The vascular plant collections of R. S. Williams from Bolivia and Peru (1901–1902): Addenda et corrigenda

Notes that augment an earlier essay on R. S. Williams’ botanical activities and the distribution of his collections from Bolivia and Peru are presented. Twelve species based on Williams’ vascular plant collections are added to a catalog that was previously published. Synonymies of an additional 22 taxa are corrected or updated. Notes also are provided concerning the typification or orthography of two species.     

Chromosome numbers in callirhoe (Malvaceae)

Forty-four chromosome counts from the North American genusCallirhoe are reported, including first-recorded counts ofC. alcaeoides, C. digitata, C. bushii, C. triangulata, andC. involucrata var.tenuissima. Information concerning chromosome numbers from present and previous reports is summarized and evaluated. Observations are presented on meiosis for the genus in general, for male sterile individuals of gynodioecious species, and for selected F₁ hybrids. Pollen fertility data are recorded for the latter. Chromosomally three species groups are recognized: an exinvolucellate, diploid species group withn=14; an involucellate, tetraploid and octoploid species pair,n=28 and 56; and an involucellate, diploid and tetraploid species group withn=15 and 30. Intrageneric relationships and those within the tribe Malveae subtribe Malvinae are discussed.     

A previously unrecognized Isidrogalvia (Liliaceae) from Bolivia

Isidrogalvia longiflora, a new combination based onAsagraea longiflora, is proposed. It is the first record ofIsidrogalvia from Bolivia.

In 1843 S. B. Buckley (1809–1884) traveled up the St. Johns River, Florida in search of plants, shells, and possibly other natural history objects. Buckley encountered many difficulties in interior Florida and his trip did not live up to his original expectations. It was unfortunate that the took years to distribute his plant specimens, since few botanists or naturalists had preceded him into that poorly explored region.

Immunohistochemistry in the evaluation of neovascularization in tumor xenografts

Angiogenesis, or neovascularization, is known to play an important role in the neoplastic progression leading to metastasis. CD31 or Factor VIII-related antigen (F VIII RAg) immunohistochemistry is widely used in experimental studies for quantifying tumor neovascularization in immunocompromised animal models implanted with transformed human cell lines. Quantification, however, can be affected by variations in the methodology used to measure vascularization including antibody selection, antigen retrieval (AR) pretreatment, and evaluation techniques. To examine this further, we investigated the microvessel density (MVD) and the intensity of microvascular staining among five different human tumor xenografts and a mouse syngeneic tumor using anti-CD31 and F VIII RAg immunohistochemical staining. Different AR methods also were evaluated. Maximal retrieval of CD31 was achieved using 0.5 M Tris (pH 10) buffer, while maximum retrieval of F VIII RAg was achieved using 0.05% pepsin treatment of tissue sections. For each optimized retrieval condition, anti-CD31 highlighted small vessels better than F VIII RAg. Furthermore, the MVD of CD31 was significantly greater than that of F VIII RAg decorated vessels (p<0.001). The choice of antibody and AR method has a significant affect on immunohistochemical findings when studying angiogenesis. One also must use caution when comparing studies in the literature that use different techniques and reagents.
