Our recent EPR and EXAFS experiments investigating the structure of the oxygen-evolving complex of PS II are discussed. PS II treatments which affect the cofactors calcium and chloride have been used to poise samples in modified forms of the S-states, S1, S2 and S3. X-ray absorption studies indicate a similar overall structure for the manganese complex between treated and native samples although the influence of the treatments and cofactors is observed. Manganese oxidation (or oxidation of a ligand to the manganese cluster) is indicated to occur on each of the transitions S1 S2 and S2 S3 in these modified samples. The cluster appears to contain at least two inequivalent Mn-Mn pairs. In the native samples the Mn-Mn distance is 2.7 Å, but in samples where the calcium site is affected, one of the pairs has a 3.0 Å Mn-Mn distance. The intensity of the 3.3/3.6 Å interaction is reduced on sodium chloride treatment (calcium depletion) perhaps indicating calcium binding close to the manganese cluster. From EPR data we also propose that treatments which affect calcium and chloride binding cause a modification of the native S2 state, slow the reduction of Yz and allow an S3 EPR signal to be observed following illumination. Common (Cinnamomum verum, C. zeylanicum) and cassia (C. aromaticum) cinnamon have a long history of use as spices and flavouring agents. A number of pharmacological and clinical effects have been observed with their use. The objective of this study was to systematically review the scientific literature for preclinical and clinical evidence of safety, efficacy, and pharmacological activity of common and cassia cinnamon. Using the principles of evidence-based practice, we searched 9 electronic databases and compiled data according to the grade of evidence found. One pharmacological study on antioxidant activity and 7 clinical studies on various medical conditions were reported in the scientific literature including type 2 diabetes (3), Helicobacter pylori infection (1), activation of olfactory cortex of the brain (1), oral candidiasis in HIV (1), and chronic salmonellosis (1). Two of 3 randomized clinical trials on type 2 diabetes provided strong scientific evidence that cassia cinnamon demonstrates a therapeutic effect in reducing fasting blood glucose by 10.3%-29%; the third clinical trial did not observe this effect. Cassia cinnamon, however, did not have an effect at lowering glycosylated hemoglobin (HbA1c). One randomized clinical trial reported that cassia cinnamon lowered total cholesterol, low-density lipoprotein cholesterol, and triglycerides; the other 2 trials, however, did not observe this effect. There was good scientific evidence that a species of cinnamon was not effective at eradicating H. pylori infection. Common cinnamon showed weak to very weak evidence of efficacy in treating oral candidiasis in HIV patients and chronic salmonellosis.  相似文献

The dynamics of phenolic galloylglucoses (di-, tri-, tetra- and penta-galloylglucose), flavonoids (quercitin and quercitin glycosides) and sideroxylonal were compared with that of xanthophyll cycle-dependent energy dissipation during rapid induction of chilling-dependent photo-inhibition. Pre-dawn xanthophyll cycle engagement of seedlings of Eucalyptus nitens transferred from mild nursery conditions to a low temperature controlled environment increased logarithmically during eight days of treatment. Photochemical efficiency and flavonoids decreased after four days of treatment and non-photochemical quenching after two days of treatment. Galloylglucoses and sideroxylonal decreased linearly during treatment. These results demonstrate that rapid changes in foliar phenolic levels are associated with abrupt changes in the plant environment. It is argued that under these growth-chamber conditions, the xanthophyll cycle facilitated dissipation of excess light energy, lessening the requirement for the dissipation of energy or antioxidant activity through phenolic metabolites.  相似文献

Background

Selenium is a natural health product widely used in the treatment and prevention of lung cancers, but large chemoprevention trials have yielded conflicting results. We conducted a systematic review of selenium for lung cancers, and assessed potential interactions with conventional therapies.

