Efforts have been made to improve the biological stability of phosphodiester (PO) oligonucleotides by the addition of various modifications to either the 3', 5' or both the 3' and 5' ends of an oligonucleotide. ISIS 1080, a phosphorothioate (PS) 21-mer oligonucleotide complementary to the internal AUG codon of UL13 mRNA in HSV-1, reduces the infectious yield of HSV-1 in HeLa cells to 9.0% +/- 11%. PO analogs of ISIS 1080 containing three PS linkages placed on the 3' (ISIS 1365), 5' (ISIS 1370), both the 3' and 5' (ISIS 1364) ends or with four linkages in the middle (ISIS 1400) demonstrated reduced antiviral efficacy compared to fully PS ISIS 1080. Thermal denaturation profiles demonstrated that these oligonucleotides hybridized to complementary DNA or RNA with equivalent binding affinities. All were able to support E. coli RNAse H cleavage of the HSV mRNA to which they were targeted. The stability of the congeners in cell culture medium containing 10% fetal calf serum (FCS), HeLa cytosolic extract, HeLa nuclear extract and in intact HeLa cells revealed that ISIS 1080 was most resistant to nucleolytic digestion through 48 hours. Partial PS oligonucleotides exhibited increased degradation compared to the fully thioated oligonucleotide by exonuclease activity in FCS and endonuclease activity in cell extracts or intact cells. Thus, the reduced efficacy of partial compared to fully PS oligonucleotides against HSV-1 in HeLa cells may result from increased degradation of the mixed PO/PS oligonucleotides. 相似文献

This paper challenges the view of Judith K. Brown (1961) and Peter Kloos (1969) that matrilocal residence tends to cause girls' puberty rites. It shows how improbable this hypothesis is for western North America, where data are plentiful, and suggests that it is unlikely for the rest of the world. Girls' puberty rites seem to be earlier in most localities because of much greater world frequency and dominance among hunters, gatherers, and fishers, while matrilocal residence appears mainly to have stemmed from female hand farming at a later date. Government‐funded flow response monitoring and modelling programmes (flow science) provided by the New South Wales Office of Water (NOW) have supported water resource management since 1997. Flow science has a core technical component defined by hypothesis‐driven long‐term monitoring and analysis, but it also represents many activities that support committees involved in environmental flow management. This is done through collaborations and contracting and has fostered considerable research and analysis into flow ecology, including modelling for the recent Murray–Darling Basin Plan. We describe the performance of environmental flows against legislated wetland objectives to improve wetland function and diversity using flow science. On‐ground monitoring at wetland sites has largely ceased but the flow science done so far indicates that the environmental flow rules written into Water Sharing Plans improve wetland diversity and function. Determination of the long‐term flow needs of NSW wetlands, including how well current Water Sharing Plans aid the delivery of environmental flows, requires finding the means to build on current flow science knowledge from across Australia. 相似文献

Background
The duration of sounds can affect the perceived duration of co-occurring visual stimuli. However, it is unclear whether this is limited to amodal processes of duration perception or affects other non-temporal qualities of visual perception.Methodology/Principal Findings
Here, we tested the hypothesis that visual sensitivity - rather than only the perceived duration of visual stimuli - can be affected by the duration of co-occurring sounds. We found that visual detection sensitivity (d’) for unimodal stimuli was higher for stimuli of longer duration. Crucially, in a cross-modal condition, we replicated previous unimodal findings, observing that visual sensitivity was shaped by the duration of co-occurring sounds. When short visual stimuli (∼24 ms) were accompanied by sounds of matching duration, visual sensitivity was decreased relative to the unimodal visual condition. However, when the same visual stimuli were accompanied by longer auditory stimuli (∼60–96 ms), visual sensitivity was increased relative to the performance for ∼24 ms auditory stimuli. Across participants, this sensitivity enhancement was observed within a critical time window of ∼60–96 ms. Moreover, the amplitude of this effect correlated with visual sensitivity enhancement found for longer lasting visual stimuli across participants.Conclusions/Significance
Our findings show that the duration of co-occurring sounds affects visual perception; it changes visual sensitivity in a similar way as altering the (actual) duration of the visual stimuli does. 相似文献7.

BackgroundHypoglycemic effects of grapefruit juice are well known but the effects of naringin, its main flavonoid on glucose intolerance and metabolic complications in type 1 diabetes are not known.ObjectivesTo investigate the effects of naringin on glucose intolerance, oxidative stress and ketonemia in type 1 diabetic rats.MethodsSprague-Dawley rats divided into 5 groups (n = 7) were orally treated daily with 3.0 ml/kg body weight (BW)/day of distilled water (group 1) or 50 mg/kg BW of naringin (groups 2 and 4, respectively). Groups 3, 4 and 5 were given a single intra-peritoneal injection of 60 mg/kg BW of streptozotocin to induce diabetes. Group 3 was further treated with subcutaneous insulin (4.0 IU/kg BW) twice daily, respectively.ResultsStretozotocin (STZ) only-treated groups exhibited hyperglycemia, polydipsia, polyuria, weight loss, glucose intolerance, low fasting plasma insulin and reduced hepatic glycogen content compared to the control group. Furthermore they had significantly elevated Malondialdehyde (MDA), acetoacetate, β-hydroxybutyrate, anion gap and significantly reduced blood pH and plasma bicarbonate compared to the control group. Naringin treatment significantly improved Fasting Plasma Insulin (FPI), hepatic glycogen content, malondialdehyde, β-hydroxybutyrate, acetoacetate, bicarbonate, blood pH and anion gap but not Fasting Blood Glucose (FBG) compared to the STZ only-treated group.ConclusionsNaringin is not hypoglycemic but ameliorates ketoacidosis and oxidative stress. BACKGROUND: Recent neuroimaging studies have found that several areas of the human brain, including parietal regions, can respond multimodally. But given single-cell evidence that responses in primate parietal cortex can be motor-related, some of the human multimodal activations might reflect convergent activation of potentially motor-related areas, rather than multimodal representations of space independent of motor factors. Here we crossed sensory stimulation of different modalities (vision or touch, in left or right hemifield) with spatially directed responses to such stimulation by different effector-systems (saccadic or manual). RESULTS: The fMRI results revealed representations of contralateral space in both the posterior part of the superior parietal gyrus and the anterior intraparietal sulcus that activated independently of both sensory modality and motor response. Multimodal saccade-related or manual-related activations were found, by contrast, in different regions of parietal cortex. CONCLUSIONS: Whereas some parietal regions have specific motor functions, others are engaged during the execution of movements to the contralateral hemifield irrespective of both input modality and the type of motor effector. 相似文献
