Tetramorphism of trypomastigote forms has been discovered in the blood stream of mice infected with Trypanosoma cruzi, that is C slender, C middle, S slender, S middle and forms intermediate between C and S. Regular changes of forms have been observed in the course of the infection. In the middle of the process, at the moment of maximum destruction of parasites affected by immunosystems of the host, C-forms prevailing on the whole substantially give place to S-forms, slender variants being replaced by middle ones within each of them. Polymorphism in the bloodstream of the vertebrate host is the general property of all trypanosomes, with their common biological value: immunological adaptation to parasitism in the blood and preadaptation to the continuation of the life cycle in the invertebrate host.  相似文献

The basic experimental data obtained at the Department of Coenzymes' Biochemistry of the A. V. Palladin Institute of Biochemistry of the Ukr. SSR Academy of Sciences as to the biological role of vitamin E are analyzed. Vitamin E, selenium and methionine are found to induce peculiar changes in the activity of glutathione-peroxidase, metabolism of sulphur-containing amino acids, biosynthesis of adenine nucleotides, proteins and nucleic acids. Participation of alpha-tocopherol and its active derivatives in the control of biosynthesis and intertransformation of ubiquinone and its cyclic isomer, ubichromenol, in the animal organism, is proved, which determines to a considerable extent the biological role of vitamin E in the bioenergy processes. It is substantiated in experiments that the detected wide range of the biological effect of vitamin E is associated with the control of RNA biosynthesis. Under these conditions the effect of vitamin E on the RNA synthesis does not depend on the manifestation of antioxidant properties of its molecule and in this sense it is a specific one. The results obtained are discussed for their significance in explanation of the molecular mechanism of the vitamin E action and in substantiation of the possibility to use the results in practical medicine and animal husbandry.  相似文献

Lipid free-radical oxidation (LFRO) on the D-deficient model with the animal administration per os by vitamin D3 (cholecalciferol), specimen from Serratula coronata plant, containing low molecular weight biological sterol--20-hydroxyecdysone (ecdysterone) was investigated. As well influence of arginine as one of the amino acid components of the specimen from Serratula coronata was investigated. The LFRO in the blood serum, liver microsomal and mitochondrial fractions were characterize by measured of the chemiluminescence kinetic parameters. Vitamin D3, the specimen from Serratula coronata plant and arginine displayed some antioxidant (AO) properties as was established. On the basis of the received experimental data the series of their antioxidizing activity: vitamin D3 = drug from Serratula coronata > arginine were offered. The possible mechanism of the 20-hydroxyecdysone and arginine participation on LFRO maybe in free-radical reactions breaking via hydroxy and amino groups in structure of their molecules.  相似文献

Concentration of lipid peroxidation products and antioxidant enzyme activities in rat brain and erythrocytes and the effects of nicotinamide and nicotinoyl-GABA administration on these parameters were estimated on 21st day of streptozotocin-induced diabetes. It was demonstrated more then two-fold diabetes-induced accumulation of conjugated dienes and malondialdehyde in tissues studied. Superoxide dismutase and glutathione reductase activities of both brain homogenate and erythrocytes as well as catalase and glutathione peroxidase activities of brain homogenate were shown to decrease significantly in diabetic rats, meanwhile, catalase activity of erythrocytes was increased and glutathione peroxidase unchanged. So the correlation between changes in enzymatic antioxidant system in brain and erythocytes failed to be found. Alterations observed were virtually prevented by the course of nicotinamide and nicotinoyl-GABA treatment. The results suggested that the suppression of antioxidant system could be primary biochemical disturbance in diabetic neuropathy progression. It was shown that the antioxidant efficacy of nicotinoyl-GABA is lower than that of nicotinamide. It was suggested that the mechanism of antioxidant action of nicotinamide and its structural analogue consists of both scavenging of lipid peroxides and NAD biosynthesis that leads to activation and normalization of altered energy and lipid metabolism.  相似文献

alpha-Tocopherol was found to decrease the level of rat thymocytes DNA fragmentation and to increase the viability of these cells under apoptosis induction by dexamethazone. So the antiapoptotic role of this vitamin is suggested. In contrast to alpha-tocopherol synthetic antioxidant ionol and alpha-tocopherylacetate, contained only 6 carbon atoms in its isoprenoid side chain caused the cell death from necrosis. This process preceded the apoptosis stimulated by dexamethazone.  相似文献

The long-term influence of low X-ray irradiation increases lipid peroxidation (LP) in radiosensitive (bone marrow, enterocytes of small intenstine) and in relatively radioresistant blood cells (erythrocytes). The activation of antioxidant system enzymes in observed cells does not decrease LP intensity. We concluded that additional administration of alpha-tocopherol provided the decrease of the first and end products of LP in the observed tissues mostly in the beginning of the experiment. Antioxidant effect of the preparation is more significant in cells with high proliferative activity but normal activity of enzymes was not determined.  相似文献

An increase in GABA uptake by isolated rat brain synaptic endings as well as a decrease of pharmacologically active GABA analogue muscimol specific binding have indicated a physiologically drastic failure in realization of GABA-mediated inhibitory effects in CNS induced by diabetic encephalopathy. In spite of the impairment of inhibitory function of GABAergic transmission in diabetes a crucial activation of benzodiazepine receptors was determined, as it is tested by the increase in specific binding of flunitrazepam by synaptic membranes. This increase may play an important role in endogenous control of neural activity associated with the factors undefined so far. Using the approach that GABA, and several synthetic GABA agonists, appear to increase the affinity of the benzodiazepine recognition sites for such ligands, presumably by some allosteric mechanism, the findings concerning the in vitro binding assay technique confirm at least some of the functional characteristics observed between GABA and benzodiazepine receptors in vivo under pathological conditions. Indeed, the absence of activating effect on the affinity of flunitrazepam specific binding in the presence of micromolar concentrations of exogenous GABA implicate diabetes-induced alterations in coupling GABA- and benzodiazepine receptors that might be linked to changes in conformantial state of this membrane-bound complex and could partially explain diabetes-induced impairments of GABAergic transmission evaluated in the present study. Our study suggests that nicotinamide and especially GABA play an important role in improving the functioning of brain GABA-benzodiazepine complex impaired in diabetes through specific ligand-mediated mechanism and can be useful in the management of diabetes-associated brain failures.  相似文献

alpha-Tocopherol added in vivo increased protein kinase C--dependent phosphorylation of rat liver nuclei proteins. Such effect of tocopherol was found also after the addition of sphingosine known as natural inhibitor of protein kinase C to incubation medium. The stimulation of phosphorilation was observed also after the addition of alpha-tocopherol to the Triton X-100 solubilized nuclear fraction which contained protein kinase C and tocopherol-binding proteins.  相似文献

While estimating the effect of tocopherol, tocopherylquinone and their complexes with the tocopherol-binding proteins from the rat liver cytosole on arachidonate 5-lipoxygenase from peritoneal-lymphocytes and soybean linoleate-5-lipoxygenase alpha-tocopherol and especially its complex with tocopherol-binding protein was defined to inhibit the activity of both vegetative- and animal nature-lipoxygenase.  相似文献