Methods and Findings

Two independent reviewers searched six databases from inception to March 2009 for evidence pertaining to the safety and efficacy of selenium for lung cancers. Pubmed and EMBASE were searched to October 2009 for evidence on interactions with chemo- or radiation-therapy. In the efficacy analysis there were nine reports of five RCTs and two biomarker-based studies, 29 reports of 26 observational studies, and 41 preclinical studies. Fifteen human studies, one case report, and 36 preclinical studies were included in the interactions analysis. Based on available evidence, there appears to be a different chemopreventive effect dependent on baseline selenium status, such that selenium supplementation may reduce risk of lung cancers in populations with lower baseline selenium status (serum<106 ng/mL), but increase risk of lung cancers in those with higher selenium (≥121.6 ng/mL). Pooling data from two trials yielded no impact to odds of lung cancer, OR 0.93 (95% confidence interval 0.61–1.43); other cancers that were the primary endpoints of these trials, OR 1.51 (95%CI 0.70–3.24); and all-cause-death, OR 0.93 (95%CI 0.79–1.10). In the treatment of lung cancers, selenium may reduce cisplatin-induced nephrotoxicity and side effects associated with radiation therapy.

Conclusions

Selenium may be effective for lung cancer prevention among individuals with lower selenium status, but at present should not be used as a general strategy for lung cancer prevention. Although promising, more evidence on the ability of selenium to reduce cisplatin and radiation therapy toxicity is required to ensure that therapeutic efficacy is maintained before any broad clinical recommendations can be made in this context.  相似文献

We investigated the effects of whole-plant nutrient-availability on the degree of distribution of some plant primary and secondary (nitrogen, fibre, flavonols, gallotannins and cineole) chemicals across young, mature and old leaves of seedlings of Eucalyptus nitens. Four treatments that ranged from low to high nutrient-application rates resulted in mean whole-plant foliar concentrations of 0.63%, 0.85%, 1.11% and 1.82% nitrogen dry matter (N%DM) for treatments A, B, C and D, respectively. Within-plant distribution (across the leaf age profile of young, mature and old leaves within a eucalypt seedling) of N%DM ranged from zero in treatment A to a wide range of distribution in treatment D (low N%DM concentrations in old leaves to high N%DM concentrations in young leaves). Similarly, the distribution of fibre ranged from zero in treatment A to a wide range of distribution in treatment D, but with high concentrations in old leaves and low concentrations in young leaves. In contrast, flavonols (weakly) and gallotannins had a wide range of distribution in treatment A (low concentrations in old leaves to high concentrations in young leaves) but were little or not distributed in the other treatments. Again in contrast, cineole was strongly distributed between old and young leaves (low concentrations in old leaves to high concentrations in young leaves) across all treatments while concentrations in mature leaves reflected one or other leaf age depending on treatment. Protein precipitable phenols in treatments A, B and C were high in young, and low in old leaves; whereas in treatment D they were low and similar between leaves of different ages.  相似文献

Does biochar influence soil physical properties and soil water availability?   总被引:10,自引:0,他引:10  

Aims

This study aims to (i) determine the effects of incorporating 47 Mg ha?1 acacia green waste biochar on soil physical properties and water relations, and (ii) to explore the different mechanisms by which biochar influences soil porosity.

Methods

The pore size distribution of the biochar was determined by scanning electron microscope and mercury porosimetry. Soil physical properties and water relations were determined by in situ tension infiltrometers, desorption and evaporative flux on intact cores, pressure chamber analysis at ?1,500 kPa, and wet aggregate sieving.

Results

Thirty months after incorporation, biochar application had no significant effect on soil moisture content, drainable porosity between –1.0 and ?10 kPa, field capacity, plant available water capacity, the van Genuchten soil water retention parameters, aggregate stability, nor the permanent wilting point. However, the biochar-amended soil had significantly higher near-saturated hydraulic conductivity, soil water content at ?0.1 kPa, and significantly lower bulk density than the unamended control. Differences were attributed to the formation of large macropores (>1,200 μm) resulting from greater earthworm burrowing in the biochar-amended soil.

Conclusion

We found no evidence to suggest application of biochar influenced soil porosity by either direct pore contribution, creation of accommodation pores, or improved aggregate stability.  相似文献
