Background

The yeast Saccharomyces cerevisiae provides unique opportunities to study roles and regulation of aqua/glyceroporins using frontline tools of genetics and genomics as well as molecular cell and systems biology.

Scope of review

S. cerevisiae has two similar orthodox aquaporins. Based on phenotypes mediated by gene deletion or overexpression as well as on their expression pattern, the yeast aquaporins play important roles in key aspects of yeast biology: establishment of freeze tolerance, during spore formation as well as determination of cell surface properties for substrate adhesion and colony formation. Exactly how the aquaporins perform those roles and the mechanisms that regulate their function under such conditions remain to be elucidated. S. cerevisiae also has two different aquaglyceroporins. While the role of one of them, Yfl054c, remains to be determined, Fps1 plays critical roles in osmoregulation by controlling the accumulation of the osmolyte glycerol. Fps1 communicates with two osmo-sensing MAPK signalling pathways to perform its functions but the details of Fps1 regulation remain to be determined.

Major conclusions

Several phenotypes associated with aqua/glyceroporin function in yeasts have been established. However, how water and glycerol transport contribute to the observed effects is not understood in detail. Also many of the basic principles of regulation of yeast aqua/glyceroporins remain to be elucidated.

General significance

Studying the yeast aquaporins and aquaglyceroporins offers rich insight into the life style, evolution and adaptive responses of yeast and rewards us with discoveries of unexpected roles and regulatory mechanisms of members of this ancient protein family. Contamination of soils with copper (Cu) has become a serious problem in the environment. Phytoremediation is an emerging green technology that uses green plants to remediate heavy metal contaminated areas. This study was conducted to evaluate the potential of Jatropha curcas for remediation of soils contaminated with Cu. Seedlings were planted in soils spiked with Cu in amount of 0, 50, 100, 200, 300, and 400 mg kg–1 (Cu0, Cu50,Cu100,Cu200,Cu300, and Cu400) for a period of five months. The maximum height and number of leaves were recorded in control (Cu0) whereas the highest basal stem diameter was found in seedlings exposed to Cu50. Copper concentrations among plant parts were in the following trend: roots > stems > leaves. The highest total Cu concentration (665 ± 1 mg kg?1) and total Cu removal (1.2 ± 0.2%) based on total plant dry biomass were found in Cu400 and Cu50 treatments, respectively. J. curcas exhibited high root concentration factor (RCF > 1) and low translocation factor (TF < 1). Although Cu accumulation by the plant didn't reach the criteria of Cu hyperaccumulators, this species showed a potential to be used in phytostabilization of mildly Cu contaminated areas. However, the plant cannot be used for phytoextraction of Cu-contaminated soils.  相似文献

Background:Decitabine is a potent anticancer hypomethylating agent and changes the gene expression through the gene''s promoter demethylation and also independently from DNA demethylation. So, the present study was designed to distinguish whether Decitabine, in addition to inhibitory effects on DNA methyltransferase, can change HDAC3 and HDAC7 mRNA expression in NALM-6 and HL-60 cancer cell lines.Methods:HL-60, NALM-6, and normal cells were cultured, and the Decitabine treatment dose was obtained (1 µM) through the MTT assay. Finally, HDAC3 and HDAC7 mRNA expression were measured by Real-Time PCR in HL-60 and NALM-6 cancerous cells before and after treatment. Furthermore, HDAC3 and HDAC7 mRNA expression in untreated HL-60 and NALM-6 cancerous cells were compared to normal cells.Results:Our results revealed that the expression of HDAC3 and HDAC7 in HL-60 and NALM-6 cells increases as compared to normal cells. After treatment of HL-60 and NALM-6 cells with Decitabine, HDAC3, and HDAC7 mRNA expression were decreased significantly.Conclusion:Our data confirmed that the effects of Decitabine are not limited to direct hypomethylation of DNMTs, but it can indirectly affect other epigenetic factors, such as HDACs activity, through converging pathways.Key Words: Decitabine, HDAC3, HDAC7, HL-60, NALM-6  相似文献

Shot hole disease is one of the most important diseases of stone fruit trees in Iran. The disease is wide spread among orchards of Prunus spp. During spring and summer of 2007, 80 monoconidial isolates of the pathogen were recovered from infected leaves, fruits and twigs of different Prunus spp. in West Azerbaijan, Tehran, Ghazvin and Razavi Khorasan provinces of Iran and were studied taxonomically. Based on morphological and physiological characteristics and growth optimal temperature, all isolates were identified as Wilsonomyces carpophilus. Seedlings of stone fruits (apricot, almond, peach, nectarine, plum, sweet cherry and sour cherry) were used for pathogenicity tests. All seedlings were susceptible to the fungal isolates and showed disease symptoms on twigs, leaves, buds and petioles. Genetic diversity of 28 selected fungal isolates was investigated based on DNA fingerprinting by random amplified polymorphic DNA-polymerase chain reaction (RAPD-PCR), using four random primers. Based on cluster analysis of the PCR results from the four primers, 10 fingerprinting groups (clonal lineages) and 27 haplotypes were identified. Clonal lineages “C”, “D” and “E”, each with six haplotypes formed the biggest clonal lineages, but other clonal lineages (“B”, “F”, “G”, “H”, “I” and “J”) included only one isolate. No correlation was detected among clonal lineages with the location of selected isolates and their host species. A correlation was found between the substrate (fruit, twig or leaf) and clonal lineages, particularly in “C” clonal lineage. The results showed that the fungus population had high genetic diversity which is distributed among the different areas of Iran.  相似文献

Application of remote sensing (RS) and geographical information system (GIS) techniques has been increased in natural sciences. In fact, it is inevitable applying of these techniques in vegetation studies due to the existence of some problems in traditional methods (e.g. sampling, calculation, analysis and so on). On this scope, scientists must have sufficient information about the efficiency of these techniques as a useful tool in their studies. This study aims to evaluate the IRS-P6 LISS III and Landsat ETM+ efficiency in plant groups’ identification. In order to this purpose, 143 training samples were collected from areas that showed homogenous composition of plant species in at least area of 3600 m2 (60 × 60 m). Coordinates of these training samples were recorded using a GPS device and transferred to a GIS database. Also, ENVI 4.2 package has used to process and analyze the satellites data. Several methods of processing such as; spectral separability, supervised classification and assessment of classification accuracy were used in order to gain a satisfy evaluation of the data efficiency. The results indicated that net farming of alfalfa and Juniperus polycarpus–Artemisia kopetdaghensisi community have the most separability on the satellite images (1.99 for Landsat and 2 for IRS). Against, the least separabilities on the Landsat data were between Ju. polycarpus–Onobrychis cornuta and Ju. polycarpus–Ar. kopetdaghensis communities (1.57) and between Ju. polycarpus–Ar. kopetdaghensis and Ju. polycarpus–Agropyron intermedium (1.53) on the IRS data. According to these results, it is concluded that the satellite data are somedeal able to identify plant groups when vegetation communities are sufficiently homogenous, abundant and spectrally and ecologically separable.  相似文献
