freeze–thawing of Merino ram sperm. Totally 40 ejaculates of five Merino ram were used in the study. Semen samples, which were diluted with a Tris-based extender containing 10 mM raffinose, 5 mM hypotaurine, 5 mM raffinose +2.5 mM hypotaurine (H + R) and no antioxidant (control), were cooled to 5 °C and frozen in 0.25 ml French straws and stored in liquid nitrogen. This study was conducted to determine the effects of methionine, inositol and carnitine on sperm (motility, abnormality, DNA integrity and in vivo fertility) and oxidative stress parameters (lipid peroxidation, total glutathione and antioxidant potential levels) of bovine semen after the freeze–thawing process. Nine ejaculates, collected with the aid of an artificial vagina twice a week from each Simmental bovine, were included in the study. Each ejaculate, splitted into seven equal groups and diluted in Tris-based extender containing methionine (2.5 and 7.5 mM), carnitine (2.5 and 7.5 mM), inositol (2.5 and 7.5 mM) and no additive (control), was cooled to 5 °C and then frozen in 0.25 ml straws. Frozen straws were then thawed individually at 37 °C for 20 s in a water bath for the evaluation.The extender supplemented with 7.5 mM doses of carnitine and inositol led to higher subjective motility percentages (61.9 ± 1.3% and 51.3 ± 1.6%) compared to the other groups. The addition of methionine and carnitine at doses of 2.5 and 7.5 mM and inositol at doses of 7.5 mM provided a greater protective effect in the percentages of total abnormality in comparison to the control and inositol 2.5 mM (P < 0.001). As regards CASA motility, 7.5 mM carnitine (41.6 ± 2.9% and 54.2 ± 4.9%) and inositol (34.9 ± 2.0% and 47.3 ± 2.2%) caused insignificant increases in CASA and total motility in comparison to the other groups. All of the antioxidants at 2.5 and 7.5 mM resulted in lower sperm with damaged DNA than that of control, thus reducing the DNA damage (P < 0.05). No significant differences were observed in CASA progressive motility and sperm motion characteristics among the groups. In fertility results based on 59-day non-returns, no significant differences were observed in non-return rates among groups. As regards biochemical parameters, supplementation with antioxidants did not significantly affect LPO and total GSH levels in comparison to the control group (P > 0.05). The maintenance of AOP level in methionine 2.5 mM was demonstrated to be higher (5.06 ± 0.38 mM) than that of control (0.96 ± 0.29 mM) following the freeze–thawing (P < 0.001). Supplementation with these antioxidants prior to the cryopreservation process protected the DNA integrity against the cryodamage. Furthermore, future research should focus on the molecular mechanisms of the antioxidative effects of the antioxidants methionine, carnitine and inositol during cryopreservation. 相似文献

A response surface methodology (RSM) experimental design was applied for improving micropropagation of a wild apricot, Prunus armeniaca Lam., from the mountains of Kazakhstan. In an initial study, woody plant medium (WPM) mineral nutrients [calcium nitrate, ammonium nitrate, mesos (calcium chloride, potassium phosphate and magnesium sulfate) potassium sulfate and minor nutrients] were tested in a response surface methodology (RSM) experiment. Shoot quality was the best when nitrogen and mesos (CaCl2, MgSO4, K2SO4, KH2PO4) compounds were altered. In this study an expanded mesos optimization experiment was run. Data taken included a subjective quality rating, shoot length, shoot number, leaf color and size, callus and physiological disorders. Data were analyzed by Classification and Regression Tree Analysis (CART), a data mining technique that provides specific cutoff values for data and easy to interpret data trees. The CART analysis indicated that the best quality would be with ≤2.4× WPM levels of KH2PO4 and ≤0.75× MgSO4. Shoot length was affected by K2SO4, but most shoots were of good size at any concentration. Shoot multiplication was affected by KH2PO4, but there were >5 shoots at any concentration. Leaf color was best with ≤2.41× KH2PO4 and ≤1.22× K2SO4. Based on the CART analysis, a recommendation for improved mesos compounds was developed. Each of the individual trees was analyzed and the cutoff points determined so that all the growth characteristics could be considered in the final concentrations chosen. Using the combined results from the CART analysis, the suggested medium would include WPM with CaCl2 2.7×, MgSO4 2.7×, K2SO4 0.8×, KH2PO4 0.75×. 相似文献

Soil quality and health are terms describing similar concepts, but the latter appeals to farmers and crop consultants as part of a holistic approach to soil management. We regard soil health as the integration and optimization of the physical, biological and chemical aspects of soils for improved productivity in an economic and sustainable manner. This paper describes the process used for the selection of soil quality/health indicators that comprise the new Cornell Soil Health Test. Over 1,500 samples collected from controlled research experiments and commercial farms were initially analyzed for 39 potential soil quality indicators. Four physical and four biological indicators were selected based on sensitivity to management, relevance to functional soil processes, ease and cost of sampling, and cost of analysis. Seven chemical indicators were also selected as they are part of the standard soil nutrient test. Soil health test reports were developed to allow for an overall assessment, as well as the identification of specific soil constraints. The new soil health test is being offered on a for-fee basis starting in 2007. In addition, visible near infrared reflectance spectroscopy was evaluated as a possible tool for low-cost soil health assessment. From preliminary analyses, the methodology shows promise for some but not all of the soil quality indicators. In conclusion, an inexpensive soil health test was developed for integrative assessment of the physical, biological, and chemical aspects of soils, thereby facilitating better soil management. 相似文献

The assumption of Hardy-Weinberg equilibrium (HWE) among alleles in a nonevolving population is of fundamental importance
in genetic studies. Deviation from HWE in a population usually indicates inbreeding, stratification and sometimes problems
in genotyping. In populations of affected individuals, these deviations can also provide evidence for association. In this
paper, we introduce a measure based on the Kullback-Leibler discrimination information function that quantifies the deviation
from HWE in a population. We use this measure to order populations. We also propose a test for HWE based on an estimate of
this measure. The test is a statistically consistent test of the null hypothesis for all alternatives and is very easy to
implement. Our proposed test statistic is compared with an earlier, widely used, test. Finally, the use of the proposed new

The new species Nonea dumanii, endemic to the mountains of the western Taurus in south Turkey (C4 Antalya), is described an illustrated based on original collections by the authors. Karyological observations and analysis of ITS1 DNA sequences showed that the species is probably hexaploid with 2n = 6x = 60 and has phylogenetic affinity to the diploid N. monticola from the Paphlagonian mountains as well as to the tetraploid N. anchusoides from northwest Iran and southeast Turkey. From these allopatric species it is morphologically distinct in characters of the indumentum, flower and fruit. Polyploidy is supported as a major driving force for speciation in Nonea, especially in the group of Anatolian mountain species with primary base number x = 10. 相似文献
